1.Correlation between polymorphism of CYP2D6 gene with effect of tamoxifen in patients with breast cancer

Objective To investigate the correlation of CYP2D6 gene polymorphism and serum tamoxifen (TAM) and its active metabolites (4-hydroxy tamoxifen,4-OH-TAM) in terms of prognosis of breast cancer patients.Methods The oral mucosas and serum were obtained from 200 breast cancer patients receiving oral TAM from Jan 2008 to Oct 2010.Real-time RT-PCR was used to determine CYP2D6* 10 gene polymorphism.The relationship between CYP2D6* 10 gene polymorphism and clinicopathological features and prognosis was assessed by Chi-square test and Cox proportional model.Serum TAM and 4-OH-TAM concentration were determined by liquid chromatography-trap mass spectrometry (LC-MS).Results Of 200 breast cancer patients,CYP2D6 * 10/* 10 homozygotes were found in 94 cases (47%),CYP2D6 wt/wt wild type in 48 cases (24%) and CYP2D6 wt/* 10 heterozygote in 58 cases (29%).The concentration of serum 4-OH-TAM in patients with CYP2D6 wt/wt wild type and wt/* 10 heterozygote was significantly higher than that in patients with CYP2D6 * 10/ * 10 homozygote (F =4.31,P =0.01).CYP2D6 gene polymorphism was not related to clinicopathological features (P > 0.05).Logrank test showed that patient's disease-free survival with mutational CYP2D6 (mean,47.2 months) was significantly shorter than that in patients without mutational CYP2D6 (mean,51.2 months) (P =0.018).Cox proportional model showed that CYP2D6 genotype was related to disease-free survival (HR =2.755,95%CI:1.230 ~6.173,P =0.014).Conclusions CYP2D6 *10/* 10 genotype is related to curative effect of TAM on breast cancer patients,detection of CYP2D6 * 10/* 10 genotype helps improve the choice of TAM therapy.
2.Progress and application of phosphoproteomics in the proteomics of liver pathological and physiological state.

The phosphorylation is one of most common protein post-translational modifications. The protein phosphorylation plays important roles in the life through the reversible process of phosphorylation and dephosphorylation by kinases and phosphatases. Systematical analysis of the phosphorylation state of proteins would greatly help to reveal the mystery of the life. Recently, with the development of mass spectrometer, bioinformatics sortwares and enrichment methods of phosphopeptides, phosphorylation stduy of orgnism proteins by mass spectrometer has become mature gradually. Liver is one of the most important metabolic and immune organs. In-depth study of protein phosphorylation in liver is of great importance to reveal its function. And booming phosphoproteomics has been applied into the study of liver, which has deepened the knowledge of molecular mechnism of its physiology and pathology states. Here, we review the recent progress on the research and development of phosphoproteomics and their application in liver proteomics study.

3.Repairment of iatrogenic bile duct injury

Objective To evaluate different surgical repairments for iatrogenic bile duct injury Methods Clinical data of 145 patients with iatrogenic bile duct injury surgically treated in our department from Mar 1990 to Sep 2002 were retrospectively analyzed Results Bile duct end to end anastomosis、 or bile duct repair using the patient′s own tissues and medical glue, or Roux en Y bile duct jejunostomy were used in these 145 cases The average follow up time was 3 6 years (3 months to 11 years) in 130 cases The eutherapeutic rate was 94 6% Conclusion With good surgical expertise iatrogenic bile duct injury could be repaired by a variety of methods with a satisfactory results
4.Prophylactic treatment of ganciclovir to prevent and cure cytomegalovirus infection after renal transplantation: a systematic review

Objective To assess the efficacy of ganciclovir to prevent and cure cytomegalovirus (CMV) infection after renal transplantation. Methods We searched PubMed, EMBASE, Cochrane Library, SCI, China Academic Journals Full-text Databases, Chinese Biomedical Literature Database, Chinese Scientific Journals Databases and Chinese Medical Association Journals to collect randomized controlled trials of ganciclovir to prevent and cure CMV infection after renal transplantation (up to June, 2009). Two reviewers extracted data independently using a designed extraction form. The quality of included trials was evaluated according to the Cochrane Handbook. RevMan 5.0 software was used for data analysis. Results Twelve randomized controlled trials were included. The results of meta-analysis showed that: ①Compared with no receive antiviral agents, ganciclovir couldn't lower CMV infection rate and disease rate in 3 months and 6 months after renal transplantation, but could lower CMV disease rate in 12 months. The delay between transplantation and CMV infection was significantly longer. ②Either valaciclovir or ganciclovir could lower CMV infection rate and disease rate after renal transplantation, without statistical difference. ③Compared with acyclovir, ganciclovir could lower CMV disease rate in 6 months after renal transplantation. ④Compared with CMV-IgG and valganciclovir, ganciclovir didn't have statistical difference in decreasing CMV disease rate (P=0.93;P=0.14). Conclusions Longer prophylaxis by ganciclovior may prevent CMV infection after renal transplantation. Its curative effect is similar to valaciclovir, CMV-IgG and valganciclovir, but better than acyclovir.
5.Expression and significance of ?-catenin and cox-2 in bulge-originated cells from rat hair follicles

Objective To study the expression of ?-catenin and cox-2 in the bulge cells of hair follicle and investigate the relationship of their expression on cell proliferation. Methods The hair follicle was prepared from the resected cheek skin of 20 Wistar rats aged 7 days. The bulge cells were resected from the intact hair follicle and cultured in vitro. Immunocytochemical technique (ICC) was applied to detect ?-catenin and cox-2 expression in bulge cells at culture day 3, 5, 8, 13. Results ?-catenin and cox-2 strongly expressed in bulge cells and the expression correlated with the culture days. ?-catenin appeared in both plasma and nucleus, while cox-2 only in nucleus. Conclusion ?-catenin and cox-2 were correlated with bulge cells proliferation, and cox-2 might be the target gene of ?-catenin signaling pathway in nucleus.
6.THE NUTRITIONAL EVALUATION AND CLINICAL TRIALS OF A TOTAL ENTERAL NUTRIENT PREPARATION

The composition, nutritional evaluation and clinical trials of a nutritionally complete enteral preparation were presented. It was composed of glucose oligosaccharides, emulsified fat, solubilized proteins, twelve vitamins, certain minerals and trace elements. Rats were forced to negative nitrogen balance by means of protein-free diet. Body weight and serum total protein declined accordingly. When the feed was changed to this preparation in one group, and a routine one with equal nitrogen content in another, nitrogen balance was turned to positive, body weight increased, serum total protein returned to normal in both groups. The increments of nitrogen balance and body weight were, however, higher significantly in the group fed with the preparation than that with routine diet. The biological value, net protein utilization and protein efficiency ratio of the solubilized proteins in the preparation were higher than casein in the routine feed. Twenty surgical patients having the preparation as the sole source of nutrients could take around 8.37MJ(2000 kcal)a day. Nitrogen balance turned from negative to positive in an average time of 3.6 days. Body weight, serum total protein, serum albumin were all increased. Fourty patients took this as dietary supplement and the similar results were observed too. Clinical effects were excellent in all cases, especially for healing of surgical wounds and fistula. Incidence of diarrhea was 3.3%.
7.PRELIMINARY OBSERVATION ON THE GROWTH CHARACTERISTIC OF BULGE-ORIGINATED CELLS FROM HUMAN HAIR FOLLICLES

Objective To observe the growth characteristic of bulge-originated cells from human hair follicles in vitro. Methods The bulges which were isolated from human hair follicles by dispase and microdissection were cultured. The morphological and biological characteristic of the cultured bulge cells were observed by light microscopy and immunocytochemistry (ICC). Results Proliferated cells could be observed in the second day after being inoculated. The number of these cells, with greater proliferation potential, reached the peak at the sixth day and maintain several days. In addition, the mitotic figures appeared and the cells behave the similar morphologic features, meanwhile the cells strongly expressed K19 and showed a descensive tendency in long-time growth.Conclusion The cultured bulge cells kept the primitive characteristic, which suggested that the putative follicle stem cells resided in the bulge area.;
8.The expression and significance of β-catenin and p28GANK in residual hepatocellular carcinoma cells after transcatheter arterial chemoembolization

Objective To investigate the expression and significance of β-catenin and p28GANK in residual hepatocellular carcinoma (HCC)cells after transcatheter arterial chemoembolization (TACE).Methods We collected forty-five cases of surgical specimens of hepatocellular carcinoma after TACE ( TACE group ) and thirty cases of surgery without any treatment (pure surgery group).The expression of β-catenin and p28GANK were detected by using immunohistochemical SP method and compared between the two groups .Results The positive expression of β-catenin and p28GANK in TACE group were 77.78%and 75.56%respectively,which were significantly higher than those in pure surgery group (46.67%and 53.33%respectively,P<0.05).In the residual hepatocellular carcinoma cells of TACE group ,the positive expression of β-catenin showed correlation with the positive expression of p28GANK(Φ=0.318,P =0.033).The high expression of β-catenin and p28GANK were closely related to portal vein thrombosis and distant metastasis (P<0.05).Conclusion The ex-pression of β-catenin and p28GANK in the residual hepatocellular carcinoma cells were increased significantly after TACE.The high expression of β-catenin and p28GANK were closely related to portal vein thrombosis and distant metastasis.The high expression of β-catenin and p28GANK may be one of the reasons of hepatocellular carcinoma invasiveness and metastasis .
9.Construction and expression of recombinant plasmid pEGFPC1uPAR in Pam 212 cells

Objective To construct the eukaryotic expression plasmid of pEGFPC1uPAR gene and explore the effect on the proliferation and invasion ability of Pam 212 cells. Methods The human uPAR cDNA was cloned by PCR, and inserted into the eukaryotic expression plasmid pEGFPC1. After identification of sequencing, the reconstructive plasmid was transformed transiently into Pam 212 cells, then the cell growth and the invasion ability were evaluated. Results The reconstructive plasmid of pEGFPC1uPAR was validated by sequencing. The reconstructive plasmid can promote the growth of Pam 212 cells and enhance the invasion ability. Conclusion The pEGFPC1uPAR plasmid was constructed successfully and uPAR was confirmed to promote the growth and the invasion ability of Pam 212 cells, which lay the foundation for further studies of uPAR in vivo.
10.Determination of Organic Solvent Residues in Testosterone Cypionate

Objective To establish a headspace GC method for the determination of residual organic solvents in testosterone cypionate. Methods The residual organic solvents were determined by GC with DB-WAX capillary colum(30 m× 0. 32 mm,0. 25 μm)and FID detector at 230℃,using high purity nitrogen as the carrier gas by headspace sampling. The flow rate was 1. 5 mL·min-1 ,the split ratio was 20:1,the temperature of injection port was 220℃,the headspace heating temperature was 70 ℃,and the headspace balance time was 40 mins. The content of residues was calculated by using n-propanol as the internal standard. Results Four residual solvents were completely separated. There were good linearity for m-ethanol,ethanol,benzene and pyridinein in the ranges of 4. 17-2. 50×103 μg·g-1(R=0. 999 9),4. 18-2. 51×103 μg·g-1(r=0. 999 6),0. 84-172 μg·g-1 (r=0. 998 1),and 2. 95-1. 77×103μg·g-1(r=0. 999 9),respectively. The detection limit was 2.08,1. 23,0.28,and 0. 87 μg·g-1,respectively. The average recovery of methanol,ethanol,benzene and pyridinein was 102. 2%(RSD=4. 0%),99. 6%(RSD=1. 9%),112. 6(RSD=5. 6%),and 98. 9%(RSD=1. 6%),respectively. Conclusion This method is reliable,sensitive, accurate and can be used for the determination of residual organic solvents in testosterone cypionate.
