Objective To investigate the expression of lentivirus-mediated short hairpin RNA (shRNA) targeting collagen typeⅠ (Col Ⅰ) in rat mesangial cells.MethodsThe Col Ⅰ shRNA lentivirus vector was constructed with three-plasmids packaging approach which encoding human/rat homology Col Ⅰ gene.Rat mesangial cells were infected by infection enhancer (control group),the empty lentiviral vector (pSC-GFP group),and lentiviral expression vector (pSC-GFP/ColⅠ group),respectively.Infection efficiency in the three groups was detected with flow cytometry,and the expression of Col Ⅰ mRNA and protein in the three groups was respectively detected by RT-PCR and Western blotting.ResultsRat mesangial cells were efficiently infected in pSC-GFP group and pSC-GFP/Col Ⅰ group.The infection efficiency was 72.42% in pSC-GFP group and 75.42% in pSC-GFP/Col Ⅰ group.RT-PCR showed that the expression of Col Ⅰ mRNA in pSC-GFP/Col Ⅰ group was significantly lower than that in control group and pSC-GFP group (P

Abstract?AIM:To investigate the clinical effect of orthokeratology for 400 juvenile with myopia astigmatism and its effects on corneal endothelial cells.?METHODS:Four hundred patients(800 eyes), of whom the average age was 11.5 ±2.3 years old, 239 male, 161 female, were divided into two groups: orthokeratology group and spectacles group. Parameters including efficacy data ( uncorrected visual acuity, corneal curvature, axial length and diopter ) and corneal endothelial cell data ( count of endothelial cell, endothelial cell density, fluorescein staining and central corneal thickness) were observed at 1d, 1, 6, 12 and 24mo after wearing.? RESULTS: The visual acuity of spectacles group recovered to normal after wearing, that of orthokeratology group recovered to normal at 1mo after wearing.At 2a after wearing, the corneal curvature, diopter of orthokeratology group decreased significantly (40.09 ±0.31D, 0.23 ±0.06D respectively); while those of spectacles group increased, the differences between the two groups were significant (P<0.05).The axial length of the two groups increased slightly at 1mo after wearing ( P>0.05 ) compared to those before wearing. At 2a after wearing, the axial length of the two groups were 23.96 ± 0.38mm, 26.49±0.88mm respectively (P<0.05).At 2a after wearing, central corneal thickness was 527.33 ± 27.69mm, 526.98±26.89μm(P>0.05).The count of endothelial cell and endothelial cell density both decreased after wearing without significant differences (P>0.05).?CONCLUSION: Orthokeratology has less effect on the corneal endothelial cells, no obvious adverse reactions and can control the prognosis of myopia.

The MPN method was used to enumerate ammonia-oxidizing bacteria (AOB) in water and sediments of several shallow lakes. The suitable incubation time, medium types and substrate (ammonium sulphate) concentrations were studied. The results showed that, MPN values increased with the incubation time, reaching a stable maximum at some time stages, which was 40 days in all the samples for MSF medium. Among the three media used (XZ-AOB、MSF、SW), MSF give the highest MPN value. In addition, am- monium sulphate concentration in medium was an important factor affecting MPN estimation of AOB. Compared to AOB in lake sediments, AOB in lake water was more sensitive to ammonium sulphate concentration.

Objective To investigate the role of NF-?B signal pathway in the stress of a rat acute colitis model. Methods Acute colitis were induced by 6% intracolonic acetic acid lavation in rats; the spinal I-?B levels were measured by Western blot at L5-S1 and C2-4 levels; the spinal COX-2 mRNA levels were measured by RT-PCR at the same segments; the expressions of spinal COX-2 were also measured by immunohistochemistry. Results There were obvious regional inflammatory responses after intracolonic acetic acid lavation in rats; the extent of inflammation was gradually aggravated with time.The expressions of spinal I-?B at L5-S1 level significantly increased after 0. 5 h in acute colitis group. A questionnaire survey was conducted in 114 medical students of 4th year in eight-year program before and after community clerkship of general practice.The survey showed that there were significant differences in the awareness of the function and features of community health service,the doctorpatient relationship and the difference with large hospitals before and after community clerkship (P ＜ 0.05).The willingness of students to work in the community health service was low (P ＞ 0.05).85.1％ students (97/114) thought that the community health service center had a harmonious doctor-patient relationship,65.0％ (74/114) thought that full communication between general practitioners and patients was an important factor to build a harmonious doctor-patient relationship,66.6％ (76/114) felt satisfied,and 72.8％ (83/114) gained a lot in the clerkship.The results indicate that the community clerkship of general practice is conducive for students to understand theory,knowledge of general practice.

OBJECTIVETo explore the effects of stromal interaction molecule 1 (STIM1) on the expression of apoptosis-related proteins in prostate cancer PC-3 cells.

METHODSWe transfected the lentivirus vector STIM1-pGCSIL-GFP carrying STIM shRNA into human hormone-independent prostate cancer PC-3 cells, and 3 days later observed the transfection efficiency by fluorescence microscopy. At 7 days after transfection, we determined the expression of STIM1 in the PC-3 cells by RT-PCR and Western blot and those of apoptosis-related proteins Bcl-2, Bax, survivin and activated Caspase-3 by Western blot.

RESULTSAt 3 days, inverted microscopy revealed a transfection efficiency of > 80%. At 7 days, the STIM1 expression was significantly inhibited at both mRNA and protein levels. The Bcl-2/Bax rate was remarkably decreased as compared with that of the control group (0. 31 vs 1.24 ) , and the survivin expression was markedly reduced, 0. 14 times that of the relative expression in the control. However, the Caspase-3 cleavage was significantly activated, 1.52 times that of the control (P <0.05).

CONCLUSIONSTIM1 can be regarded as an oncogene in prostate cancer PC-3 cells. Inhibition of its expression can induce PC-3 cell apoptosis by reducing the Bcl-2/Bax rate, decreasing the survivin expression, and activating the Caspase-3 pathway.

ABSTRACT:To explore the feasibility of DNA barcoding in the identification of sandflies ,the mitochondrial cytochrome c oxidase subunit I (COI) genes of 9 dominant species in China from three genera in the Phlebotominae were studied .The in‐traspecies and interspecies genetic distances were calculated using the Kimura‐2‐parameter model ,and the phylogenetic trees were reconstructed by neighbor‐joining (NJ) method .Results showed that all of sandflies species were successfully distinguish‐ed by the phylogenetic trees .The average intraspecific genetic distance (0 .8% ) was much less than the average interspecific ge‐netic distance (11 .2% ) .The study based on COI sequences indicates that the sandflies from China could be well distinguished with the commonly used COI barcodes ,which are potentially well utilized in sandflies species identification .

Objective To investigate the surgical treatment for patients with isolated recurrence and metastasis of colon cancer after primary curative-intent resection. Methods In this stuay,58 cases suffering from postoperative recurrence of colon cancer between the year of 1996-2005 were analyzed retrospectively.Data were analyzed using statistical package for the social sciences(Release 11.5,SPSS,Inc).x2 analysis was used to determine the related factors,survival analysis was estimated using the Kaplan-Meier method and compared using tlle log-rank test. Results In this reoperative group radical resection was achieved in 81%(47/58)cases,and palliative operation in 11 cases,with median survival time of 48 months and 12 months respectively(x2=12.30,P=0.0005). Conclusion Postoperative isolated recurrence and metastasis of colon cancer usually allows radical resection in most cases with favorable long-time survival.
