4.Epidemiological investigation of an outbreak of P1-Ⅱ Mycoplasma pneumoniae infection in a school

Abstract
On October 13 2023, the Beijing Economic and Technological Development Area Center for Disease Control and Prevention recAAAAAeived a report from a school indicating an outbreak of fever among students in Class 2, Grade 4. An on-site epidemiological investigation was immediately conducted. A total of 14 cases were reported, all of whom were students, with an attack rate of 34.15%. The onset of illness was concentrated between October 6 and 15, with the main clinical symptoms being fever, cough and fatigue. There were no cases of hospitalization, severe illness or death. Seven cases tested positive for Mycoplasma pneumoniae P1-Ⅱ. Based on the on-site investigation and laboratory test results, it was an outbreak of influenza-like illness caused by Mycoplasma pneumoniae infection. The likely cause of the spread was cases infecting other classmates through respiratory droplets while attending class. The outbreak occurred only in one class and did not spread to neighboring classes. Schools should actively carry out health education to raise awareness among parents for reporting infectious diseases, and strengthen morning and afternoon screenings to improve the sensitivity of identification.
5.Expression of nuclear factor kappaB, MCP-1 and RANTES concentration in peritoneal fluid in women with endometriosis

Objective To compare the concentration of nuclear factor-kappa B (NF-κB), MCP-1 and RANTES in peritoneal fluid in women with endometriosis and normal peritonal fluid ,To explore the relationship between these cytokines with pathogenesis of endometriosis. Methods There were 2 groups:the endometriosis group and the control group.Double-antibody sandwich ABC-ELISA technique was adopted for determinating concentration of NF-кB、MCP-1 and RANTES in peritoneal fluid. Results Peritoneal fluid level of NF-κB MCP-1 and RANTES in endometriosis patients was higher than the control group (P < 0.05). NF-κB and MCP-1, NF-κB and RANTES expressed positive correlation by linear correlation analysis in both EMs group and control group , correlation coefficients were 0.385, 0.569(EMs group), 0.474, 0.388(P < 0.05). Conclusions NF-кB、MCP-1 and RANTES are mutual affection and promote endometriosis in the pathogenesis.

Sperm ultrastructural abnormalities are often associated with sperm motility, the integrity of genetic material, and the fertilization potential. The investigation of sperm ultrastructural abnormalities is based on the evolution of microscopy techniques. In his paper, we review the improvement of the microscopy techniques and the ultrastructure of several specific morphological defects and he apoptotic spermatogenic cells in order to expound the significance of sperm ultrastructural observation in clinical practice. We deem it necessary to analyze the sperm ultrastructure before exploring the pathology and adopting assisted reproductive technology for some special patients with teratozoospermia.

7.Study on the correlation of interferon induced virus-related genes and the clinical features of patients with systemic lupus erythematosus

Objective To investigate the expression of the genes correlated with interferon induced genes virus (MX1, OAS1, IFI44) in the peripheral blood leukocytes of patients with systemic lupus erythematosus (SLE), and to evaluate the relationships between the expression levels of these genes and diseaseactivity. Methods The clinical data of 100 SLE patients, 40 non-SLE patients with rheumatic diseases, and 40 normal controls were collected. Peripheral blood samples were collected. Total RNA was extracted and transcribed into cDNA. SYBR green dye based real-time quantitative PCR method was used to compare the expression levels (indicated as △CT value) of MX1, OAS1 and IFI44 in patients with SLE and those in the controls. Comparisons between groups were performed with ANOVA and Spearman correlations. Results ①The △CT value of MX1, OAS1 and IFI44 expression level of the SLE patients (3.4±1.8, 4.2±1.5, 8.8±2.2)was significantly higher than those of the non-SLE patients (2.4±0.4, 3.4±0.7, 5.4±2.1 ) and normal controls (2.3±1.1, 2.6±0.7, 5.2±2.0). ② The △CT value of OAS1 and IFI44 expression level of the SLE patients in severe disease was significantly higher than those of the SLE patients in mild disease and the SLE patients with stable disease. ③The ACT value of OASI and IFI44 were correlated with the SLEDAI scores (r=0.038,0.380). ④ The △CT values of MX1, OAS1 and IFI44 expression level of the SLE patients with arthritis were significantly higher than those of SLE patients without arthritis. ⑤ The △CT value of IFI44 expression level of the SLE patients with lupus nephritis (3.2±2.1,2.2±1.1) was significantly higher than that of the SLE patients without lupus nephritis. ⑥ There was correlations among these genes in SLE patients (P<0.05). Conclusion The value of MX1, OAS1 and FFF44 expression level of SLE patients is up-regulated. The real time expression levels of OAS1, IFI44 genes are associated with SLE disease activity and there are close correlation among these genes with interferon induce virus-relationed genes (MX1, OAS1, IFI44) in SLE patients.

9.Determination of Organic Solvent Residues in Testosterone Cypionate

Objective To establish a headspace GC method for the determination of residual organic solvents in testosterone cypionate. Methods The residual organic solvents were determined by GC with DB-WAX capillary colum(30 m× 0. 32 mm,0. 25 μm)and FID detector at 230℃,using high purity nitrogen as the carrier gas by headspace sampling. The flow rate was 1. 5 mL·min-1 ,the split ratio was 20:1,the temperature of injection port was 220℃,the headspace heating temperature was 70 ℃,and the headspace balance time was 40 mins. The content of residues was calculated by using n-propanol as the internal standard. Results Four residual solvents were completely separated. There were good linearity for m-ethanol,ethanol,benzene and pyridinein in the ranges of 4. 17-2. 50×103 μg·g-1(R=0. 999 9),4. 18-2. 51×103 μg·g-1(r=0. 999 6),0. 84-172 μg·g-1 (r=0. 998 1),and 2. 95-1. 77×103μg·g-1(r=0. 999 9),respectively. The detection limit was 2.08,1. 23,0.28,and 0. 87 μg·g-1,respectively. The average recovery of methanol,ethanol,benzene and pyridinein was 102. 2%(RSD=4. 0%),99. 6%(RSD=1. 9%),112. 6(RSD=5. 6%),and 98. 9%(RSD=1. 6%),respectively. Conclusion This method is reliable,sensitive, accurate and can be used for the determination of residual organic solvents in testosterone cypionate.
10.Effect of gap junction on permeability of blood-brain barrier in rats after cerebral ischemia- reperfusion

Objective To investigate the possible mechanism of the gap junctional influence on the change in permeability of the blood-brain barrier(BBB)after reperfusion subsequent to cerebral ischemia.Methods In the test laser scanning confocal microscope(LSCM)was used to investigate the change of Cx43 levels and distribution.The MCAO/R model was induced using intraluminal suture technique first described by Longa with a little modification.A total of 60 Wistar rats were divided into 4 groups:the sham-operation group,control group,octanol-treatment group and DMSO vehicle control group. Control group were further divided into seven subgroups at different time points of reperfusion after middle cerebral artery occlusion.To observe the change in permeability of BBB,Evans blue(EB)in the brain tissue was surveyed by the means of EB fluorescent quantitation.Octanol-treatment group and DMSO vehicle control group were done at the point of the peak of permeability of BBB.Octanol,the specific blocker for gap junctions(GJ)was used in an intervention study.To compare the amount of EB with the same point of groups,the influence of octanol on BBB permeability was investigated.Results At 3 h of reperfusion after cerebral ischemia for 2 h,the permeability of BBB began to increase,reached the peak at 24 h of reperfusion and was still elevated at 72 h.The Cx43 expression formed into bigger plague and remained linear disposition in the penumbra after reperfusion subsequent to cerebral ischemia.Octanol group was done at 24 h of reperfusion after cerebral ischemia.The amount of EB of octanol group((4.924?0.296)?g/g)was significantly lower than that of corresponding operation control group(5.543?0.506)?g/g.Conclusions (1)Cx43 expression is concentrated around vessels in brain.The Cx43 forms into bigger plague and the function maybe strengthens after reperfusion.Gap junction might aggravate the disruption of BBB.(2) Octanol,the specific blocker of gap junctions,could effectively prevent the permeability of BBB from increasing and has a protective effect on BBB.
