Objective To study the effects of basic fibroblast growth factor gene in enhancing collateral vessel formation of coronary artery in the ischemic myocardium of swine. Methods All swines were distributed into the operation control group(n=6) and treatment group. The treatment group was further divided into 2 groups according to the route of injection either through the right coronary artery or the left coronary(n=6 respectively). The animal models of AMI were prepared by ligating the left circumflex(LCX) coronary artery. Two weeks after the operation,2?000 ?g of pcDNA_3-bFGF eukaryotic expression plasmid was directly injected into the coronary artery by catheter. Two weeks after the gene injection, evaluation of collateral circulation formation was made by means of coronary angiography and immuohistological staining ect. Results (1) Through immuotistological staining, the vessels count in both treatment groups was more than that in the control group;(2) Selective coronary angiography at 4 weeks after the operation showed that the number of newly formed collateral vessels in the bFGF gene treatment group was more than that in the control group. On the other hand, more collateral vessels were found in the group through left coronary injection compared with the group through right coronary injection. Conclusion Intra-coronary artery injection of bFGF gene can improve collateral vascular formation in the ischemic myocardium of swine.

4.Changes of Ultrastructure of Muscle in Rats with Spastic Palsy after Injected with Botulinum Toxin
Journal of Applied Clinical Pediatrics 2006;0(14):-
Objective To observe the ultrastructural changes of end-plate and gastrocnemius muscle of rats with spastic palsy after injected with botulinum toxin type A in order to provide scientific base of histomorphology for clinical use of botulinum toxin type A.Methods Wistar rats′ pyramidal tracts in experiment group were injured by electric current 2.5 mA for 30 s,twice, but 36 normal control rats were only drilled hole in skull(control group).Seventy-two rats were successful in building spastic palsy model.Botulinum toxin type A (6 U/kg) were injected into right gastrocnemius muscles of 36 spastic palsy rats (group B),and physiological saline were injected into the same muscles in 36 spastic palsy rats as control(group A).Control group were not injected. The rats of group A,group B were sacrificed on 3,7,15,30,60 and 90 days after botulinum toxin therapy, and right gastrocnemius muscles was separated for histological analysis. Results Under the electron microscope, the structure of muscle fiber in group B had changed(Z line rupture, disorder and abolition, myofilament lysis, triad tract diso-rder,vacuolar degeneration) and these changes in group B occurred earlier and were severer than those in group A.There was compensatory hypertrophy of muscle fiber at later stages in group B.During the first 15 days after botulinum toxin type A injection, there were many synaptic vesicle without pre-synaptic membranes near terminal portion of nonmedullated nerve fibers.Following this, between 15 to 30 days, there were many folds similar to postsynaptic membranes in the cellular membranes and there were many grains in it.But there was no synaptic vesicle and pre-synaptic membranes near them.Compensatory hypertrophy and atrophy of muscle fiber coexist at 90 days after injection in group B.Conclusions Injection of botulinum toxin type A maybe induce the sprout of nerve and degeneration of the partial postsynaptic where there are many grains. It also accelerates muscle atrophy, but it induces compensatory hypertrophy of muscle fiber at later stage. It implies that injection of botulinum toxin type A can improve spasm symptom of the gastrocnemius muscles and this is good for muscle fiber repairing.

Chinese Journal of Gastrointestinal Surgery 2011;14(5):311-313
Robotic system helps surgeons in performing surgery. Currently Da Vinci system is the most popular. Da Vinci system has been used for the stomach and bowel diseases in 27 cases(18 cases of stomach and 9 cases of colon and rectum) in the Zhongshan Hospital, Fudan University. Accurate preoperative staging is crucial, and Da Vinci system is advantageous in lymph node dissection, preservation of nerve plexus, and complete resection of mesorectum. Adoption of gastrointestinal tract reconstruction technique should depend on the operation and experience in surgery. Though Da Vinci system has limitations and the cost is high, it is believed to be the future trend.

Objective To prepare Exosomes secreted by mouse hepatoma cell (H_(22)) and heat stressed Exosomes (HS-Exo) derived from heat stress-treated mouse hepatoma cell (H_(22)), in order to study the possible anti-tumor immune mechanism. Methods Exosomes and HS-Exo were purified by serial ultracentrifugation and sucrose density gradiant centrifugation, and were observed and identified by electron microscope. The components and production of the protein and the effects of the host immune response against hepatocellular carcinoma of HS-Exo were observed by using Exosomes as the control. Their immunological factors were detected by Western blot. Lymphocyte proliferation and specific cytotoxic activity of mouse splenic cells were determined by MTT. CD4~+ and CD8~+ lymphocytes infiltration in mouse tumor tissues immunized by both were analysed by immunohistochemical staining. Results HS-Exo was similar in morphology to the Exosomes, the important immune-ralated protein expressed in HS-Exo was increased (P<0.05). HS-Exo immunized mouse group showed more effective inhibition of tumor growth, better-induced lymphocyte proliferation, more significantly enhanced the cytotoxic activity of spleen lymphocytes, as well as a more prominent role in tumor therapy than Exosomes immunized mouse control group (P<0.05). Conclusions Heat stress treatment method for the preparation of HS-Exo was feasible. HS-Exo had a stronger role in the immuneactivity and tumor treatment than control Exosomes.

Journal of Kunming Medical University 2001;22(1):39-41
Objective an experimental animal model of acute myoc ardial infarction (ANI) was established by opening chest and ligation of the left anterior descending (LAD) coronary artery. Methods a total of 20 rabbits were opened chest and ligated LAD under sterilization. Electrocardiogram (ECG) and myocardial-enzymes in blood serum were investigated. Results ECG of all rabbi t s showed normal before operation. Irmediately after and 1/2 hour after ligation , ST-segment elevated and ECG showed ambulatory changes for 7 and 9 rabbits respectively. Two hours after LAD ligation, the change of ECG for 2 rabbits wa s not typical and 2 of them died during experiment. Four weeks after operation, E CG of 18 rabbits showed the chest leads had pathologic Q waves. Twenty-four ho urs after LAD ligation, AST, LDH, LDH-1, CK and CK-MB in blood serum were significantly increased. There was significant difference compared with before operation (except LDH) (P<0. 0l). Conclusions:The method was sim ple and well repeated. The formation of myocardial infarction was reliable and rabbits were maintained for a long time after operation. It provides a valuable animal mode l for the experiment study of coronary heart disease.
