2.Statistical analysis of monitoring data regarding the state of Keshan disease in Wudu district, Longnan city, Gansu province in 2009

Objective To study the current cases of Keshan disease aims at providing scientific and basic evidence for prevention and control of the disease. Methods In accordance with historical cases, in 2009, an investigation was conducted in the permanent resident population in seriously infected representative villages of landscapes and ways of living such as Xujialeng, Zhuping and some other adjacent villages in Wudu district, Longnan city. People in those areas were examined and carefully observed based on the scheme of Keshan disease surveillance. Then they were enquired about their medical history, given normal clinical examinations and electrocardiogram(ECG) examination, the suspected cases were chest X-rayed. At the same time, some samples of their hair of some children under 12 and adults were collected to be examined for the selenium content. Results Among the total number of 515 subjects, 20 were diagnosed with Keshan disease with a total detection rate of 3.9% (20/515), among whom 13 were potential patients with a detection rate of 2.5%( 13/515), 7 were chronic patients with a detection rate of 1.4%(7/515). In the total number of 515 ECG, 48 were found with abnormal ECG at a detection of 9.3%(48/515). In the 20 patients, the potential patients were mostly featured with incomplete right bundle-branch block(5 cases), complete right bundle-branch block(3 cases), frequent ventricular premature beat (2 cases) and left ventricle high voltage(2 cases), while the chronic patients were mostly featured with incomplete right bundle-branch block(2 cases). Of the 29 chest X-ray films, 17 cases had heart enlargement with a detection rate of 58.6%( 17/29). The selenium content in the hair of 10 adults and 10 children were (0.38±0.09), (0.31±0.12) mg/kg, respectively. Conclusion There are still existing some cases and obvious virulence factors of Keshan disease in Wudu district, Longnan city.
3.Study on Degradation of Rhizoma Typhonii Polysaccharide with Sulfuric Acid

Objective To study the degradation of Rhizoma Typhonii polysaccharide with sulfuric acid under the normal and ultrasonic condition. Methods The orthogonal test table L9(34) was used to optimize 4 factors including acid concentration, reaction time, reaction temperature and ultrasonic power, with the yield of the low-molecular-weight polysaccharide as the index. Results The optimal conditions of discomposing Rhizoma Typhonii polysaccharide with sulfuric acid were 10% sulfuric acid, 100 ℃ for 2 h under the normal condition,and the yield of low-molecular-weight polysaccharide was 36.17%. Under the ultrasonic condition, the optimal conditions were 150 W of the ultrasonic power, 5% sulfuric acid, 40 ℃ for 1 h, and the yield was 44.63%. Conclusion The degrading yield of the low-molecular-weight polysaccharide can be increased under ultrasonic condition with sulfuric acid.

5.Ultrasound imaging features of hepatic regenerative nodules in Budd-Chiari syndrome after portacaval anastomosis

Objective To investigate the routine ultrasound and contrast-enhanced ultrasound (CEUS) features of hepatic nodules in Budd-Chiari syndrome(BCS) after portacaval anastomosis.Methods Routine ultrasonography and CEUS were performed in 18 BCS patients with hepatic nodules after portacaval anastomosis.Results Appearance after portosystemic anastomosis,multiplicity,small size,presence of peripheral rim and hypervascularization were important ultrasound imaging features of hepatic regenerative nodules in patients with BCS.Nodules showedquick wash-in and slow wash-out pattern in CEUS.Sixteen cases showed center-to-periphery enhancement pattern in arterial phase and hyper-enhancement in portal phase and late phase.Two cases showed periphery-to-center enhancement pattern in arterial phase and periphery enhancement in portal phase and late phase.Conclusions Hepatic regenerative have different features on routine ultrasound and CEUS in patients with BCS after portacaval anastomosis,which are useful for differential diagnosis.
6.Progress in the isolation and purification methods of adipose-derived stem cells

Adipose-derived stem cells ( ASCs ) as potential seeded cells have been widely used in tissue engineering.Thus to obtain enough, high activity, high purity adipose-derived stem cells is the particular important premise of the application in tissue engineering.In this paper, the isolation and purification methods of ASCs were reviewed and the merit and demerit of different methods were compared in order to provide theoretical basis for safe and high-effective isolation and purification of ASCs.
7.Three-dimensional double echo steady state with water excitation MR imaging of facial nerve at 1.5T: A preliminary study

Objective To assess the value of three-dimensional double echo steady state with water excitation (3D-DESSwe) MR sequence in depicting the normal anatomy of facial nerve. Methods 3D-DESSwe MR sequence was performed in 15 healthy volunteers on a 1.5T MR. The 3D datasets obtained were constructed in relation to the course of the facial nerve using multi-planar reconstruction (MPR) and thin slice maximum intensity projection (MIP) programs, and the images acquired were compared with anatomical textbook diagrams. Two doctors analyzed the images in collaboration and recorded on an arbitrary scale of 0-2 grades (grade 2, homogeneity and facial nerve identified with certainty;grade 1, mild inhomogeneity and facial nerve probably identified;grade 0, severe inhomogeneity and facial nerve not identified). Results In all subjects, 3D-DESSwe images provided reliable definition of the normal facial nerve anatomy bilaterally and high contrast between the facial nerve and adjacent structures were obtained. The cisternal segment of facial nerve showed high signal intensity while the moving CSF flow surrounding it showed low signal intensity. The internal auditory canal (IAC) segment of facial nerve showed relatively low signal intensity while the stationary CSF surrounding it showed high signal intensity. The labyrinthine, tympanic, mastoid and extracranial segments of facial nerve showed high signal intensity while the bone and soft tissue surrounding it showed low signal intensity, and parts of facial nerve branches could be demonstrated. Conclusion 3D-DESSwe sequence can display the complete course of the normal facial nerve, especially for the intratemporal and extracranial parts.
8.Studies on Mutagenesis of Virginiamycin Producing Strain X-435

Streptomyces sp.X-435 isolated from a soil sample collected in the suburbs of Beijing was proved to be a produce Virginiamycin.To improve the productivity of the Virginiamycin of Streptomyces sp.X-435,the spores of strain X-435 were treated with UV.The three types of colony,strawhat,wrinkled,blad,were isolated on Gaose's medium plates after mutation.Among them,the colonies of strawhat type exhibited positive mutation and were picked up as objects of screening.After five generation of mutation,the mutant F5-25-u-28 was selected which potency of Virginiamycin was about 20times higher than that of the beginning strain by flask fermentation and was also genetic stable.
9.Culture of fibroblast-like synoviocytes from rabbits with adjuvant arthritis

BACKGROUND:Currently, fibroblast-like synoviocytes from the synovial tissue of patients with rheumatoid arthritis or adjuvant arthritis rats are isolated and cultured mainly using enzyme digestion method, but tissue explant adherent method is rarely reported in primary culture. OBJECTIVE:To establish a culture method of fibroblast-like synoviocytes from adjuvant arthritis rabbits and to investigate cell biological characteristics. METHODS:Rabbit models of adjuvant arthritis were prepared by injected with adjuvant at multiple sites and times, the knee synovial tissues were obtained, and fibroblast-like synoviocytes were separated and cultured by the tissue explant adherent method. Furthermore, the cell morphology was observed. The cell proliferation was measured using cell counting kit-8 assay, and the expression of Vimentin was detected by immunofluorescence.RESULTS AND CONCLUSION:The primary cultured rabbit synoviocytes were long spindle fiber-like and coincided with the fibroblast-like synoviocytes. Moreover, cells were positive for Vimentin. In conclusion, fibroblast-like synoviocytes from rabbits with adjuvant arthritis are successfully isolated and cultured by the tissue explant method.
10.Recent progress in researching the reactive oxygen species-mediated myocardial remodeling

Reactive oxygen species(ROS) in the cardiac muscle are increased in cardiovascular diseases such as hypertension,coronary heart disease and heart failure.Oxidative stress mediates myocyte hypertrophy,myocyte apoptosis and interstitial fibrosis.And these changes are particularly pronounced in elderly patients or senescent animal disease models.The major sources of ROS in myocardium are mitochondria and the reduced nicotinamide adenine dinucleotide phosphate(NADPH) oxidase.Specific ROS such as hydrogen peroxide play an important role in myocardial remodeling in heart failure.Oxidative modifications of calcium handling proteins result in reduction of their activities,leading to decreased myocardial contractility.Reduction of oxidant levels and inhibition of oxidative modifications of specific proteins through inhibiting ROS sources may provide new strategies for the treatment of myocardial remodeling.
