4.Evaluation of acute liver injury in mice model with different does of CCl 4

Objective To establish a simple ,stable acute liver injury model induced by CCl4 to observe effects of hepatocyte transplantation .Methods CCl4 plant oil with different concentration of 20% and 50% was used in mice by intraperitoneal injection , of which the dose was 2 mL/kg ,and then materials were taken at different time points respectively .Mice survival rate ,alanine amin-otransferase (ALT) ,aspartate aminotransferase (AST) and the pathological changes of the liver were detected .Results Mice sur-vival rate in 20% CCl4 intraperitoneal injection was significantly higher than that of 50% .ALT and AST in experiment group were significantly higher than that of control group ,but there was no significant difference between two experimental groups .Pathologi-cal examination showed that mice liver cells showed typical cytoplasmic ,ballooning ,scattered punctate ,piecemeal necrosis and in-flammatory cell infiltration in 20% CCl4 intraperitoneal injection ;while in 50% CCl4 ,there was obvious fibrosis ,in addition to the mentioned heavier lesions .Conclusion 20% -50% CCl4 intraperitoneal injection in 2 mL/kg dose can induce different degrees of relatively stable liver injury ,and its concentration determines the degree of liver injury .Acute liver injury induced by 20% -50%CCl4 was an ideal model for hepatocyte transplantation experiment .

To explore the clinical significance of changes in maximal expiratory flow in 50% and 25% vital capacity (Vmax50% & Vmax25%) before and after bronchodilator reversibility testing in patients with asthma.Forced expiratory volume in one second (FEV1),Vmax50% and Vmax25% were measured before and after bronchodilator reversibility testing in 118 patients with asthma and 82 with chronic obstructive pulmonary disease (COPD).The rate of positive reversibility in Vmax50% was significantly higher than that in FEV1 in 118 asthmatics (x2 =7.995,P =0.007).The rates of positive reversibility in Vmax50% and Vmax25% were significantly higher in asthmatics than those in COPD patients (x2 =9.335,P =0.009).

Objective To study the anti-tumor effect of 5-aza-2′-deoxycytidine(5-aza-dC) on breast carcinoma xenografted in nude mice. 8.Methodological evaluation on ELISA for detecting concentration of apo A-V in human plasma

Objective To establish an ELISA to detect the concentration of apo A-V in human plasma and evaluate the sensitivity and stability of the method.Methods The standard curve of recombinant human apo A-V, the lowest detectable concentration, the day-to-day variation and between-plate-within-day variation, the interference of other non-specialized apolipoprotein were examined, and the reference range was established by detecting the plasma concentration of apo A-V in 505 healthy subjects.Results The linear range of the established method was 5.86 - 187.5 ng/ml with lowest detectable concentration of 2.34 ng/ml. The interferences for apo A-V detection due to Lp(a), apo AI and apo B was never found in our study. The day-to-day variation and between-plate-within-day variation were less than 10% respectively.Conclusion An ELISA for detecting the concentration of apo A-V in human plasma was established and the method can serve as a useful tool for research on mechanism and function of apo A-V in TG metabolism.
9.Diagnosis and treatment of urothelial tumors in multiple organs

Objective To explore the experiences of the diagnosis and treatment of urothelial tumor in multiple organs.Methods Clinical data of 10 patients with urothelial tumor in multiple organs were retrospectively reviewed.Urothelial tumors were found in two or more organs at the same time by B ultrasound,IVU,R-P,CTU,MRU,cystoscopy,ureteroscopy and so on before operation.Results 6 cases were operated by radical total nephroureterectomy and partial cystectomy,3 cases were operated by radical total nephroureterectomy and cystectomy with urinary diversion,1 case was operated by partial ureterectomy and total cystectomy.8 of them were alive,1 case was operated by total urethrectomy because of tumor recurrence in the posterior urethra,one died of metastasis tumor 18 months after operation,and the other died 32 month after operation.Conclusions Combined use of various kinds of the diagnostic means (ultrasound,IVU,R-P,CTU,MRU,cystoscopy,ureteroscopy) are important for the diagnosis of urothelial tumor in multiple organs.It needs to select the operate mode according to the tumor staging and grade and the patient's condition.Reinforcement surveillance and close follow up is required after operation.
10.Peripheral blood leukocyte double strand RNA-dependent protein kinase gene expression in patients with systemic lupus erythematosus

Objective To investigate the expression of the double-stranded RNA-dependent protein kinase (PKR) gene in the peripheral blood leukocyte of patients with systemic lupus erythematosus (SLE),and to evaluate the relationship between the gene expression and the disease activity.Methods The clinical data of 100 SLE patients,40 non-SLE patients with rheumatic diseases,and 40 normal controls were collected.Total RNA was extracted from the peripheral blood and then reverse transcribed into cDNA.Sybr green dye based real-time quantitative PCR method was used to compare the expression levels (indicated as 2-△Ct value) of PKR in the three groups.Results (1) The 2-△Ct value of PKR expression level in the SLE patients was (14.69 ± 7.62),which was significantly higher than those in the non-SLE patients (5.09 ±4.73,P =0.012) and normal controls (4.79 ± 3.49,P =0.005).(2) The 2-△Ct value of PKR expression level in the SLE patients with severe activity was (22.57 ±2.61),which was significantly higher than those in the SLE patients with mild activity and no activity(12.94±2.41,P =0.000 ;8.85 ± 2.17,P =0.000).(3) The 2 △Ct value of PKR expression level in the SLE patients with lupus nephritis was significantly higher than that in the SLE patients without lupus nephritis (16.85 ± 7.32 vs 8.35 ± 2.04,P =0.034).(4) The 2-△Ct value of PKR was correlated with the systemic lupus erythematosus index(SLEDAI) scores(r =0.32,P =0.000),WBC (r =0.46,P =0.000),Hb (r =-0.22,P =0.035),the quantitation of urine protein in 24 hours (r =0.21,P =0.000),HDL-C (r =0.21,P =0.022),and anti-RNP antibody (rs =-0.21,P =0.025).Conclusions The expression of PKR in the SLE patients is up-regulated,especially in those with severe activity.The expression level of PKR gene is associated with SLE disease activity.
