1.The expression and clinical signification of tM2-PK in colorectal cancer

Objective To investigate the expression and clinical signification of tumor M2 pyruvate kinase in pa-tients with CRC, and to try to find a reliable and atraumatic method to predict the progress and pronosis in the CRC. Methods ELISA method was used to determine the serum levels of tM2-PK in 68 patients with CRC,40 pa-tients with colorectal polyps and 40 cases of normal subjects; immunohistochemistry ( IHC ) methods were respec-tively used to detect the expression of tM2-PK in tissues. Results ① The level of serum tM2-PK in patients with CRC was significantly higher than the group of patients with polyps and the normal subjects ( P<0.05 ) . The serum tM2-PK levels was closely correlated to tumor size and Duke stage.②The positive rate of tissue tM2-PK was signif-icantly higher in CRC than in colorectal polyps and normal subjects ( P<0.05 ) . Tissue tM2-PK levels was closely correlated to tumor size,Duke stage and differentiation grade. ③Serum tM2-PK levels was correlated to tissue tM2-PK levels (r=0.357,P=0.003). Conclusion Combined detection of expression of tM2-PK can provide the basis theory for the diagnosis and prognosis evaluation of CRC.
2.The expression and clinical significance of Livin and Caspase-9 in duodenal adenocarcinoma

The expressions of Livin and Caspase-9 were detected by immunohistochemical technique in 60 duode-nal adenocarcinoma tissues and 30 adjacent normal tissue. The positive expression rate of Livin in duodenal adeno-carcinoma tissues was significantly higher than that of adjacent normal tissue,and the expression of Caspase-9 was lower than that of adjacent normal tissue ( P<0.01 ) . The expression of Livin was negatively correlated with the ex-pression of Caspase-9 ( P<0.01 ) . The expression of Livin and Caspase-9 was closely correlated with the progress of pathological changes, degree of development,lymph nods metastasis,and clinical stages,but not with sex,age and tumor size ( P>0.05 ) . Livin and Caspase-9 were associated with occurrence and development of duodenal adeno-carcinoma,and the expression of the above two were negatively correlated. Livin and Caspase-9 may be considered as reliable markers for clinical diagnosis,therapy efficacy and the prognosis of duodenal adenocarcinoma.
3.Relation between breast cancer susceptibility gene 1 expression and the efficacy of platinum-based chemotherapy in colorectal cancer

Objective To explore the expression of breast cancer susceptibility gene 1 (BRCA1) in human colorectal cancer and its correlation with the efficacy of platinum-based chemotherapy.Methods Seventy-eight cases of colorectal cancer patients treated with platinum-based chemotherapy after surgery were collected.The surgical specimens of them were taken.Fourteen normal colonic mucosa specimens and 12 non-cancerous tissues of colorectal cancer specimens were obtained.The expression of BRCA1 in tissues was detected by immunohistochemistry and analyzed by x2 test.The colorectal cancer patients were followed up for survival time.Comparison of survival analysis was performed by Kaplan-Meier survival curves and Log-rank test.Results The positive rate of the BRCA1 expression in colorectal cancer tissue (52.6 %,41/78) was lower than that of para-cancer tissue (11/12,x2 =6.518,P=0.011) and normal colonic mucosa tissue (13/14,x2 =7.949,P=0.005).The poorer the differentiation of colorectal cancer,the lower the positive rate of BRCA1 (x2=14.160,P=0.001).The median disease-free survival time of BRCA1 negative colorectal cancer patients was 51.0 months (95 % CI:47.7 to 54.4 months),which was longer than that of BRCA1 positive patients (45.0 months,95 %CI:36.6 to 53.4 months,x2 =4.367,P=0.032).Conclusions Receiving oxaliplatin based chemotherapy may be a survival benefit for BRCA1 negative colorectal cancer patients.The expression of BRCA1 may be an index for chemotherapy options and prognosis evaluation for colorectal cancer patients after surgery.

5.Effect of MACC1 down-regulation on proliferation of gastric adenocarcinoma cell line

Objective To study the effects of MACC1 down-regulation on the growth of gastric adenocarcinoma cells. Methods siRNA (MACC1-siRNA1 and MACC1-siRNA2) that can transiently silenced MACC1 was designed, syn?thesized and transfected into MGC-803 cells by lipofectamine 2000. Non-specific siRNA was transfected to be used as nega?tive control. The efficiency of MACC1 depletion was determined by Real-time quantitative PCR. MTT, colony formation and flow cytometry assay were performed to examine cell proliferation. The expressions of MACC1, P21,CDK4, CCND1 and c-myc were determined by Western blot. Results Compared with cells in negative control group, transiently silencing MACC1 decreased the expression of MACC1 in MGC-803 cells shown by Real-time PCR. MACC1 downregulation drastical?ly changed the proliferation, colony formation and cell cycle of gastric adenocarcinoma cells in vitro ( P<0.05). The expres?sions of MACC1 , CDK4, CCND1 and c-myc proteins in cells of MACC1 silence group were much lower while P21 expres?sion level was much higher than those in negative control. Conclusion Down-regulation of MACC1 result in blocking cell cycle, inhibiting proliferation of MGC-803 cells. So it may serve as a promising target in the treatment of gastric cancer.
6.Rapid diagnosis of autosomal dominant polycystic kidney disease by detecting microsatellite SM7 with modified PCR amplification

Objective To diagnose Chinese autosomal dominant polycystic kidney disease (ADPKD) rapidly by detecting microsatellite SM7. Methods Microsatellite SM7 was amplified by modified PCR in two temperatures. Gene linkage analysis was performed. Rusults DNA analysis predicted 9 asymptomatic members as PKD1 gene carriers. Conclusion The presymptomatic gene diagnosis could be done rapidly by detecting microsatellite SM7 with modified PCR amplification in two temperatures.
7.Experimental study on properties of stress-relaxation of the middle and lower cervical spine

Objective To study the properties of stress relaxation of the middle and lower cervical vertebrae, and to evaluate the effects of laminotomy and anterior diskectomy and spinal fusion (ASF) on the properties . Methods The dynamic responses of stress relaxation of six intact and post operative fresh human cadaveric cervical vertebrae were measured in vitro. Results Under the condition of the constant strain, the functional equations and the curves of stress relaxation of the six intact and post operative fresh human cadaveric cervical vertebrae were obtained. The Gex(t)(the ratio of the ensuing instant stress to the original stress) of the post operative ones was significantly bigger than that of the intact ones; The Gex(t) of the laminotomic ones was significantly bigger than that of the ones having undergone the anterior discectomy and spinal fusion(ASF). Conclusion Either in flexion or in extension, cervical vertebrae have the similar behaviors of stress relaxation. The laminotomy and anterior discectomy and spinal fusion (ASF) all reduce the stress relaxation effect of the cervical vertebrae, but ASF is more significant.
8.Summary of orthotopic cardiac transplantation in 28 cases

Objective To summarize the clinical data of cardiac transplantation in 28 patients of our hospital,and study the effectiveness of operation and the methods of immunosuppressive regime.Methods From January 2000 to May 2005,in our center,28 patients were subjected to orthotopic cardiac transplantation.In early stage(before 2003) ciclosporin A and mycophenolate mofetil were used for immunosuppression.In late stage(after 2003) Daclizumab and mycophenolate mofetil were used for immunosuppression.Results Orthotopic cardiac transplantations were done successfully in 28 patients.There was no haemorrhage,twist and right heart insufficiency postoperatively.In the patients before 2003,the complications of hepatic-renal inadequacy to varying degrees occurred in 6 cases and infection in 3 cases respectively.In the patients after 2003,there were no complications of hepatic-renal inadequacy and infection.Conclusions Cardiac transplantion is an effective way for end stage cardiomyopathy.Immunosuppressive effect of Daclizumab and mycophenolate mofetil is satisfactory.
9.Influence of age and gender on the healing of three-and four-part proximal humerus fractures following cloverleaf plate fixation

Thirty-two patients with comminuted proximal humerus fractures(Neer Ⅲ or Ⅳ) were admitted to Department of Orthopedics,First Hospital of Jilin University from March 2001 to April 2006.The patients consisted of 19 males and 13 females,aged 19-70 years,including 15 of 19-49 years and 17 of 50-70 years.All patients underwent treatment of cloverleaf plate-screw fixation.During the follow-up for 8 months,union time and scores from Constant & Murley functional scale were similar between male and female groups.Compared with patients aged 50 years,and the scores from Constant & Murley functional scale were significantly lower(P 50 years old is relatively longer with unfavorable functional recovery in the shoulder joint.
10.Mechano-chemical Signal Pathway of Endothelial Cells Migration Induced by Fluid Shear Stress

The migration of endothelial cell induced by fluid shear stress is related to many physiological and pathological responses in organism.The redistribution of membrane receptors and proteins,caused by shear stress,is a vital step for cell migration,followed by chemical reactions and signal transduction.Then cell morphological changes are induced,such as polarity,protrusion,and adhesion,which lead to the migration of endothelial cell at last.The goal of this paper is to review what is known about the mechanical models of endothelial cell migration and the responses of cells to fluid flow.It is helpful for better understanding the intrinsic mechanism of mechano-chemical signal pathway of the migration of endothelial cells induced by fluid shear stress.
