3.Ultrasonic Extraction and Purification Technology of Solanesol from Tobacco Leaf

OBJECTIVE:To optimize ultrasonic extraction and purification technology of solanesol from tobacco leaf. METH-ODS:Using extraction rate and transport rate of solanesol as indexes,single factor test was used to investigate liquid-solid ratio, ultrasonic extraction temperature and time,ultrasonic power and extraction times,and the amount of soap alkali lye(volume ratio of soap alkali lye to extraction liquid),acidizing fluids (volume ratio of acidizing fluids to soap alkali lye extract),extraction times of purification technology. Optimized technology was validated,and the purity of solanesol was calculated;the amount of ex-tracted solanesol was compared between this method and traditional extraction method (spending 30 h),solvent continuous cyclic extraction (spending 5-6 h). RESULTS:Optimized extraction technology was as follows as volume ratio of soap alkali lye to ex-traction liquid 1∶14,ultrasonic extraction temperature 70 ℃,ultrasonic extraction time 60 min,ultrasonic power 120 W,extract-ing for 3 times;optimized purification technology was as follows as volume ratio of soap alkali lye to extraction liquid 2∶35,vol-ume ratio of acidizing fluids to soap alkali lye extract 2∶14,extracting for 4 times. In validation test,extraction rate,transport rate and purity were 92.45%(RSD=0.46%,n=3),79.88%(RSD=0.30%,n=3)and 55.86%(RSD=0.40%,n=3). The amount of solanesol extracted with 3 methods were 52.22,45.22 and 26.10 mg/g. CONCLUSIONS:The optimized technology is simple and stable,costs less time and saves source with high extraction amount and purity,which is suitable for production,extraction and purification of solanesol from tobacco leaf.

Objective To explore the changes of cardiomyocytes apoptosis in the effect of irhesartan and imidapril on regressing cardiac hypertrophy in spontaneous hypertensive rat (SHR), and its mechanism. Methods Thirty 13-week-old SHR were randomly divided into three groups: SHR positive control group, irhesartan treated group(50 mg ?kg-1 ?day -1 ) , imidapril treated group(3 mg ?kg -1 ?day-1 ), and normotensive Wistar-Kyoto rats(WKY) as controls. Blood pressure of rats were monitored periodically. After 15 weeks, all rats were killed and left ventricle weight(LVW) were measured, plasma and myocardium angiotensin Ⅱ concentrations were examined by radioimmunoassay. Then the changes of cardiomyocytes apoptosis using in situ TDT-mediated dUTP nick end labeling(Tunel) were studied. Objective To investigate the effects of ATP1A1 knockdown by RNA interference(RNAi) on proliferation of human U251 glioma stem cells .Methods The human U251 glioma stem cells were infected with lentivirus expressing ATP1A1-shRNA . The mRNA and protein expressions of ATP1A1 in U251 glioma stem cells were detected by RT-qPCR and Western blotting ,re-spectively .The cell cycle and apoptosis were evaluated by flow cytometry .The proliferation of U251 glioma stem cells was deter-mined by MTT assay .Results The expressions of ATP1A1 in U251 glioma stem cells transfected with ATP1A1-shRNA were in-hibited significantly at both mRNA and protein levels ,with an inhibitory rate of 84 .15% for ATP1A1 mRNA and of 55 .33% for ATP1A1 protein respectively .The proliferation of cells was inhibited ,the cell apoptotic rate was significantly increased and the cell cycle was arrested in G1 phase and S phase decreased significantly in ATP1A1-shRNA cells(P<0 .05) .Conclusion RNAi targe-ting ATP1A1 gene could down-regulates the ATP1A1 expression ,induces cell apoptosis ,regulates cell phase redistribution and in-hibits cell proliferation in U 251 glioma stem cells .
8.Surgical treatment for chronic instability of the elbow

Objective To study the cause and treatment of chronic instability of the elbow.Methods Between August 1998 and August 2002,12 patients with chronic instability of the elbow were treated in our de- partment:They underwent complete medical check-ups and imaging examinations.Surgical exploration was done to determine the causes of their conditions.Their radial and(or)ulnar collateral ligaments were repaired respectively or simultaneously through the medial and lateral approaches for all the patients.All the patients were followed up for two to six years,with an average of three years.Results The follow-ups revealed no recurrent dislocation of elbow in this group.The stress valgus test and lateral pivot shift test(PST)were negative for all the patients.No elbow pain occurred.The mean flexion and extension of elbow ranged from 0 to 135?.The mean pronation was 85?and mean supination 80?.According the evaluation system of Chinese Medical Association,eight cases were rated as excellent and four as fine,with the excellent and fine rate of 100%.Conclusions Lesions of radial and(or) ulnar collateral ligaments are the major cause for chronic instability of the elbow.Surgical reconstruction of the collateral ligaments can obtain good effects in treatment of recurrent dislocation of the elbow.
9.Individualization of tacrolimus dosage based on CYP3A5 * 3 gene polymorphism: a prospective,controlled study

Objective To investigate the value of Cytochrome P450 (CYP3A5) * 3 gene polymorphism in providing individualized administration for the use of tacrolimus (Tac) in renal transplantation recipients.Method Pyrophosphate sequencing method was used to determine the CYP3A5 * 3 genotype of renal transplant patients in the first day after surgery.Sixty recipients were divided into experiment group and control group.Both groups of patients were routinely given the initial dose of Tac-4.0 mg/day in the first day after surgery.The experiment group of patients were given different doses of Tac based on the different CYP3A5 * 3 genotypes at the third day after surgery [for AA:0.12 mg/(kg· day),and for GG:0.06 mg/(kg· day)],and the control group of patients were given different dosages of Tac according to drug concentration.Different parameters were compared between two groups of patients:percentage of patients reaching the target concentration (3-8 μg/L) at the fifth day after surgery,days required to reach the target concentration level,times needed to adjust the dosage of Tac within two weeks.Result The percentage of patients reaching the target concentration in experiment group and control group was 90% and 46.67%,respectively (P< 0.05).Days required to reach the target concentration were (3.67 ± 1.32) and (7.57 ± 3.42) on average,respectively (P < 0.05).Times of adjusting the Tac dose in experiment group was significantly less than those in the control group (P<0.05).In the experiment group,the target concentration was obtained even without dosage adjustment (70%).Conclusion Individualized adjustment of Tac doses for patients according to recipients' different CYP3A5 * 3 genotypes is beneficial for reaching target concentration as soon as possible,which is superior to traditional dosage regimen.
10.Study on blood lipids of patients with ischemic cerebral vascular disease

Objective To study blood lipids levels of patients with ischemic cerebral vascular disease (ICVD).Methods The blood lipids levels were measured in 2886 subjects with ICVD enrolled from 1991 to 2004,in which 1430 subjects from 2000 to 2004 were classified into a sub-group;400 healthy persons receiving physical examination were enrolled as controls.Results The blood TC,TG and LDL-C levels were significantly higher in the sub-group than those in the control group.Both in males and females, the morbidity of abnormal blood lipid metabolism and blood lipids level gradually increased with the age and the lapse of decades.Total cholesterol level of male younger than fifty,averaging (4.43?0.51) mmol/L, increased to (4.96?0.85) mmol/L after fifty years old.The same thing happened to females,from (4.30?0.49) mmol/L to (5.01?0.90) mmol/L.TG and LDL-C increased in patients older than fifty compared with those younger than fifty.In the groups above 50 years old,the blood lipids levels of females were significantly higher than those of males.The morbidity of abnormal blood lipid metabolism of male increased from 24.5% in (1991 to 1994) to 38.1% (2001 to 2004) and of female from 22.3% (1991 to 1994) to 38.5% (2001 to 2004).Conclusions Abnormal blood lipid in Foshan is one of the most important reasons to ICVD.
