1.Effect of MACC1 down-regulation on proliferation of gastric adenocarcinoma cell line

Objective To study the effects of MACC1 down-regulation on the growth of gastric adenocarcinoma cells. Methods siRNA (MACC1-siRNA1 and MACC1-siRNA2) that can transiently silenced MACC1 was designed, syn?thesized and transfected into MGC-803 cells by lipofectamine 2000. Non-specific siRNA was transfected to be used as nega?tive control. The efficiency of MACC1 depletion was determined by Real-time quantitative PCR. MTT, colony formation and flow cytometry assay were performed to examine cell proliferation. The expressions of MACC1, P21,CDK4, CCND1 and c-myc were determined by Western blot. Results Compared with cells in negative control group, transiently silencing MACC1 decreased the expression of MACC1 in MGC-803 cells shown by Real-time PCR. MACC1 downregulation drastical?ly changed the proliferation, colony formation and cell cycle of gastric adenocarcinoma cells in vitro ( P<0.05). The expres?sions of MACC1 , CDK4, CCND1 and c-myc proteins in cells of MACC1 silence group were much lower while P21 expres?sion level was much higher than those in negative control. Conclusion Down-regulation of MACC1 result in blocking cell cycle, inhibiting proliferation of MGC-803 cells. So it may serve as a promising target in the treatment of gastric cancer.
2.Comparative Analysis on Simultaneous Determination of α-pinene, β-pinene, Eucalyptol and Linalool in Baeckea Frutescens Preparations by Reference Substances Method and Reference Extract Method

Objective To determinate the contents of α-pinene, β-pinene, eucalyptol and linalool in Baeckea frutescens by reference substances method and reference extract method respectively; To explore the feasibility of replacing single component reference by control extracts in assay. Methods The GC system consisted of a quartz column DB-5 (60 m×0.25 mm×0.25 μm); The temperature programming rose from 80 ℃ (15 min) to 90 ℃ by 1 ℃/min, lasting 2 min, then 10 ℃/min to 110 ℃, then 25 ℃/min to 240 ℃, lasting 8 min in the end; The temperature of the entrance of capillary vessel column was 250 ℃, and the temperature of the detector was 250 ℃. Results α-pinene, β-pinene, eucalyptol and linalool were in a good linear relationship within each concentration scope (r≥0.999). The average recovery rates were in the range of 96.5%–102.2%. The results of t-test demonstrated that there is no significant difference between the two methods. Conclusion The reference extract method can be used as a quality evaluation pattern for Baeckea frutescens.
3.Effect of Folic Acid on the Homocysteine level and Flow-mediated Vasodilation and Cognitive Function in the Treatment of Elderly Patients with Cerebral Infarction

OBJECTIVE:To discuss the effect of folic acid on the homocysteine(Hcy)and flow-mediated vasodilation and cog-nitove function in the treatment of elderly patients with cerebral infarction. METHODS:The data of 198 elderly patients with cere-bral infarction was randomly divided into the observation group and the control group by different medication. Control group was treated with conventional treatment;based on the treatment of control group,observation group was orally treated with Folic acid tabelts 5 mg,once a day. The treatment course of 2 groups was 8 weeks. The Hcy level and flow-mediated vasodilation(FMD)be-fore and after treatment,mini-mental state examination (MMSE) score and incidence of adverse reactions in 2 groups were ob-served. RESULTS:After treatment,Hcy level in observation group was significantly lower than before and control group,FMD was significantly higher than before and control group(P<0.01). MMSE score in observation group was significantly higher than control group(P<0.01). There were no obvious adverse reactions in 2 groups during treatment. CONCLUSIONS:Based on the con-ventional treatment,folic acid can effectively reduce the Hcy level of elderly patients with cerebral infarction,improve the cogni-tive function and reduce the brain tissue damage,with good safety.
4.Determination of 100 Pesticide Residues in Moras alba by LC-MS/MS

OBJECTIVE:To establish a method for the determination of 100 pesticide residues in Morus alba. METHODS:LC-MS/MS was conducted on the column of ZORBAX Eclipse plus C18 with mobile phase of acetonitrile (containing 5% water, 0.1% formic acid,5 mmol/L ammonium formate)- water(containing 0.1% formic acid,5 mmol/L ammonium formate)(gradient elution)at a flow rate of 0.4 ml/min,column temperature was 40 ℃,injection volume was 5 μl;MS conditions:ionization source was electrospray ionization with positive ion mode,scanning mode was dynamic MRM,scanning time window was 2 min;atomiz-ing gas was nitrogen,atomizing gas pressure was 40 psi,drying gas flow rate was 5 L/min,capillary voltage was 4 000 V,and ion spray voltage was 500 V. RESULTS:The linear range of 100 pesticide residues was 2.4-150 ng/ml(r>0.990 0),recovery was 69.3%-128.2%,and the determination limit was 0.003-16 μg/kg. CONCLUSIONS:The method is simple,stable and reproduc-ible,and can be used for the determination of pesticide residues in M. alba.
5.Diagnosis and treatment of adult-onset Still′s disease

Adult-onset Still′s disease (AOSD) is a type of systemic inflammatory disease of unknown etiology, with diverse clinical manifestations, and there are some difficulties in its diagnosis and treatment. In recent years, it has been found that some new markers, such as heme oxygenase 1, calreticulin, inflammatory cytokines and advanced glycation end products, can be used for a comprehensive assessment of the activity and severity of AOSD. Moreover, new biological agents, such as tumor necrosis factor inhibitors, interleukin-1 (IL-1) inhibitors, IL-6 inhibitors and recombinant IL-18 binding proteins, bring new hope for the treatment of AOSD. This review mainly summarizes progress in the diagnosis and treatment of AOSD.
6.Eplerenone partly reverses thyroid hormone induced myocardial electrical remodeling

AIM:To investigate the effects of eplerenone (Epl) on thyroid hormone (T3) induced myocardial electrical remodeling .METHODS:The ventricles of 1~3 d neonatal rats were digested with 0.125%trypsin and 0.08%collagenase type 2.The cell suspension was replated for 90 min to reduce the proportion of non-myocardial cells.The isola-ted cardiomyocytes were randomly divided into control group , T3 group, Epl group and T3+Epl group.The cardiomyocytes were identified by immunofluorescence staining .The viability of the cardiomyocytes was measured by CCK-8 assay.The ex-pression of Kv1.5, Kv4.3, Cav1.2, connexin 40 (Cx40) and Cx43 at mRNA and protein levels was determined by immu-nofluorescence staining , real-time PCR and Western blot .RESULTS:The results of the cell immunofluorescence labeling conformed that the cultured cells were cardiomyocytes with more than 95%positive staining of sarcomeric α-actinin.Com-pared with control group , the mRNA and protein levels of Kv1.5, Kv4.3, Cav1.2 and Cx40 were increased, but the ex-pression of Cx43 was decreased in T3 group.The mRNA and protein levels of Kv1.5, Kv4.3, Cav1.2 and Cx40 were de-creased, but the expression of Cx43 was increased in Eplerenone group .Compared with T3 group, the mRNA and protein expression levels of Kv1.5, Kv4.3, Cav1.2 and Cx40 were decreased, but the expression of Cx43 was increased in T3+Epl group.CONCLUSION:Eplerenone partly reverses T3-induced myocardial electrical remodeling .

8.Influence of long-term exercise training on carotid intima-media thickness in patients with mild-moderate hypertension

Objective: To observe influence of long-term exercise training on carotid intima-media thickness (IMT) in patients with mild-moderate hypertension. Methods: A total of 92 patients with essential mild-moderate hypertension were randomly divided into routine treatment group (n=46) and exercise group (n=46, received exercise training based on routine treatment) according to number table. All patients were followed up for one year, and the blood pressure, carotid artery diameter and carotid IMT were measured, compared and analyzed in two groups before and after treatment. Results: There was no significant difference in systolic and diastolic blood pressure before treatment between two groups (P>0.05). Compared with baseline value, there were significant decrease in systolic blood pressure [(176.66±l1.78)mmHg vs. (130.89±13.01)mmHg], diastolic blood pressure [(101.43±6.41)mmHg vs. (81.71±8.45)mmHg], carotid artery diameter [(6.62±0.97)mm vs. (6.22±1.01)mm] and carotid IMT [(0.98±0.12)mm vs. (0.84±0.11)mm] in exercise group after one-year training (P<0.05 all), and they were all significantly lower than those of routine treatment group (P<0.05 all ). Conclusion: Long-term exercise training can effectively control blood pressure, decrease carotid artery diameter and carotid intima-media thickness.
9.The relationship between the extent and severity of angiographic coronary artery disease and impaired fasting glucose

Objective To evaluate the correlation between impaired fasting glucose and the extent and severity of angiographic coronary artery disease( CAD ) in patients who underwent coronary angiography.Methods A total of 630 consecutivc patients who underwent coronary angiography were selected in the stndy.The extent and severity of angiographic CAD were diagnosed based on the followings:whether or not with CAD diagnosis by angiography,the number of diseased vessels,the CAD Gensini cumulative index.According to fasting plasma glucose ( FPG),all patients were divided into three group,group 1 ( FPG <5.6 mmol/L),group 2 ( 5.6 mmol/L ≤ FPC < 6.1 mmol/L) and group 3 ( 6.1 mmol/L ≤ FPG < 7.0mmol/L).The CHD risk factors and the lesion degree of coronary arteries were compared among three groups.The relation of FPG and CAD were analyzed by univariate and multivariate analyses.Results The incidence of CH and,the CAD Gensini cumulative index and the incidence of mult-vessel lesions were significantly different among the three subgroups ( all P < 0.05 ).After adjusting other inffluencing factors,there were significantly positive correlation between the number of diseased vessels and the CAD Gensini cumulative index and FPG( P <0.05).The FPG had significant correlation with the diagnosis or not of CAD by angiography( OR =3.042,95% CI:2.589 -6.275,P <0.01).Conclusions In prediabetic period,the extent and severity of angiographic coronary artery disease have significant correlations with the impaired fasting glucose,and the extent and severity can increase along with the increasing FPG.
10.The efficacy and safety of tirofiban in the treatment of elderly patients with acute coronary syndrome during primary percutaneous coronary intervention

Objective To investigate the efficacy and safety of tirofiban in the treatment of aged patients with acute coronary syndrome (ACS) during primary percutaneous coronary intervention (PCI). Methods Two hundred and fifty-six patients with ACS who underwent primary PCI were randomly divided into two groups: tirofiban group (tirofiban + PCI treatment, n= 130) and control group (routine PCI treatment, n = 126). Tirofiban group was further divided into two subgroups according to the age: aged group(age ≥60 years, n= 68) and non-elderly group(age<60 years, n=62). At the end of PCI procedure, angiographic features such as thrombolysis in myocardial infarction (TIMI) flow grade, corrected TIMI frames count and TIMI myocardial perfusion grades (TMPG) were analyzed immediately. The incidence of major adverse cardiac events (MACE) was observed within 36 hours and 30 days after PCI, and the incidence rates of thrombocytopenia and bleeding were assessed. Results There was no significant difference between both the two study groups and the two subgroups in the basic clinical or angiographic characteristics before PCI (all P>0.05). There was no significant difference between two groups in TIMI 3 flow achievement rate in culprit vessels after PCI (93.6% vs. 91.3%, χ~2 = 1.02, P= 0.313). In tirofiban group, corrected TIMI frames count was significantly higher than that in control group (31.6±7.7 vs. 23.8±6.1, t = 2.49, P 0.026), and TMPG 3 achievement rate was also higher (83.1% vs. 67.5% ,χ~2=4.05, P=0.046). The incidence of MACE was significantly lower in tirofiban group than that in control group both within 36 hours and 30 days after procedure(6.9% vs. 19.0%, χ~2= 6.30, P= 0.013; 3.8% vs. 11.90%, χ~2= 5.82,P=0.018, respectively). No statistical difference was found in mild bleeding complications between the two groups (20.2% vs. 15.2%, χ~2 =3.65, P=0.065), but the incidence of mild bleeding was higher in aged group than that in the non-elderly group(25.0% vs. 14.5%,χ~2=4.98, P=0.026), and the incidences of serious bleeding and thrombocytopenia were similar between the two subgroups. Conclusions Intravenous tirofiban infusion is safe in aged ACS patients during primary PCI, and has favorable short-term outcomes compared with routine treatment, although there is a slight risk of mild bleeding.
