Chinese Journal of Laboratory Medicine 2013;36(10):942-946
Objective To establish the first national standard of Cystatin C.Methods The candidate standard was prepared from human recombinant Cystatin C,diluted and dispensed aseptically.Homogeneity and stability study were carried out on the automated biochemical analyzer. The target value was assigned by six laboratories using widely recognized immumoassay under strict conditions, and traceable to ERM-DA471.Commutabilities of the candidate standard and its saline dilutions were evaluated according to EP14-Evaluation of Matrix Effects.Excel 2007 was used to analyze the results. Results The preparation had good immunological activity and was proved to be homogeneous and stable (6 months sealed at 2-8 ℃,30 days sealed at room temperature and 37 ℃,30 days open-vial at 2-8 ℃).The assigned value of the preparation was (4.47 ± 0.25) mg/L (coverage factor k =2).The candidate standard and/or its saline dilutions have commutability on the 10 evaluated systems. Conclusion The preparation meets the requirements of national secondary standard and could be used in quality control and evaluation of Cystatin C assays in China.

Objective To study the effect of the extract of Chrysanthemum Indicum(CI)on the expression of tumor necro-sis factor-α (TNF-α) and neutrophil function in chronic bronchitis (CB)rats and to explore the therapeutic mechanism for chronic bronchitis. Methods The extract of CI was prepared. Wistar rats were randomly divided into blank control group, model group, Chuanxiling group, and low-, middle-and high-dose CI extract groups. Rat model of CB was established by intratracheal injection with low-dose lipopolysaccharide. After drug intervention, the expression of TNF-α in rat serum and bronchoalveolar lavage fluid was detected by ELISA method. Phagocytic function of the neu-trophil and respiratory burst of the rats were measured by flow cytometry (FCM). Results Compared with the blank con-trol group, the phagocytic function of the neutrophil, respiratory burst of the rats, and the expression of TNF-α in rat serum and bronchoalveolar lavage fluid of the model group were increased significantly. CI extract significantly decreased the abnormal rising of the above indexes. Conclusion Down-Regulation of TNF-α expression, and decrease of the neutrophil phagocytic function and rat respiratory burst may be one of the therapeutic mechanisms of Chrysanthemum In-dicum extract for the treatment of CB.

BACKGROUND: Silver-matrix composite is usually used as a kind of electrical conductive matedal, which has excellent properties in electrical and heat conducting. MoS2, which used as a common solid lubricating material, has unique anti-frictional and lubricating properties in the process of wearing. Ag-MoS2 composites using in the area of electrical sliding wear were fabricated by adding MoS2 to the silver-matrix. OBJECTIVE: Ag-MoS2 composites with different content of MoS2 were produced to gain the mixture ratio for best properties by the experiments of mechanical and wear test. DESIGN, TIME AND SETTING: The controlled experiment was performed at the Department of Matedal Science and Engineering, Hefei University of Technology from April to May 2008. MATERIALS: Silver powder (Bengbu Gold and Silver Article Factory, China) and MoS2 powder (Sinopharm Chemical Reagent Co., Ltd., China) were used in this study. METHODS: Ag-MoS2 composites for five different contents were fabricated by powder metallurgy method, which included the processes of powder mixture, drying, initial pressure, sintering, and second pressure. MoS2 contents were the MoS2 composite of 12.0%, 13.5%, 15.0%, 16.5%, 18.0% silver- MoS2. MAIN OUTCOME MEASURES: Flexural strength was measured by versatility mechanical testing machine. The microstructures of the composites after sintering were observed by an optical microscope. The electrical sliding friction and wear properties were tested by the electrical wear test machine. The worn surfaces of the samples were observed by a scanning electron microscope. RESULTS: With the increase in MoS2, the flexural strength decreased, the resistivity increased, and the wear loss decreased first, and then increased, which comes the lowest point when the MoS2 content was 15%. CONCLUSION: When the content of MoS2 is 15%, the average properties was the best.

Background and purpose:Drug-resistance is the main obstacle in terms of efficacy of chemotherapy for leukemia, RNA interference(RNAi) strategy possesses the characteristics of specilization, high-efficiency and low-toxicity, and can effectively and specifically inhibit the overexpression of given gene. This study was designed to investigate the effect of small interfering RNA (siRNA) on expression of mdr1 gene and drug-resistance in multidrug-resistant human leukemia K562/ADM cell. Methods:Human multidrug-resistant leukemia cell line K562/ADM over-expressing mdr1 gene was used as the target cells, Two siRNAs (si-mdr1-1 and si-mdr1-2) targeted mdr1 gene were chemically synthesized and transfected into K562/ADM cells. Expression of mdr1 mRNA was determined by RT-PCR, P-glycoprotein (P-gp) expression was measured using flow cytometry (FCM), and the sensitivity of K562/ADM cells to adriamycin was assessed with a MTT colorimetric assay. Results:Two siRNAs (si-mdr1-1 and si-mdr1-2) specially designed in this study could markedly down-regulate the expression of mdr1 mRNA and its product P-gp in K562/ADM cells. After cells transfected with si-mdr1-1 or si-mdr1-2 for 24h and 48h, the inhibition of mdr1 mRNA expression in the cells for si-mdr1-1 was 55.5% and 22.5%; and for si-mdr1-2, 16.0% and 57.6%, respectively. Treated with siRNA for 72h, the expression intensity of P-gp in the two transfected cell lines decreased 74% and 85%, respectively. Both si-mdr1-1 and si-mdr1-2 significantly enhanced the sensitivity of K562/ADM cells to adriamycin and reversed their drug-resistance, the reversal efficiency was 2.52-folds and 1.96-folds, respectively. Conclusions:The siRNA could effectively and specifically silence the expression of mdr1 gene and overcome the drug-resistance mediated by P-gp in K562/ADM cells.

Many risk factors lead to hypohepatia and hepatic failure, causing people suffering from end-stage liver disease. The conventional treatment for end-stage liver disease is not good enough.Orthotopic liver transplantation is effective. However, the high cost, lack of liver source, immune rejection and other factors limit the large-scale clinical application. Thus, cell therapy is a good option.Studies on common cell sources for the treatment of liver disease and the induction of hepatocytes by embryonic stem cells or pluripotent stem cells have made progress. With the development of stem cell technology, cell transplantation has become a new option, which brings hope to people with end-stage liver diseasetransplantation has become a new option. It brings hope to people with endstage liver disease.

Objective To evaluate the role of early cleavage embryo in combination with embryo growth rate and morphology scoring in embryo selection in in vitro fertilization/intracytoplasmic sperm injection (IVF/ICSI) cycles. Methods Six hundred and ten IVF/ICSI cycles were randomly assigned to group A(269 cycles) and group B(341 cycles). In group A, transferred embryos were chosen according to embryo growth rate and morphology scoring by 72 h(D3) after fertilization, while early cleavage embryo was added to the selecting system in group B. The pregnancy rate and implantation rate were compared between two groups, and the clinic outcomes were compared between transfers with early cleavage embryos and without early cleavage embryos in group B. Results The pregnancy rate and implantation rate in group B were significantly higher than those in group A (P < 0.05). Transfers with early cleavage embryos also achieved much higher pregnancy rate and implantation rate in group B (P < 0.01). Conclusion Compared with embryo growth rate and morphology scoring, early cleavage embryo in combination with embryo growth rate and morphology scoring can improve the clinical outcomes in IVF/ICSI cycles.
