1.Apoptosis and Bcl-2 protein expression in rat kidney tissue after severe burn with delayed fluid resuscitation in areas of different altitude

Objective To observe the changes in Bcl-2 protein expression in rat kidney tissue after severe burn with delayed fluid resuscitation in areas of different altitude,and explore the relationship between its expression and cell apoptosis.Methods The experiments were performed at two altitudes(1 517m and 3 848m).A 30% TBSA Ⅲ? scald model was reproduced with 240 male Wistar rats(120 rats for each altitude).Rats were randomly assigned into delayed fluid resuscitation group(DFR,n=50),immediate fluid resuscitation group(IFR,n=60) and normal control group(NC,n=10).Renal tissue samples were harvested at 1,6,12,24,72 and 168 hours after scald,respectively.The cell apoptosis was detected by tissue chips technic and terminal uridin nick end labeling(TUNEL).The expression of Bcl-2 was detected by immunohistochemistry and image analysis.Results In higher altitude,cellular edema,granular degeneration,necrosis and disintegration of renal tubular epithelial cells;dilation and engorgement of renal glomerular capillaries,degeneration and necrosis of endothelial cells,and congestion,edema and inflammatory cell infiltration in renal interstitium were found.The pathological changes were more serious in DFR group than that in IFR group,and they were worse in the rats at 3 848m altitude than that in those rats at 1 517m altitude.The levels of cell apoptosis and Bcl-2 protein expression were higher in DFR group than that in IFR group,and in the rats at 3 848m altitude than that in those rats at 1 517m altitude(P
2.Identification of Chaetomium sp. CQ31 and Optimization of Xylanase Production in Solid State Fermentation

A fungus CQ31 isolated from soil samples was identified as Chaetomium sp.. This strain produced effectively xylanases utilizing several liguocellulosic materials in the solid-state fermentation (SSF), and corn straw was the best carbon source. The results of single-factor-experiment showed that the corn straw as the carbon source, tryptone as the nitrogen source, initial moisture content of 80% and initial pH 9.0 were the optimal conditions for xylanase production. Under the optimized conditions, it produced xylanase which was 4897 U/g dry substrate while mannanase was 803 U/g dry substrate after 7 days of cultivation. Therefore, xylanase and mannanase production by Chaetomium sp. CQ31 in SSF possess potential for commercial applications.
3.Effect of Serum Containing Rhizoma Chuanxiong on Proliferation and Differentiation of Mouse Embryonic Stem Cells

Objective To observe the influence of serum containing Rhizoma Chuanxiong on the proliferation and differentiation of mouse embryos stem cells(ESCs).Methods Mouse ESCs were co-cultured with serum containing Rhizoma Chuanxiong.The proliferation of ESCs was detected by CCK-8 method.The expression level of specific gene beta-myosin heavy chain (β-MHC) in cardiac myoblasts was detected by reverse transcription-polymerase chain reaction (RT-PCR).Results Compared with the blank rat serum group and the blank fetal bovine serum group,the differences of activities of ESCs in serum containing Rhizoma Chuanxiong group were in significant(P >0.05) and the expression level of specific gene β-MHC in cardiac myoblasts was increased (P < 0.05).Conclusion Serum containing Rhizoma Chuanxiong can promote the differentiation of ESCs into cardiac myoblasts.

5.Expression and signiifcance of thrombonspondin-1 in oxygen-induced retinopathy in mice

Objective:To examine the expression and function of thrombonspondin-1 (TSP-1) in oxygen-induced retinopathy in new-born mice, and to investigate its role in retinal neovascularization. Methods:A total of 40 C57BL/6J newborn mice were divided equally into a model group (n=20)andanormalcontrolgroup(n=20).Miceinthemodelgroupwereexposedto75%oxygentoestablish the oxygen-induced retinopathy (OIR) model. On the 7th, 9th, and 11th day after the birth of mice, 5 mice were randomly selected each time from the 2 groups to examine the expression of TSP-1 mRNA with reverse transcription polymerase chain reaction (RT-PCR). After that, 5 mice were selected on the 11th day to observe the retinal neovascularization by fluorescein angiography retinal flatmount. Results:On the 11th day, fluorescein angiography retinal flatmount showed that the retinal blood vessels presented mean network distribution in the normal control group, while in the model group, a lot of dilatated areas in the retinal main vessels surrounded the optic disc. Meanwhile lots of new blood vessels were found surrounding the optic disc with irregular distribution but well distributed peripherial retinal small vessels, which was typical of early stage OIR. There was no signiifcant difference in the retinal TSP-1 mRNA level between the model group and the normal control group in the postnatal 7-day mice (P>0.05). Compared with the normal control group, the expression of TSP-1 mRNA in the model group was signiifcantly lower in postnatal 9-day and 11-day mice (P<0.05, P<0.01) , and the expression of TSP-1 mRNA in postnatal 9-day mice was lower than that in the postnatal 11-day mice(P<0.05).
Conclusion:In the early stage of OIR model (retinal vascular growth and development stage), the expression of TSP-1 mRNA in the retinal tissue is gradually decreased, implying that TSP-1 (as a negative regulatory factor) may be involved in the formation of retinal neovascularization in the early stage.
6.Effect of cytotoxicity T lymphocyte on acute rejection of xenogenic liver transplantation

Objective To study the mechanism of cytotoxic T lympocytes in the acute rejection of xenogenic liver transplantation(XLT). Methods Hamster to rat orthotopic liver transplantation model was performed with three cuff method. In XLT,lymphocytes including CD8 and CD4 subsets were observed by histology and immunohistochemistry; the expression of perforin and Fas- L was observed by immunohistochemistry; and apoptotic cells of XLT were observed in situ end-labelling of fragmented DNA. Results In the XLT, T cell infiltration firstly ocurred on the 2nd day posttansplantation and located at the portal triads; the infiltrating lymphocytes proliferated increased with time and reached the peak on the 5th to 6th days.Perforin and Fas-ligand were expressed on the 4th day after XLT, and peaked on the 5th to 6th day,and the expression of perforing was still higher than the expression of Fas-ligand. Conclusions T lymphocyte participates in the acute rejection of XLT through the expression of perforin and Fas-ligand, which leads to apoptosis or necrosis of hepatocytes.

8.Clinical features, treatment and prognosis analysis of the asymptomatic patients with retinoblastoma

Objective:To analyze the clinical features, treatment and prognosis of asymptomatic patients with retinoblastoma.Methods:A retrospective series of case study. Eight asymptomatic patients (11 eyes) with the diagnosis of retinoblastoma by screening enrolled in Department of Ophthalmology of The Eye-ENT Hospital of Fudan University from January 2006 to March 2019 were included. There were 6 males and 3 females ranging from 2 days to 20 months, with a median age of 6 months. Five patients were unilateral retinoblastoma while 3 patients were bilateral. Based on the International Classification of Intraocular Retinoblastoma, 4 eyes were stage A, 3 eyes were stage B and 4 eyes were stage C. One patient had family history. Four patients were evaluated the Rb1 mutation. Routine ophthalmic examinations and ultra-wide field fundus imaging were performed on the 16 parents and 3 siblings of the 8 patients. Systemic intravenous chemotherapy was performed using the Carboplatin, Vincristine, Etoposide protocol, intra-arterial chemotherapy using Carboplatin and Melphalan, and local treatment involved cryotherapy and transpupillary thermotherapy. The mean follow-up time is 47.25 months.Results:None of the 8 children had any ocular symptoms. Six patients received intravenous chemotherapy (5-6 times), 1 patient received intra-arterial chemotherapy (3 times), and 1 patient just received local treatment. Among the 11 eyes, 9 eyes were treated with local cryotherapy and 8 eyes were treated with transpupillary thermotherapy. During the follow-up period, 2 patients had new tumor, and the average time was 6.3 months after the last chemotherapy. At the last follow-up, the tumor disappeared in 11 eyes, remained stable in 11 eyes. The eye protection rate was 100% (8/8) for patients without eyeball excision. The best corrected visual acuity was 0.1 for 3 eyes and 1.0 for 5 eyes. Three eyes were not found. One heterozygous mutation of Rb1 gene [1c.35_69del (p.T12fs)] was identified in 1 patient, and the other 3 patients were not detected. One had bilateral bulbar tuberculosis of the 16 parents, 1 had bilateral RB of the 3 siblings. They were the mother and brother of a child with bilateral RB.Conclusions:Fundus screening is helpful for the detection of early RB. The eye protection rate is high and the long-term vision prognosis is good after systemic or topical chemical drugs (IVC, IAC) and ocular topical treatment (cryopreservation and transpupillary thermotherapy).
9. Effect of miR-1269a expression on the radiotherapy sensitivity of H460 stem cell like cells

Objective To investigate the effect of miR-1269a expression on the radiotherapy sensitivity of H460 stem cell-like cells of non-small cell lung cancer. Methods Non-small cell lung cancer H460 cell line was selected for serum-free suspension culture, and CD133 and CD44 positive H460 stem cells were screened by flow cytometry. qRT-PCR was performed to detect the expression levels of miR-30a, miR-148a, miR-148b, miR-152, miR-411, miR-497, miR-598, miR-424, miR-761, miR-1269a, miR-1280 and CD44, CD133 mRNA in H460 stem cell-like cells. The effects of miR-1269a expression on the radiotherapy sensitivity of H460 stem cell-like cells were analyzed using CCK-8 experiments, stem cell pelleting experiments, flow cytometry, and comet experiments. Results The expressions of CD133 and CD44 mRNA increased 1.30±0.03 times and 1.19±0.02 times, respectively, in H460 stem cell-like cells than in H460 cells (P<0.05). The expression level of miR-1269a was the highest in H460 stem cell-like cells, and was higher than that in H460 cells (P<0.05). After transfection of miR-1269a inhibitor and X-ray treatment, the growth inhibition rate of H460 stem cell-like cells increased significantly (72.11%±2.45%), the apoptosis rate reached to 17.70%±0.54%, and the fragmentation of DNA double-strand breaks in cells also increased significantly. After 2 Gy of X-ray radiation, the tail moment was 1.19±0.02 times of that before transfection, implying that transfection of miR-1269a inhibitor significantly increased the sensitivity of H460 stem cell-like cells to radiotherapy (P<0.05). After transfection of pcDNA3.1-SOX7, the effect of X-ray treatment on H460 stem cell-like cells was similar to that of miR-1269a inhibitor transfection. Simultaneous transfection of pcDNA3.1-SOX7 and miR-1269a mimics reversed the inhibitory effect of transfection of pcDNA3.1-SOX7 on the malignant biological behavior of H460 stem cell-like cells. Conclusion Inhibition of miR-1269a may enhance the radiotherapy sensitivity of non-small cell lung cancer, and the mechanism may be caused by up-regulation of SOX7 expression.
10.The expression and significance of Caspase-3/Bax in TCM differentiation of syndromes typing for ulcerative colitis

Objective To study the correlation between TCM differentiation of syndromes typing for ulcerative colitis (UC) and expressions of Bax and Caspase-3.Methods 50 patients with UC were divided into 2 groups according to TCM syndrome differentiation including dampness-heat internal accumulation syndrome group (25cases),deficiency of spleen and stomach syndrome group (25cases).Immunohistochemistry was used to study expression of Bax and Caspase-3 in colonic mucosas and compare with the normal group (25cases).Results The expressions of Bax were highest in dampness-heat internal accumulation syndrome group (4.56±1.58).The expressions of Bax of deficiency of spleen and stomach syndrome group were lower than dampness-heat internal accumulation syndrome group(3.28± 1.14)scores.There were significant differences between the three groups (P<0.05).However,There were no significant difference between dampness-heat internal accumulation syndrome group and deficiency of spleen and stomach syndrome group in terms of the Caspase-3expressions (P>0.05).In addition,Caspase-3 and Bax were positively related in each group(r=0.23and 0.21).Conclusion The high expressions of Bax,Caspase-3 in ulcerative colitis were closely related to TCM typing according to syndrome differentiation.Caspase-3 and Bax were adjustment in apoptosis.The detection of expression of Bax,Caspase-3 is helpful to the TCM syndrome differentiation and to the selection of treatment plan.At the same time,it can provide more important experiment basses for the pathogenesis of ulcerative colitis.
