1.Measurement of serum levels of angiogenic factors and inflammatory cytokines in patients w ith moyamoya disease by cytometric bead array

Objective To detect the serum levels of angiogenic factors and inflammatory cytokines in patients w ith moyamoya disease and explore their roles in the pathogenesis of the disease. Methods The serum levels of vascular endothelial grow th factor (VEGF), angiopoietin -1 (Ang-1), interleukin-8 (IL-8), granulocyte colony stimulating factor (G -CSF), granulocyte-macrophage colony stimulating factor ( GM-CSF) and monocyte chemotactic protein 1 (MCP -1) in 56 patients w ith moyamoya disease and 26 healthy controls w ere measured by cytometric bead array. Results The serum levels of VEGF (2.81 ± 1.77 pg/ml vs.1.98 ±0.66 pg/ml; t = 3.081, P = 0.003 ) and IL-8 (0.89 ±0.69 pg/ml vs.0.63 ± 0.45 pg/ml; t'=2.0371, P < 0.05) in the moyamoya disease group w ere significantly higher than those in the healthy control group, and the serum level of Ang -1 in the moyamoya disease group w as significantly low er than that in the healthy control group (830.01 ±289.29 pg/ml vs.961.65 ±232.87 pg/ml; t =-2.032, P =0.045). Conclusions There are significant difference in serum levels of VEGF, Ang -1 and IL-8 betw een patients w ith moyamoya disease and healthy controls. The results indicate that angiogenic factors and inflammatory cytokines play some roles in the pathogenesis of moyamoya disease.
3.Association between mitochondrial uncoupling protein 2 gene promoter -866G>A polymorphism and ischemic stroke in diabetic patients

Objective To investigate the association of uncoupling protein 2 ( UCP-2 ) gene promoter -866G>A polymorphism and ischemic stroke in diabetic patients.Methods A total of 844 type 2 diabetic patients including 404 cases with ischemic stroke and 440 cases without ischemic stroke were selected for the 4 year prospective study,Genomic DNA was extracted from the whole blood samples of subjects,UCP-2 gene promoter -866G > A polymorphism was detected by TaqMan MGB probe method,and then the genotype and allele gene frequencies were compared.Results The risk of ischemic stroke in type 2 diabetic female patients with AA+GA genotypes of UCP-2 was higher than that with GG genotype (P<0.05),but there was no difference among male patients with three genotypes.Conclusions UCP-2 gene promoter -866G > A polymorphism increases the risk of ischemic stroke in Chinese diabetic women.

OBJECTIVE:To evaluation of the efficacy and safety of azithromycin in the treatment of acute infections METHODS:A randomized controlled multicenter clinical study was used to compare the clinical efficacy of azithromycin(200mg q d for 3~5 days) with erythromycin(500mg b i d for 5 days) RESULTS:The cure rate,effective rate,and bacterial clearance rate were 76 7%,95 0%,and 94 3% respectively for azithromycin group and were 48 3%,76 7%,and 77 4% respectively for erythromycin group with a side-effect incidence of 10 0% for azithromycin group and 39 3% for erythromycin group CONCLUSION:Azithromycin is superior to erythromycin in clinical efficacy,safety and side-effect incidence

ObjectiveTo evaluate the clinical effect of integrated Chinese and western medicine (ICWM) on the post-stroke comorbid anxiety and depression (PSCAD).Methods35 PSCAD patients were treated with ICWM therapy and scores of self-rating anxiety scale (SAS) and self-rating depression scale (SDS) were analyzed statis tically before and after treatment.ResultsThe curi ng rate of PSCAD is about 74.29% and scores of SAS and SDS were significantly d ecreased after treatment (P<0.001).Conclusions The treatment of ICWM can produce a good effect on PSCAD.
6.The diagnosis and treatment of late acute rejection after adult orthotopic liver transplantation.

OBJECTIVETo explore the diagnosis, treatment and long-term outcome of late acute rejection (LAR) following adult orthotropic liver transplantation (OLT).
METHODSA total of 398 consecutive adult patients who underwent OLT in Organ Transplant Center, the First Affiliated Hospital, Sun Yat-sen University between January 2007 and December 2012 were reviewed retrospectively. There were 48 patients (12. 1%) developed to LAR, including 43 male patients and 5 female patients, with an average age of (52 ± 13) years(18 - 70 years). The mean body mass index was (22.1 ± 4. 5) kg/m2 (15. 4 - 30. 4 kg/m2). The indications of the liver transplantation recipients included 16 cases of end-staged liver cirrhosis after hepatitis B or C(33. 3%), 14 cases with severe hepatitis (29. 2%), 9 cases of primary liver cancer(18. 5%), 5 cases of alcoholic liver cirrhosis (10. 4%), 1 case with autoimmune liver disease (2. 1%) , the other 3 cases (6. 3%). They were followed up by outpatient service, telephone and other means. Survival curves were generated with the Kaplan-Meier method and Cox proportional hazards modeling was used for predictors of mortality. Statistically significant variables found by single factor regression analysis were put into the Cox proportional hazards regression model of multivariate analysis.
RESULTSThe time-to-event was 23. 6 months after OLT which were more common in the first year to the third year post-transplant (26/48,52. 4%). Thirty-five cases were assessed as mild, 11 cases were assessed as moderate, and 2 cases were assessed as severe ,based on the Banff schema. After adjustment to the immunosuppressive regimen, the overall recovery rate reached to 81. 3%. The rate of steroid-resistant acute rejection was 11. 8% (4/34). Inadequate immunosuppression and steroid pulsation were two independent risk factors affecting the prognosis of LAR (P = 0. 008, P = 0. 003, respectively).
CONCLUSIONSLAR is an uncommon complication after OLT. Inadequate immunosuppression and steroid pulsation are the major risk factors for prognosis of LAR. Improving patient compliance and strengthening blood concentration surveillance can increase the patient survival.

7.Effects of lentivirus-mediated RWDD3 silencing on proliferation and inva-sion of human glioma U251 cells

AIM:To investigate the effect of RWDD3 gene silencing on the biological characteristics of human glioma U251 cells.METHODS: A lentiviral vector expressing RWDD3 shRNA was constructed and transfeeted into the U251 cells.The expression of RWDD3 at mRNA and protein levels was detected by real-time PCR and Western blot , re-spectively .The cell activity was determined by MTT assay .The colony formation ability was detected by the colony forma-tion assay .The cell proliferation ability was detected by BrdU incorporation assay .The cell invasion and migration were evaluated by Transwell assay .Flow cytometry was used to monitor the changes of cell cycle distribution and apoptosis .RE-SULTS:Recombinant lentivirus was successfully transfected into U 251 cells.Compared with the cells transfected with the scrambled shRNA and control cells, the cell activity, colony formation ability, and the invasive and migratory activities were inhibited, the cell cycle was arrested in G 0/G1 phase, and the apoptosis was increased in the U 251 cells transfected with RWDD3 shRNA ( P <0.05 ) .CONCLUSION: RWDD3 plays a vital role in proliferation and invasion of glioma cells.It may serve as a potential target of gene therapy for glioma .

0.05).The concentration of urinary KYNA,metabolite of the KYN,was significantly lower in SHRs compared to WKYs(7.8?1.8 vs 19.9?3.5 ?mol/24 h P=0.013).Both KAT activity in renal cortex and KYNA content in urine were negatively correlated to blood pressure(r=-0.418,P=0.023;r=-0.723,P=0.001).Conclusion The declined activity of KAT in renal cortex and the deficiency of KYNA concentration in urinary may affect blood pressure regulation in SHR by renal metabolite of the KYN.
9.Clinical study on 52 patients of post-stroke depressive disorder treated with traditional and western medicine

ObjectiveTo evaluate the clinical effect of integrated traditional and western medicine in the treatment of post stroke depressive disorder (PSDD). MethodsThe data of 52 patients of PSDD evaluated on self rating depression scale(SDS) before and after treatment were analyzed statistically. ResultsThe curing rate of PSDD is about 78.85%.There was an apparent decrease in the level of SDS before and after treatment(P<0.001). ConclusionsThe treatment of integrated traditional Chinese and western medicine can produce a good effect on PSDD.
10.Construction of miRNA expression vector dual-targeting on HIF-1α/survivin genes and its effects on proliferation of pancreatic cancer Panc-1 cell line.

OBJECTIVETo design and construct miRNA expression vector dual-targeting on HIF-1α and survivin genes and to investigate its effects on proliferation of human pancreatic cancer cells.
METHODSThe specific pre-miRNA single strand DNA oligos for HIF-1 α and survivin genes were designed and synthesized, then via annealing and ligating with pcDNA6.2-GW/EmGFP-miR plasmids in order, two kinds (eight in total) of miRNA expression vectors for HIF-1α and survivin genes were constructed. The vectors, which were most effective to knockdown target genes, were screened with real-time RT-PCR and combined by chaining technology to construct dual-targeting plasmid. The recombined dual-targeting plasmid, mono-targeting plasmids and negative plasmid were transfected into Panc-1 cells, the suppression effect on two genes was identified by real-time RT-PCR, Western blot and MTT assays.
RESULTSThe miRNA expression plasmids anti-H, anti-S and anti-H+S were successfully constructed by identification of sequencing analysis, and they were able to effectively inhibit the target genes expressing. MTT assays showed that the inhibition effect of dual-targeting vector anti-H+S was higher than that of mono-targeting vectors anti-H and anti-S 72 h after transfection (P<0.05).
CONCLUSIONThe effective miRNA expression vector dual-targeting on HIF-1α and survivin genes has been successfully constructed. The inhibition effect on proliferation of pancreatic cancer Panc-1 cells by dual-targeting plasmid was higher than that by mono-target plasmids.
