1. Correlation of comminution degree and astragalus polysaccharides yield in Mt. Hengshan Astragalus membranaceus

Objective: To study the correlation between the comminution degree and astragalus polysaccharides yield in Mt. Hengshan Astragalus membranaceus (MHAM). Methods: MHAM was processed with the ultrafine treatment by mini plant mill, and grouped according to the different sizes. The yields of astragalus polysaccharides in each group were obtained under the different temperatures and ratios of material to liquid by water and CaO extraction methods. Results: When the powder size ≥ 600-mesh, the yield of astragalus polysaccharides with CaO extraction is higher than that with water extraction; When the powder size ≤ 800-mesh, the yields are almost the same by the two extraction methods. With the methods of both water and CaO extraction, the highest yields of astragalus polysaccharides (8.36% and 8.49%) were obtained at the following conditions: 800-mesh crushing granularity, 80°C extraction temperature, and 1:8 solid-liquid ratio. Conclusion: The yield of astragalus polysaccharides and the comminution degree of MHAM are related.
3.Cluster Analysis of Medication Laws for Treating Coronary Heart Disease by Distinguished Veteran Doctors of Traditional Chinese Medicine.

OBJECTIVETo provide inspiration and ideas for clinical treatment of coronary heart disease (CHD) by data mining technology based frequency analysis and cluster analysis of medical records, prescriptions and herbs in treating CHD by distinguished veteran doctors of traditional Chinese Dedicine (TCM).
METHODSTotally 386 medical cases were retrieved from Wanfang Data, Chinese Scientific Journals Database (VIP medical information resources system, China National Knowledge Infrastructure (CNKI), and Typical Collections of Medical Cases by Contemporary Distinguished Veteran Doctors of Traditional Chinese Medicine. They input into database trimmed after unified standard. Medication laws of CHD by distinguished veteran doctors of TCM were analyzed using frequency analysis and cluster analysis, and so on.
RESULTSDistinguished veteran doctors of TCM frequently used top ten herbs in treatment of C D as Salvia miltiorrhiz , Ligusticum wallichii, Trichosanthes kirilowi, Pinellia ternat, Angelica sinensis, Poria coco stragalu , Panax ginseng, Allium macrostemon, and Radix Ophiopogonis. Cluster analysis summarized that there were 16 herb pairs commonly used, 7drug assemblies consisting of 3 herbs and 5 drug assemblies consisting of multiple herbs.
CONCLUSIONSDistinguished veteran doctors of TCM mainly used herbs assemblies capable for invigorating Pi to resolve phlegm, and promoting qi and activating blood circulation in treating CHD. Meanwhile, they concurrently used herbs combination of nourishing Xin and tranquilization, and regulating yin and yang.

4.Immune modulation by mouse lymphocytes co-culturedwith cord blood-derived multipotent stem cells in Alzheimer's disease model mice

Objective To investigate the immune modulation effect of lymphocytes co-cultured with human cord blood derived-multipotent stem cells(CB-SCs) and to explore their therapeutic potential for Alzheimer's Disease (AD) model mice.Methods CB-SCs were isolated from human cord blood.Lymphocytes were isolated from spleens of AD model mice.The lymphocytes were co-cultured with CB-SCs or cultured alone for 72 h.AD model mice were divided into experimental group and control group randomly,and then the experimental group mice were administrated with lymphocytes co-cultured with CB-SCs and control group were administrated with lymphocytes cultured alone by caudal vein injection.Then,the behavior experiment was carried out.The CD4+CD25+Foxp3+Tregswere detected by flow cytometry.The protein expression of TNF-αand IL-10 in peripheral blood was detected by ELISA,The mRNA expression of TNF-α and IL-10 in brain tissue was detected by PCR.The amyloid-β(Aβ)plaques were detected by immunohistochemistry.Results 1)There was spatial learning and memory improvement on experimental group.2)The Aβ plaques of experimental group decreased.3)The percentage of CD4+CD25+Foxp3+Tregs in peripheral blood and IL-10 level in plasma were higher in experimental group(P<0.05).The pro-inflammatory cytokines,TNF-α level in plasma of experimental group was lower than that in the control group(P<0.001).4)The mRNA expression of IL-10 in brain was higher in experimental group as compared to those in the control group(P<0.05),and the mRNA expression of TNF-α of experimental group was lower than that in the control group(P<0.05).Conclusions The lymphocytes co-cultured with CB-SCs have immunotherapeutic effect in AD model mice,which is mainly displayed with increased proportion of Tregs and enhanced anti-inflammatory function of Tregs.

6.Effect of bee pollen on development of immune organ of animal.

OBJECTIVETo study on the effect of been pollen on development of immune organ of animal.
METHODA total of 144 one day-old broilers were randomly divided into 2 groups, in which each group included 72 chickens. The control group was fed on the basal diet for 42 days, and that of experiment group supplemented 1.5% bee pollen. Six chickens in each group were selected and slaughtered at 7, 14, 21, 28, 35, 42 days respectively, and the thymuses, cloacal bursa and spleens were obtained, weighted, fixed in Bouin liquid and made into paraffin section.
RESULTCompared with control group, the weight and the relative weight of thymuses, cloacal bursa and spleens of experiment group increased significantly (P < 0.05) or extremely significantly (P < 0.01). In experiment group, the cortex of thymic lobule, bursa nodule and Periarterial Lymphatic Sheaths thicken obviously; the volume of bursa nodule, splenic nodule and ellipsoid augmented, and the germinal center of splenic nodule were obvious; the thymic corpuscle increased; the plica of cloacal bursa developed well and the degenerating of it retarded.
CONCLUSIONThe diet supplemented bee pollen could boost the early development of thymus and cloacal bursa, retard the degenerating of cloacal bursa and promote the immune response of spleen.

7.Construction of eukaryotic expression vector of unknown KH gene and its effect on cell proliferation.

OBJECTIVETo construct an eukaryotic expression vector of open reading frame of unknown KH gene (KH-ORF), and investigate its effect on cell proliferation.
METHODSThe pCI-neo-KH-ORF expression vector was constructed by DNA recombinant technique and was introduced into COS-7 cells and K562 cells by lipofectactin-mediated DNA transfection. Expression of KH-ORF mRNA was detected by RT-PCR. The effect of KH-ORF on cell cycle of COS-7 cells and K562 cells was evaluated by flow cytometry (FCM). Effect on cell proliferation of COS-7 cells was tested by MTT assay and that on K562 cells was analyzed by growth curves and LDH activity measurement.
RESULTS(1) KH-ORF mRNA was expressed both in COS-7 cells and K562 cells. (2) The cell cycle and cell proliferation of COS-7 cells were unaffected significantly. (3) The proportion of cells in S phase was increased in pCI-neo-KH-ORF-transfected K562 cells; and growth curves and LDH activity indicated enhanced cell proliferation.
CONCLUSIONKH gene may be a leukemia gene related to proliferation of K562 cells.

8.Correlation analysis of clinic and SCN1A mutation in febrile seizures plus with autism

Objective To study the clinical characteristics of autism in febrile seizures plus (FS+) and the relationship between autism and SCN1A mutation. Methods Clinical data of 103 patients with FS+ treated in epilepsy centre of the Second Affiliated Hospital of Guangzhou Medical University were collected and analyzed. According to the international criteria, generalized epilepsy with febrile seizures plus (GEFS+), partial seizures with febrile seizures plus (PEFS+), Dravet syndrome (DS) and autism were diagnosed. Genomic DNA was obtained from blood samples. SCN1A were PCR amplified and mutations were detected by DHPLC and sequencing. Result Mental retardation was found in 53.8%of patients with GEFS+, 69.2%of patients with PEFS+, and all patients with DS, respectively. One in GEFS+, one in PEFS+and nine in DS patients were accompanied with autism (P<0.01). Among FS+patients with autism, one SCN1A mutation was found in PEFS+patients, while six SCN1A mutations were found in DS patients. Conclusions Majority of GEFS+and PEFS+patients showed mental retardation, while all the DS patients were accompanied with retardation. The occurrence of autism with DS is higher than GEFS+and PEFS+. No definite relationship between autism and SCN1A mutation was indicated.
