2.Effect of compound K on spinal Toll-like receptor 4 expression during morphine-induced hyperalge-sia in rats

Objective To evaluate the effect of compound K ( CK) on spinal Toll?like receptor 4 ( TLR4) expression during morphine?induced hyperalgesia in rats. Methods Thirty?six healthy male Spra?gue?Dawley rats in which intrathecal catheters were successfully implanted were randomly divided into 3 groups ( n=12 each) using a random number table: control group ( group C) , morphine?induced hyperal?gesia group ( group M) , and morphine + CK group ( group M+CK) . Starting from 5 days after successful implantation, normal saline 10 μl, morphine 10 μg, and morphine 10 μg + CK 10 μg were injected in?trathecally twice a day for 7 consecutive days. Tail?flick latency ( TFL) to a thermal nociceptive stimulus was measured at 1 day before administration ( T0 , baseline) and at 30 min after the initial administration on 1st, 3rd, 5th and 7th days (T1?4), and the percentage of maximum possible effect (MPE) was calculated. The rats were sacrificed after the last measurement of tail?flick latency, and the lumbar segment ( L3?5 ) of the spinal cord was removed for determination of the expression of TLR4 by Western blot. Results MPE was significantly lower at T3,4 than at T1 in M and M+CK groups ( P<0?05) . Compared with group C, MPE was significantly lower at T2?4 , and the expression of TLR4 was up?regulated in M and M+CK groups ( P<0?05). Compared with group M, MPE was significantly increased at T1?4, and the expression of TLR4 was down?regulated in group M+CK ( P<0?05 ) . Conclusion The mechanism by which CK alleviates mor?phine?induced hyperalgesia is associated with down?regulation of TLR4 expression in rats.

5.Therapeutical effect of Irbesartan for coronary atherosclerosis mice (ApoE-/-) and its possible mechanisms

Objective:To analyze the therapeutical effect of Irbesartan for coronary atherosclerosis mice ( ApoE-/-) and its possible mechanisms. Methods: A total of 16 male ApoE-/- mice were randomly divided into control group and Treatment group [Irbesartan,50 mg/(kg·d),4 weeks]. HE,immunofluorescence,Western blot and ELISA were performed to analyze the effect of Irbesartan on ApoE-/- and the changes of related signaling pathways. Results: Compared with control group,the treatment group had lower atheroma macular areas and inflammatory cytokines in blood vessel (P<0. 05). Treatment group had lower levels of leptin,but higher levels of PPAR-γ and adiponectin in perivascular adipose tissues ( PVAT ) than these of control group, the difference were statistically significant ( P<0. 05 ) . Western blot and immunofluorescence analysis shown that Irbesartan treatment significantly depressed the expression of p-p65 and p-IKK in PVAT when compared with these of control group (P<0. 05). Conclusion:Irbesartan has significantly therapeutic effect on ApoE-/-mice,the possible mechanisms including anti-inflammatory effects in PVAT,improved the adipose tissue function and regulated the PPAR-γ-NF-κB signaling pathways.
6.Bilateral same-session ureteroscopic lithotripsy-safety and efficacy

Objective To investigate the safety and efficacy of same session ureteroscopical lithotripsy as a valuable treatment for the bilateral upper urinary stones.Methods 32 cases with bilateral upper urinary stones were enrolled into this study from Jan 2013 to Dec,including 19 male and 13 female.The age ranged from 19 to 79 years old [mean (53.2 ± 14.2) years old].Ultrasound,CT and IVU were used to evaluate the details about the stone burden,number and location.The total stone burden was (23.7± 7.6)mm,ranged from 14 to 40mm.The kidney stone burden ranged from 0 to 37mm [mean (15.4 ±9.7)mm].and ureter stone burden ranged from 0 to 34 mm [mean (8.2 ±7.2)mm].Total stone number was 109,including 78 stones in kidney and 31 stones in ureter.23 (71.9%) patients had stents placed before the surgery.All the patients were treated with FURL and(or) URL by the same surgeon under general anesthesia,and C arm was used routinely to monitor the position of the guide wire,sheath and residual stones.The stents were placed for 2 to 4 weeks post-operatively.The ureteral catheters were placed for 24 to 48 hours.Stone-free rates(SFRs) were judged by KUB and(or)NCCT on the first day after the surgery and one month after withdraw the stent.The number of procedures,operative duration,hospital stay,SFRs in different time phase,serum creatinine,stone composition and complications were evaluated.All patients were divided into low burden group and high burden group,based on the borderline of stone burden (20mm).The SFR and complication was compared among those group.Results Among 32 cases,30 cases underwent the one stage procedure and 2 cases underwent two stage procedure.Operative time was (99.2 ± 32.5) mins.There was no significant difference of serum creatinines before and after the surgery.[(78.3 ± 15.0) μmoL/L vs.(77.9 ± 15.3) μmol/L,P =0.711].The overall SFRs aftcr 1,2 procedures were 84.0% (27/32) and 91.0% (29/32),respectively.The immediate SFRs for patients with a preoperative stone burden ≤20 and >20 mm showed significant difference(100% vs.50%,P =0.002).Only 5 minor postoperative complications,including fever in 4 patients,hematuria in 1 were recorded.Longterm complication,such as ureteral stricture,was not noticed.Conclusions For selected cases with bilateral upper urinary stones,bilateral same-session ureteroscopy is effective and safe with little influence on the kidney function.For those whose stone burden less than 20mm,the immediate SFRs are much higher.
7.Relation between breast cancer susceptibility gene 1 expression and the efficacy of platinum-based chemotherapy in colorectal cancer

Objective To explore the expression of breast cancer susceptibility gene 1 (BRCA1) in human colorectal cancer and its correlation with the efficacy of platinum-based chemotherapy.Methods Seventy-eight cases of colorectal cancer patients treated with platinum-based chemotherapy after surgery were collected.The surgical specimens of them were taken.Fourteen normal colonic mucosa specimens and 12 non-cancerous tissues of colorectal cancer specimens were obtained.The expression of BRCA1 in tissues was detected by immunohistochemistry and analyzed by x2 test.The colorectal cancer patients were followed up for survival time.Comparison of survival analysis was performed by Kaplan-Meier survival curves and Log-rank test.Results The positive rate of the BRCA1 expression in colorectal cancer tissue (52.6 %,41/78) was lower than that of para-cancer tissue (11/12,x2 =6.518,P=0.011) and normal colonic mucosa tissue (13/14,x2 =7.949,P=0.005).The poorer the differentiation of colorectal cancer,the lower the positive rate of BRCA1 (x2=14.160,P=0.001).The median disease-free survival time of BRCA1 negative colorectal cancer patients was 51.0 months (95 % CI:47.7 to 54.4 months),which was longer than that of BRCA1 positive patients (45.0 months,95 %CI:36.6 to 53.4 months,x2 =4.367,P=0.032).Conclusions Receiving oxaliplatin based chemotherapy may be a survival benefit for BRCA1 negative colorectal cancer patients.The expression of BRCA1 may be an index for chemotherapy options and prognosis evaluation for colorectal cancer patients after surgery.
8.The Preliminary Study on the Plasma Normal Reference Value of Heart-type Fatty Acid-binding Protein

Objective To know normal reference value of heart-type fatty acid-binding protein (H-FABP) in serum. Methods Serum H-FABP levels of 126 healthy individuals were measured by ELISA. Levels of MYO, cTnI and CK-MB were assayed in the same time as control. The normal threshold value was calculated as mean value?2s. Results The normal concentration of H-FABP in serum was 3 79?3 52?g/L and the difference in age and gender wasn’t significant. Conclusions The normal reference value of serum H-FABP is 0~10 83?g/L.
9.Study of M?nchausen syndrome

Objective To explore the methods of identifying factitious/fraudulent disorders (M?n-chausen syndrome), enhancing the understanding of their predisposing factors, clinical features, and underlying psychopathology. Method After a final diagnosis of factitious fever was made, data of 4 patients presenting with a puzzling fever of unknown origin were analyzed and compared with those of other cases reported in the English literature. Result It was confirmed that the elevated temperature recorded in the clinical charts of the 4 patients was faked by manipulating of the thermometer. Conclusion These 4 cases show the existence of factitious/fraudulent disorders in China, possibly related to an abnormal psychology, but the tricks employed by the patients are relatively simple compared with those used abroad.
10.Analysis of lost goodwill target proteomic fingerprints drifting from negative to positive in advanced lung adenocarcinoma patients

Objective To analyze the serum related proteomic fingerprints when Lost Goodwill Target (LGT) proteomic fingerprints drifting from negative to positive in the advanced lung adenocarcinoma patients. Methods The serum proteomic fingerprints of 31 advanced lung adenocarcinoma patients whose LGT fingerprints drifted from negative to positive were detected by SELDI and CM10 protein chip. More than 10 % cluster and M/Z values from 11,000+H to 12,000+H was regarded as LGT positive, otherwise as negative. Different fingerprints were screened by Biomarker Wizard 3.1 and Biomarker Wizard software and the decision tree model was established. Results There were 16 statistically different protein peaks when LGT fingerprints drifting from negative to positive, including 10 up-regulation proteomic fingerprints (M/Z:11531, 11483, 11686, 11394, 11822, 11323, 11911, 12450, 5811 and 5709) and 6 down regulation proteomic fingerprints( M/Z: 4126, 13927, 13784, 7001, 1959 and 2741). Conclusion By SELDI and CM10 protein chip detection, up-regulating fingerprints of M/Z 11531, 11483, 11686, 11394, 11822 and 11323 were regarded as the subtype of LGT when it drifting from negative to positive, while up-regulation of M/Z 11911,12450, 5811 and 5709 and down-regulating of M/Z 4126, 13927, 13784, 7001 and 1959 were regarded as the related fingerprints when LGT drifting from negative to positive. The above different fingerprints are constituted of the fingerprints library when LGT fingerprints drifting from negative to positive and it will provide a platform for studying the LGT proteins.
