1.Aerobic exercise protects cardiac function of T2DM mice by activation of PI3K (p110α)/Akt signaling pathway

AIM: To study the protective effect of aerobic exercise on cardiac dysfunction in mice and its mechanism, and to provide theoretical and practical basis for the exercise therapy of diabetic cardiac dysfunction .METH-ODS:The mice were divided into normal control non-exercise (NNC) group, normal control exercise (ENC) group, dia-betic non-exercise (NDM) group and diabetic exercise (EDM) group.At the end of the experiment , the cardiac function was evaluated by echocardiography .The pathological changes of the myocardial tissues and the development of fibrosis were observed.The mRNA expression of ANP, and the protein levels of PI3K (p110α) and Akt were determined.RESULTS:The decrease in cardiac function of diabetic mice was observed , and the cardiac function recovered after exercise interven-tion (P<0.05).Under light microscope with HE and Masson staining , the myocardial structure in NDM group was in ex-treme disorder , cell arrangement was not neat , and the degree of fibrosis increased , but the myocardial damage was im-proved in ENC group .Compared with NNC group , the mRNA expression of ANP in the myocardium of diabetic mice was up-regulated (P<0.05).The protein levels of PI3K (p110α) and Akt were decreased (P<0.05), and the cascade was inactivated.Compared with NDM group , the mRNA expression of ANP was down-regulated and the protein levels of PI 3K (p110α) and Akt were up-regulated in EDM group (P<0.05).CONCLUSION:Diabetes results in myocardial damage in mice, and reduces cardiac function .Exercise intervention alleviates the heart dysfunction induced by high glucose via activating PI3K( p110α)/Akt signaling pathway to protect the structure and function of the myocardium .
3.Comparison of Preventive Effect of Maternal-Infant Hepatitis B Virus Vertical Transmission by Symphysial Immunoprophylaxis

Objective To investigate the efficacy by inoculated with hepatitis B immunoglobulin(HBIG) and hepatitis B vaccine in dif-ferent doses and time points to prevent maternal-infant vertical transmission of hepatitis B virus(HBV).Methods Gravidas positive for HBV were selected and measured HBV DNA,according to difference HBV DNA degrees divided into A,B,C groups.Every group was randomly divided into 4 groups using a variety of combined immunity methods to compare its efficacies.Results There were no significant differences in the positive rate of HBV and HBsAb in A,B groups by statistics analysis.There were significant differences in the positive rate of HBV and HBsAb in group C by statistics analysis.Conclusion According to the different HBV DNA take different combined immunodeficiency approach to prevent maternal-infant vertical transmission.

6.Curative effects of alfacalcidol combined with losartan potassium tablets in the treatment of early diabetic ;nephropathy

Objective To discuss the curative effects of alfacalcidol combined with losartan potassium tablets in the treatment of early diabetic nephropathy (EDN).Methods 90 EDN patients in our hospital were chosen and randomly divided into the observation group and control group (45 cases in each group).The control group was given losartan potassium tablets treatment,while the observation group was given alfacalcidol combiend with telmisartan treatment.All the two groups were treated for 3 months.Before and after treatment,the fasting blood glucose (FBG), 24h urine trace albumin quantitative (UAER),24h urine protein (24h pro),serum creatinine (SCr),25 -hydroxyl vitamin D [2 5 (OH )D ],blood calcium (Ca2+),potassium (K+),glycosylated hemoglobin (HbA 1 C )and serum inflammatory factors[C-reactive protein (CRP),tumor necrosis factor alpha (TNF-α),interleukin-6 (IL-6 )] were observed,and the correlation between 25 (OH)D and UAER,24h pro was analyzed.At the same time,the clinical curative effects and adverse reactions during treatment were evaluated.Results In the observation group,the total effective rate was 93.3%,which was significantly higher than 71.1% in the control group (χ2 =7.601,P<0.05).In the two groups after treatment,the FBG,HbA1c,blood Ca2+and K+had no significant changes (all P>0.05).After treatment,Scr,24h pro and UAER in the two groups were all significantly reduced compared with before treatment (P<0.05),and 24h pro and UAER in the observation group were significantly lower than those in the control group (t=6.296,11.530,all P<0.05),but Scr had no statistically significant difference between two groups (t=0.331,P>0.331).After treatment,25(OH)D in the observation group decreased significantly compared with before treatment (t=12.000,P<0.05),which was significantly higher than that in the control group after treatment (t=11.278,P<0.05).Compared with before treatment,25(OH)D in the control group had no significant change after treatment (t=0.436,P>0.436).Pearson correlation analysis showed that 25 (OH)D level was negatively correlated with 24h pro and UAER (r=0.483,0.778,all P<0.05 ).In the two groups after treatment,the CRP, TNF-αand IL-6 levels significantly reduced (P<0.05 ),which in the observation group decreased more obviously (all P<0.05).In the two groups,no serious adverse events were found,the difference was not statistically significant (χ2 =0.212,P >0.151 ).Conclusion Alfacalcidol combined with losartan potassium tablets can significantly reduce the proteinuria levels of EDN patients and inflammation,which has better clinical curative effects and higher safety.
7.Methodological and Applicability Study of Rat Skin Anaphylactoid Test

Objective To study the sensitivity and reliability of rat skin anaphylactoid test method, detect and evaluate the anaphylactoid reaction of traditional Chinese medecine injection. Methods The condition of rat skin anaphylactoid test was optimized by studying the influencing factors of sensitivity and reliability of test with C48/80 as a tool drug, Tween80, endotoxin, China cobra toxin, trichosanthin injection, Tanreqing injection and Xuesaitong injection were investigated. Results The best conditions of rat skin anaphylactoid test was as follows:intrademal inject the drug with insulin syringe, 50-100μL per point, immediately inject 0.5%Evans blue dye 1 mL though caudal vein, 15 min later, kill the rat by carotid artery bleeding, clip dorsal skin to do the test. With this method, Tween80, endotoxin, China cobra toxin and trichosanthin injection all can induce blue stain in rat skin. Tanreqing injection showed no blue stain at the clinical dose. Xuesaitong injection although can induce blue stain in rat skin at the clinical dose, but the results cannot exclude the interference of its pharmacological function. Conclusion The method is simple with short test cycle, less dose of test drug, high detection sensitivity and good reproducibility, but some drug can show false positive result due to its own property.
8.ENHANCING ADHESION OF TETRAHEDRAL AMORPHOUS CARBON FILMS

Objective The high energy ion bombardment technique is applied to enhancing the adhesion of the tetrahedral amorphous carbon (TAC) films deposited by the filtered cathode vacuum arc (FCVA). Methods The abrasion method, scratch method, heating and shaking method as well as boiling salt solution method is used to test the adhesion of the TAC films on various material substrates. Results The test results show that the adhesion is increased as the ion bombardment energy increases. However, if the bombardment energy were over the corresponding optimum value, the adhesion would be enhanced very slowly for the harder material substrates and drops quickly, for the softer ones. Conclusion The optimum values of the ion bombardment energy are larger for the harder materials than that for the softer ones.
9.Sleeve resection of bronchus and pulmonary artery for central type lung cancer

Background and purpose:In recent years,sleeve resection of bronchus and pulmonary artery has been more extensively applied in treatments of lung cancer patients with poor pulmonary functions.This kind of surgery meets the basic surgical principles of lung cancer being maximum both in resecting the carcinoma of the lungs and preserving pulmonary functions,has a good prospect.Therefore,we summarize our recent experience in treating central type lung cancer by this method and discuss its principles and methods in the application of the surgical operation.Methods:From Dec.1993 to Dec.2003,63 patients with central type lung cancer underwent sleeve lobectomy of bronchus and pulmonary artery,including sleeve resection of bronchus and pulmonary artery in 43 cases,wedge resection of bronchus and pulmonary artery in 3 cases,sleeve resection of bronchus and wedge resection of pulmonary artery in 17 cases.The surgical procedures of sleeve resection included sleeve resection of right upper lobe and pulmonary artery in 20 cases,sleeve resection of left upper lobe and pulmonary artery in 23 cases,and the length of pulmonary artery that had been respected is from 1.5cm to(4.5cm),the average length is 2.5cm.The surgical procedures of wedge resection included wedge resection of right upper lobe and pulmonary artery in 2 cases, wedge resection of left upper lobe and pulmonary artery in 1 case.The group of sleeve resection of bronchus with wedge resection of pulmonary artery included sleeve resection of right upper lobe and wedge resection of pulmonary artery in 5 cases,sleeve resection of left upper lobe and wedge resection of pulmonary artery in 12 cases.Results:No case of death and anastomotic leak occurred.Conclusions:Because sleeve resection of bronchus and pulmonary artery can not only effectively resect the tumor but also maximally preserve the pulmonary function at the same time,it enlarges the surgical indications of lung cancer,and is an effective and safe method for the treatment of lung cancer.

AIM: To study mitochondrial mass and structural protein changes in dexamethasone (DEX)-mediated mouse thymocyte apoptosis process.
