1.Application of self-made quality control materials on chemiluminescence analyzer

Objective To investigate the feasibility of self-made quality control materials on chemiluminescence analyzer .Meth-ods Residual serum in blood bags were collected and served as matrixes .Complex quality control materials of tumor markers [car-cinoembryonic antigen ,alpha-fetoprotein ,carbohydrate antigen 125(CA125) ,CA199 ,CA153 ,human chorionic gonadotrophin and total prostate specific antigen ] and hormonal markers (luteinizing hormone ,follicle-stimulating hormone ,estradiol ,testosterone , progesterone and prolactin) were prepared .The monthly average value ,coefficient of variation(CV)% value ,cumulative average value and the number of out-of-control of each marker during the 12 months were calculated .Results All self-made quality control serum which stored and packaged separately showed appearance of clear and transparent after freezing and thawing ,without turbidi-ty and sedimentation .Difference of monthly average value and cumulative average value of each marker had no statistical significance (P>0 .05) .Compared with monthly average value and cumulative average value of each marker ,no obvious drift and trends were found .Monthly indoor imprecision of each hormonal marker were controlled within the allowable range .36 times of out-of-control alarm were monitored during a year ,among them ,25 for system error and 11 for random error .Conclusion Self-made quality con-trol materials has good stability and may find the instability factors of detection system timely to guarantee the reliability of test re-sults of chemiluminescence analyzer .
2.Small interfering RNA targeting connective tissue growth factor prevents rats liver fibrosis

Objective To explore whether portal vein injection of small interfering RNA(siRNA) targeting connective tissue growth factor(CTGF) could inhibit CTGF expression in rats liver in vivo and prevent rats hepatic fibrosis. Methods Twenty four male rats were equaly divided into four groups. Rats received subcutaneous injection of 40% CCl4 (3 ml/kg) together with portal vein injection of saline every three days for 6 consecutive weeks were served as model group; CCl4 together with portal vein delivery of siRNA (0.1 mg/kg) as preventive group; CCl4 for 2 weeks followed by CCl4 and CTGF siRNA for more than 4 weeks as curative group, and only intraportal injection of saline as control group. Portal vein pressure in all rats at 3 days after the last CCl4 injection were measured, and blood and hepatic tissue from rats were harvested. Serum concentration of transaminases, hyaluronic acid(HA) and procollagen type Ⅲ (PⅢNP) were measured. Expression of CTGF mRNA and protein in rats liver was evaluated by RT-PCR and Western blot, respectively. Inflammation and fibrosis in rats liver was analyzed by H-E and Sirius red collagen staining. The fibrotic area was measured quantitatively by a computer-aided manipulator. Results Compared with model group, the expression of CTGF mRNA and protein in liver in both preventive and curative groups were markedly down-regulated. Inflammation, necrosis and fibrosis in hepatic tissue was significantly attenuated. In addition, the serum concentration of transaminases, HA and PⅢNP were greatly reduced. 3.Relationship Between p53 Gene Mutation and Apoptosis and Cell Ploidy of Ovarian Carcinoma

Objective To study the relationship among p53 gene mutation, apoptosis and cellploidy as well as to explore the role of mutated p53 gene in tumorigenesis. MethodsA total of 85 speci-mens (20 normal ovarian specimens, 20 ovarian benign tumors, 20 non - metastatic carcinomas and 25metastatic carcinomas) were chosen. p53 gene mutations were detected by PCR- SSCP (single strand con-formation polymorphism). Cell ploidy, apoptosis role of carcinoma cells and cell distribution in each cellstage were detected by flow cytometry. Resultsp53 gene mutation and non- mutation of ovarian carci-noma was 60% and 40% respectively ( P >0.05). The rate of apoptosis of carcinoma cells with mutationof p53 gene (20.25%) was much lower than that without p53 mutation (41.68%, P <0.05). Aneuploidcarcinoma cells had been found in 17/23 carcinoma specimens with p53 gene mutation and 8/22 carcinomaspecimens without p53 mutation ( P < 0.05). Aneuploid carcinoma cells metastasize more easily thandiploid carcinoma cells ( P < 0.05). The rate of apoptosis was not different between aneuploid and diploidpatients. ConclusionMutation of p53 gene decreases the apoptosis rate of ovarian carcinoma and maypromote tumorigenesis. Cell ploidy had a linkage with p53 gene mutation, but not with cell apoptosis.

6.Efficacy of olanzapine combined with fluoxetine in treatment of depression

Objective To explore the efficacy of low dosage of olanzapine combined with fluoxetine in the treatment of depression.Methods A 8-week study was conducted in 130 patients met the diagnostic criteria for de- pression.Subjects were randomly assigned to two groups:fluoxetine(20mg/d)alone and olanzapine(2.5~5 mg/d) plus fluoxetine(20mg/d).They were evaluated with Hamilton depression scale(HAMD).Hamilton anxiety scale (HAMA)at baseline,the 1 week,2 weeks,4 weeks and 8 weeks subsequently.Results(1)There were significant differences in the total scores and reduction rates of HAMD between two groups in every interview.(2)The combi- nation group had greater reduction in depressive and anxiety symptoms than that in fluoxetine group.(3)The re- sponse rate in combination group was higher than that of fluoxetine group in 1 week,2 weeks and 4 weeks.There were no significant differences in response and remission rate between combination group and fluoxetine group.Con- clusion The combination of olanzapine with fluoxetine demonstrated a rapid,effective antidepressant action.

8.Survey of CT radiation dose to 1200 cancer patients

Objective To survey the CT radiation dose to cancer patients,and to compare it with diagnostic reference level (DRL) provided by newly issued national standard.Methods Computed tomography dose index weighted (CTDIw) of 5 CT equipments was measured by the standard phantom with typical scanning protocol and radiation dose parameters including average computed tomography dose index volume (CTDIvol),dose length product (DLP),75% DLP with 900 adult and 300 pediatric cancer patients were collected.According to age stage,effective dose was estimated.Results The measured CTDIw for head scanning was on the same level with DRL,while lumbar vertebra and abdominal scanning were lower than DRL.To adults,DLP of cerebrovascular enhanced scanning and effective dose of abdominal enhanced scanning were the highest.To pediatric patients,there was no significant difference in DLP among different ages.Conclusion CTDI only reflects the dose contribution from a slice,but do not reflect the cumulative dose.For the patients need multiphase scanning,because they may be exposed to high dose,national standard should provide DLP value as the diagnostic reference level.Using individual scanning protocol is an effective method to reduce radiation dose of pediatric patient.
9.Experimental Study-Methycobal Promotes Peripheral Nerve Regeneration

Objective:To investigate the effect of methycobal on peripheral nerve regeneration.Methods:Thirty Wistar rats were divided into normal group(N),model group(M)and methycobal group(B).Each group contained 10 rats.The model of rat with sciatic nerve half injury was made by clamp.Rats were fed with methycobal in B group,and same dose of normal saline was fed in N and M groups.The nerve tissue(silver nitrate stain)and gastrocnemius tissue(HE stain)were obtained to observe the morphologic index,functional index(sciatic nerve function index,differential value of hind legs tensile force,gastrocnemius wet weight index)after 14 days.Results:There were nerve fiber sparseness,disposition disorder,and decrease of gastrocnemius cross section area,sciatic nerve function index,and differential value of hind legs tensile force,gastrocnemius wet weight index diminutu in M group compared with those of N group(P < 0.01).There were more nerve fiber,normal gastrocnemius cross section area,and higher sciatic nerve function index and gastrocnemius wet weight index in B group compared with those of M group(P< 0.01),whereas which were also different compared with those of N group(P< 0.01).Conclusion:Methycobal can promote the axon growth,postpone sarcous anaiosis of denervation,and enhance functional recovery of peripheral nerve injury.
10.Progress in urinary protein tumor markers

The development of proteomic technique has sparked new focus on novel protein markers for many diseases including cancer. Urine is one of the most useful biofluids for routine testing to provide an ideal resource for the discovery of novel biomarkers, with the advantage over tissue biopsy samples due to the ease and the less invasive nature of urinary collection. This review summarizes urinary protein tumor markers found in prostate cancer, bladder cancer, lung cancer and breast cancer that have shown potential for urogenital and non-urogenital cancer, so as to provide reference on high sensitive and high specific urinary protein tumor markers for early prevention, early diagnosis and treatment of cancer.
