1.Effects of Sinomenine on Proliferation of Human Fibroblast-like Synoviocyte and Expression of MMP-3 mRNA from Patients with Rheumatoid Arthritis

Objective To observe the effect of sinomenine on the proliferation of fibroblast-like synoviocytes (FLS) and expression level of MMP-3 secreted by FLS in rheumatoid arthritis in vitro. Methods FLS were obtained by digesting synovial tissues with collagens and were divided into four groups:sinomenine high, middle, low concentration and control group. The proliferation of FLS was assessed by methyl- thiazolyl-tetrazolium (MTT) assay and the mRNA expression of MMP-3 was measured by semi-quantitative RT-PCR respectively after treated by sinomenine. 4. HPLC fingerprint of Xintongtai Granules

Objective: To establish an HPLC method for fingerprint analysis of Xintongtai Granules (XG) for its quality control. Methods: The chromatographic behaviors were obtained by a Phenomenext Luna C18 column (250 mm × 4.6 mm, 5 μm) with gradient elution using 0.1% formic acid solution-methanol as the mobile phase. The detection wavelength was 280 nm, the volume flow rate was 1.0 mL/min, and the column temperature was 30 ℃. The samples of 10 batches of XG were determined, and the chromatographic fingerprint similarity evaluation system of Chinese medicine (2012 edition) was used to establish the common pattern of XG fingerprints, and the similarity was calculated. Then the common peaks were identified by the reference chromatogram. Results: HPLC fingerprints of XG were established by the determination of 10 batches of samples. The similarity was above 0.95. A total of 26 common peaks were calibrated. Three mutual peaks (No. 21, 22, 26 peaks) were from Salviae Miltiorrhizae Radix et Rhizoma, eight mutual peaks (No. 4—10, 16 peaks) were from Puerariae Lobatae Radix, seven mutual peaks (No. 13, 15, 17—20, 23 peaks) were from Aurantii Fructus, No. 12 peak was from Chuanxiong Rhizoma, No. 14 peaks was from Glycyrrhizae Radix et Rhizoma, No. 1 peak was shared by Notoginseng Radix et Rhizoma and Glycyrrhizae Radix et Rhizoma, No. 2 peaks was shared by Notoginseng Radix et Rhizoma, Aucklandiae Radix, and Crataegi Fructus, No. 3 peaks was shared by Chuanxiong Rhizoma and Aucklandiae Radix, No. 11 peaks was shared by Puerariae Lobatae Radix and Curcumae Radix, the No. 24 peaks was shared by Chuanxiong Rhizoma and Aurantii Fructus, and No. 25 peaks was shared by Curcumae Radix and Aurantii Fructus. The common peaks were all assigned to each medicinal material and identified by reference chromatograms: No. 7, 12, 22, and 26 peaks were puerarin, ferulic acid, salvianolic acid B, and tanshinone IIA. Conclusion: The similarity results of 10 batches of samples indicate that the particle preparation process is stable and feasible, and the established HPLC fingerprint method is stable and reliable. It can be used to measure the stability of XG production process and the controllability of finished product quality.
5. Optimization of water extraction technology of Yiqi Huoxue Prescription based on orthogonal test design and BP neural network

Objective: To optimize the water extraction technology parameters of Yiqi Huoxue Prescription (YHP). Methods: On the basis of single factor experiment, orthogonal experiment design was used to evaluate the transfer rate and extraction yield of salvianolic acid B and hydroxysafflower yellow A by using adding water, extraction time and soaking time as factors. The comprehensive score was obtained by G1-entropy weight method. The optimal water extraction technology was obtained by orthogonal test design, and another method-BP neural network modeling was used to optimize the network model and target optimization. The two analytical methods were compared in the verification experiment to find the optimal water extraction technology parameters of YHP. Results: Based on the comparison of the two analytical methods, it was found that the comprehensive score of the optimal water extraction technology obtained by orthogonal test analysis was slightly higher than that obtained by BP neural network modeling. Therefore, it was finally determined that the optimal water extraction technology parameters of YHP were as follow: water extraction for three times, soaking for 0.5 h, adding water of 20 times, and extracting time for 3.5 h. Conclusion: The optimal water extraction technology of YHP is stable and feasible, which provides new ideas and references for the development and modernization of new drugs of compound Chinese medicine.
6.EFFECTS OF (-)EPIGALLOCATECHIN GALLATE AND BLACK TEA POLYPHENOLS ON LIPID METABOLISM RELATED GENE EXPRESSION OF HepG_2 CELLS

Objective: To investigate the effect of (-) epigallocatechin gallate (EGCG) and black tea polyphenols on the lipid metabolism related gene expression profile of human hepatocellular carcinoma HepG2 cells. Method: The total RNA was isolated from HepG2 cells treated with EGCG (5? mol/L), black tea polyphenols (5?g/ml) or 0.1%DMSO (control) for 8 h and was hybridized to Human 14k cDNA microarray for gene expression profile analysis. Real-time RT-PCR was conducted to confirm microarray data. Results: A total of 13 and 29 genes related to lipid metabolism showed differential change after EGCG or black tea polphenols treatment respectively, of which six genes showed consistent expression. The results of real-time PCR were consistent with the microarray data. Conclusion: The mechanism(s) by which EGCG and black tea polyphenols exerts its effects on lipid metabolism are comprehensive. The novel target identified in this study may provide new evidence for further investigation in the hypolipidemic effects of tea polyphenols.
7.Investigation and analysis of aircrew ametropia and related factors

AIM: To investigate the refractive distribution and analysis risk factors for aircrew ametropia.
METHODS: The number of 49 cases with ametropia from 1031 aircrew during May 2013 to May 2014 were reviewed. Various types of refraction composition, age, type, position, time of flight with the subjective assessment of aircrew were analyzed and compared.
RESULTS:Of 49 cases, 43 cases (88%) were myopia, 6 cases ( 12%) were hypermetropia. , Detection rates were higher in age over 50 years aircrew and flight time more than 3000h. Detection rates were lower in self-conscious symptom heavy aircrew, fighter aircrew and good habit of using eyes.
CONCLUSION: The myopia incidence in aircrew with age >50 years and long flight time is higher, than that of fighter pilots and good habit of using eyes. We should pay attention to the increasing late-onset myopia of aviators and habit of using eyes, work intensity and time of using eyes about aircrew.

9.A study on retinal angiomatous proliferation and its mechanism in apolipoprotein E-deficient mouse with dyslipidemia

Background Retinal angiomatous proliferation (RAP) is a subtype of neovascular age-related macular degeneration (AMD).There are currently very few studies on RAP.Objective This study was to explore the pathogenic mechanism of RAP in apolipoprotein E-deficient (ApoE-/-) mice with dyslipidemia.Methods Twenty-four 2-month-old SPF ApoE-/-mice were randomly divided into the high fat diet group and the normal diet group,and twelve 2-month-old C57BL/6 mice received the normal diet as controls.A diet with a higher content of fat was given for 4 consecutive months in the high fat diet group,and normal diet was given in the same way in the mice of the normal diet group.The mice were sacrificed at 6 months of age.The expression of vascular endothelial growth factor (VEGF) in the retinal pigment epithelial (RPE) cells,the expression of vascular endothelial growth factor receptor-2 (VEGFR-2) in the outer plexiform layer (OPL),microvascular density (MVD) and microvascular area (MVA) in the OPL were examined by immunohistochemistry and semi-quantitatively by histopathology with the Mias 2000 Imaging Analyzer System.The expression of VEGF protein in the retina was examined by Western blot.Results The MVD in the retinal OPL were (20.67±3.20) and (19.50± 1.87),respectively,in the ApoE-/-mice of the high fat diet group and the normal diet group,which were significantly higher than that (12.50±1.87) of the C57BL/6 normal diet group (all at P<0.01).MVA in the retinal OPL were (626.49± 120.99) μm2 and (514.06±88.83) μm2 in the ApoE-/-mice of the high fat diet group and the normal diet group,respectively,showing a significant increase in comparison with the (336.52±84.96) μm2 of the C57BL/6 normal diet group (P<0.01).The staining area of VEGF in RPE cells was (21 048±1849) μm2 in the ApoE-/-mice of the high fat diet group,showing a significant increase in comparison with the (17 116±2023) μm2 of the C57BL/6 normal diet group.However,no significant difference was found in the staining area of VEGF between the ApoE-/-mice with normal diet group and the C57BL/6 normal diet group ([17 854±2967] μm2 vs.[17 116±2023] μm2) (P>0.05).Significant elevation was also seen in the staining area of VEGFR-2 in the retinal OPL of the ApoE-/-mice of the high fat diet group (12 193±3806)μm2 and the ApoE-/-mice of the normal diet group (11 969± 3616)xm2 compared with C57BL/6 mice of the normal diet group (5387±2225)μm2(all at P<0.01).The relative expression values (VEGF/β-actin) of VEGF in the retinas were (1.51 ±0.32) and (1.17±0.39) in the ApoE-/-mice of the high fat diet group and the normal diet group,respectively,showing a significant increase in comparison with (0.28±0.14) of the C57BL/6 normal diet group (P<0.01).Conclusions The expression of VEGF and VEGFR-2 in the retinas increases in the ApoE-/-mouse,which leads to the enlargement of MVD and MVA in the retinal OPL and subsequent RAP occurrence.
10.Ultrasound guided positioning of laryngeal mask airway in children

Objective To evaluate the efficacy of ultrasound-guided laryngeal mask airway (LMA) in children.Methods ASA Ⅰ and Ⅱ children (aged 1 to 6 years) scheduled for orthopedic surgery of limbs were recruited in this study.ALMA was inserted after induction of general anesthesia.After mechanical ventilation for 5 minutes,the air-filled cuff was deflated and then inflated with saline.Ultrasonography was then performed by a high frequency linear-array transducer to confirm the correct placement of the LMA.Results The LMA can be easily recognized on the transverse view of the thyroid cartilage notch,appearing as a circle around the glottis,which was called the dumbbell sign.On the sagittal view of the thyroid cartilage,the LMA has an archlike appearance which ended below the thyroid cartilage.Oblique scan over the cricoid cartilage shows the tip of LMA is located at the upper portion of the esophagus.Conclusions Filling the LMA cuff with saline instead of air allows better LMA visualization by ultrasound.This method enhances correct positioning of the LMA,which is crucial to ensure safe ventilation.
