2.Effect of vimentin expression on contraction of hypertrophic scar

Objective Vimentin is one of the proteins of cytoskele to n and cell movement. To investigate the effect of gene expression of fibroblast vimentin on the formation and contraction of hypertrophic scar in this report. Methods According to the results of differently expressed genes in hypertrophic scar by gene microarray, Vimentin, one of the most important g enes , was selected and made into oligonucleotide probe. 24 cases of hypertrophi c scar and 24 non-hypertrophic scar and 12 normal skin were used and these scar were taken on 3, 6, 9 and 12 months after burned. Frozen section and cultured f ibroblasts were made to detect the expression of the gene by in situ hybridizati on. Results Expression of vimentin was defected in scar tissue, but those in the hypertrophic scar on 3 and 6 months after burn were significan tly stronger than those in the hypertrophic scar on 9 and 12 months after burn a nd non-hypertrophic scar. Conclusions Over-expression of cyto skeletal relative genes causes the contraction of scar and vimentin acts leading and key functions.
3.Research advances in the pathogenesis of radiation-induced myocardial damage

The Medline and CNKI databases were searched with the key words of radiation-induced myocardial damage,damage pathway,pathogenesis,and intervention,and 37 articles were obtained.The pathogenesis of radiation-induced myocardial damage may be related to various mechanisms such as oxidative stress,transforming growth factor-β,renin-angiotensin system,mast cells,and endothelial dysfunction.The pathogenesis of radiation-induced myocardial damage is a complex process involving various mechanisms,and currently,there are ongoing studies on the cellular and molecular mechanisms involved in its pathogenesis and interventions.
4.High Wet Package Rate of Rent Surrgical Instrument:Reasons and Countermeasures

OBJECTIVE To explore the reasons of high wet package rate of the rent surgical instrument and to look for improvement method.METHODS A total of 2872 packages of hospitals in Chongqing city were investigated during the last two years.RESULTS Among 2872 rent surgical instrument packages,1064 packages were found to be damp(37.05%).CONCLUSIONS Unstandardized and too large packaging are the main reasons.After elimination of above two reasons,the damp rate will be decreased.
5.The influence of different operation film drape styles on incision infection in cesarean section

Objective To analyze the influence of different operation film drape styles on incision infection in cesarean section . Methods 396 patients of cesarean section in this hospital were divided into two groups from October 2011 to April 2012 .There were 198 patients in group A and 198 in group B .The operation film was pasted after operation sheet draped in group A ,the opera-tion film was pasted before operation sheet draped in group B .Subcutaneous tissue fluid was dipped after skin incision and before skin suture in every patient ,and the tissue fluid was bacterial cultured .Observed dressing infiltration degree after operation ,and fol-low up wound healing of incision after 5 to 7 d .Results The preoperational tissue fluid after skin incision had no bacterial growth in two groups (P>0 .05) .The dressing infiltration in group B (47 cases) was higher than in group A (16 cases) (P<0 .05) .The postoperative tissue fluid had one bacterial growth in group A ,and 4 in group B .The bacterial growth rate in group B was higher than in group A ,but there was no statistically significant (P>0 .05) .There was no postoperative infection happened in group A , and one in group B ,the difference was no significant between two groups (P>0 .05) .Conclusion Two operation film drape styles in cesarean section have no influence on postoperative incision infection ,but the dressing infiltration degree is different .So compre-hensive prevention and control is important to prevent postoperative incision infection .
6.MRI evaluation of prognosis for hypoxic ischemic encephalopathy in term neonates

Objectives To explore the characteristics and value of magnetic resonance imaging (MRI) in predicting prognosis of hypoxic ischemic encephalopathy (HIE) in neonatal period, at 4 months and 4 years old. Methods Twenty-four patients with HIE were examined by MRI. Their MRI results in the neonatal period, at month 4 and year 4 and neurological functions at year 4 were compared. Results Periventricular signal alterations and deep gray matter involvement were common in HIE neonates. The neonates with deep gray matter involvement usually had neurological malfunction and poor prognosis. The patients with encephalomalacia and periventricular leukomalacia at 4 months and 4 years old also had poor prognosis. The abnormal MRI ifndings in HIE children at 4 month and 4 year old predicted the occurrence of neurological malfunction. Con-clusions The MRI of infant at 4 months old is important in prediction of neurological malfunction and provides guidance of clinical intervention for children with HIE.
7.The impact of pregnancy on differentiated thyroid cancer

Objective To study the impact of pregnancy on differentiated thyroid cancer. Methods Retrospective comparative analysis were made between pregnancy-related thyroid cancer patients ( including nine months before delivery and one year after delivery) and the age-matched non-pregnancy-related thyroid cancer patients treated in the Department of head and neck, Fudan University Shanghai Cancer Center. Results There were 125 cases of pregnancy-related thyroid cancer patients from 1998 to 2008, including 28 cases (22.4%) of neck recurrence and metastasis after the surgery of thyroid cancer, meanwhile there were 691 cases of the agematched non-pregnancy thyroid cancer patients, including 90 cases ( 13% ) of neck recurrence and metastasis after the surgery of thyroid cancer. The rate of neck recurrence and metastasis after the surgery of thyroid cancer in the pregnancy-related group was higher than that in the non-pregnancy related group ( P < 0.05). The maximum diameter of tumor in the pregnancy-related group and the non-pregnancy group is 2. 1 cm and 2.6cm respectively.There were 19 cases of multi-focal cancer ( 19.6% ) in the pregnancy-related group and 58 cases (9. 6% ) in the non-pregnant group, P <0.05. There were 23 patients (23.7%) of extrathyroid invasion in the pregnancy-related group and 65 patients ( 10.8% ) in the non-pregnant group, P <0.05. There were 67 cases (53.6%) and 35 cases (28%) of the central and lateral cervical lymph node metastasis in the pregnancy-related group respectively and 296 cases (42.8%) and 117 cases (16. 9% )in the non-pregnant group, P <0.05. There were 19 cases (15.2%) of extralymph invasion in the pregnancy-related group and 32 cases (4.6%) in the non-pregnant group, P < 0.05. Conclusions The incidence of Extrathyroid invasion, multi-focal cancer, cervieal lymph node metastasis and extralymph invasion of the differentiated thyroid cancer is higher in pregnancy-related patients.Pregnancy may deteriorate the prognosis of thyroid cancer. For patients after the surgery of thyroid cancer, pregnancy may increase the risk of neck recurrence and metastasis. However, pregnancy does not affect the stage of thyroid cancer and the prognosis is still good.
8. Preparation of curcumin thermoresponsive nanogels and their characteristics

Objective: To prepare curcumim thermoresponsive nanogels and study its release characteristics in vitro. Methods: Poly (N-isopropyl acrylamide) (PNIPAm) thermoresponsive nanogels were synthesized by emulsion polymerization method, and then curcumin loaded-PNIPAm nanogels were prepared. The characteristics of PNIPAm nanogels, the drug loading, and the drug release in vitro were investigated. Results: Atomic force micriscopy (AFM) observation showed that PNIPAm nanogels were spherical in shape with homogenous size. Moreover, PNIPAm No. 5 showed a high drug loading (27%). In vitro release study revealed that the 24 h accumulative in vitro release reached 75.2%. Conclusion: The method used to prepare curcumin thermoresponsive nanogels is simple. It is expected to become a novel vehicle potentially nanoparticle drug delivery system of curcumin.
9.Magnetic nanoparticles versus a commercial kit for apolipoprotein E gene polymorphism analysis

BACKGROUND:Relative to blood samples, mouth swab samples are more beneficial for apolipoprotein E gene polymorphism analysis among large cohorts. However, agreement has not yet been reached about how to extract genomic DNA form mouth swab samples.
OBJECTIVE:To develop an appropriate method to extract genomic DNA form mouth swab samples, which are suitable for apolipoprotein E gene polymorphism analysis.
METHODS:Fifty mouth swab samples from patients with sporadic Alzheimer’s disease were col ected. Magnetic nanoparticles and PicoDNA trace nucleic acid extraction kit were used to extract genomic DNA form mouth swab samples. And the purity and concentration of the genomic DNA extracted by the two methods were analyzed. Then PCR amplifications and DNA electrophoresis were performed to confirm whether the genomic DNA was able to amplify desired DNA fragments. DNA sequencing was applied to analyze apolipoprotein E gene polymorphisms.
RESULTS AND CONCLUSION:Genomic DNA extracted by the two methods was of high purity. The concentration of genomic DNA extracted by magnetic nanoparticles was higher than by PicoDNA trace nucleic acid extraction kit, and the difference had statistical significance (P<0.05). Al the genomic DNA were able to performed PCR amplifications to obtain desired PCR products, but results of DNA electrophoresis showed that DNA fragments were more clear by nanoparticles method. The results of DNA sequencing were the same by the two methods. The distribution ofε2,ε3,ε4 genotypes of apolipoprotein E gene was 6%, 71%, 23%, respectively. Magnetic nanoparticles were better than PicoDNA trace nucleic acid extraction kit for extracting genomic DNA form mouth swab samples for apolipoprotein E gene polymorphism analysis.
10.Expression of human CTLA4 extracellular domain in Bacmid-baculovirus expression system

Objective To express human cytotoxic T lymphocyte associated antigen-4 (CTLA4) extracelluar domain in Bacmid-baculovirus expression system. Methods The cDNA of human CTLA4 extracelluar domain was isolated from plasmid pUC19-CTLA4Ig by PCR.The specific cDNA was subcloned into plasmid pFastBacl. The insertion was confirmed by DNA sequencing, and then transposed into Hz8 Bacmid in DH10B. The recombinant Bacmid was used to transform Hz insect cells. The supernatant and cellular lysate were analyzed by using SDS-PAGE. Results A specific protein in the cellular lysate of Hz insect cells containing human CTLA4 extracelluar domain with MW38 x 103 was found. Conclusion Human CTLA4 extracellular domain is expressed in the Bacmid-baculovirus expression system.
