2.An improved method for directional differentiation and efficient production of neurons from embryonic stem cells in vitro.

To establish a method of directional differentiation and efficient production of neurons from embryonic stem cells (ES cells) in vitro, based on the 4-/4+ protocol described by Bain, a new method was established to induce ES cells differentiating into neurons by means of three-step differentiation using all-trans retinoic acid (ATRA) combined with astrocyte-conditioned medium (ACM) in Vitro. The totipotency of ES cells was identified by observation of cells' morphology and formations of teratoma in immunocompromised mice. The cells' differentiation was evaluated continuously by the detection of the specific cellular markers of neural stem cells, neurons and astrocytes, including nestin, NSE and GFAP using immunohistochemistry assay. The NSE positive cells' ratio of the differentiated cells was determined by flow cytometry. It was found that the transparent circular clusters surrounding embryoid bodies induced with combining induction protocol formed just after 24 h and gradually enlarged later. This phenomenon could not be observed in EBs induced only by ATRA. The NSE positive cells' ratio in the cells induced with ATRA and ACM was higher than that of the cells induced by ATRA at different time points of differentiation, and finally reached up to 73.5% among the total differentiated population. It was concluded that ES cells could be induced into neurons with high purity and yield by means of inducing method combining with ATRA and ACM.

3.Correlation analysis of NLR,PLR and tyrosine-kinase inhibitors in the treatment of advanced lung adenocarcinoma

Objective To analyze the correlation between neutrophil-to-lymphocyte ratio(NLR),platelet-to-lymphocyte ratio(PLR) in peripheral blood and the therapeutic effect and prognosis of patients with advanced lung adenocarcinoma treated with tyrosine-kinase inhibitors.Methods Retrospective analysis was utilized to measure the clinical pathological characteristics and follow-up data of 40 cases with advanced lung adenocarcinoma definitely diagnosed by pathology.Correlation analysis was performed by Spearman test,and comparison of the sample rate was measured by the chi square test.Survival analysis was performed by Kaplan-Meier method,Log-rank test was used to compare patients survival difference among different groups.COX regression model was utilized to analyze the prognostic factors of patients with lung cancer.Results Through the ROC curve,the optimal cut-off points of NLR and PLR were calculated to be 3.74 and 205.1 respectively.In NLR<3.74 group,objective response rate(ORR),the median progression-free survival(PFS) and the median overall survival(OS) were 86.9%(20/23),9.02 months and 12.37 months respectively.In NLR ≥3.74 group,ORR,the median PFS and the median OS was ORR were 35.2%(6/17),3.15 months and 5.12 months respectively.Compared with NLR ≥ 3.74 group,NLR<3.74 group had a higher ORR(P=0.036),PFS(P=0.011) and OS(P=0.021),the difference was statistically significant.In PLR<205.1 group,ORR,the median PFS and the median OS was ORR were 85.7%(18/21),9.67 months and 14.35 months respectively.In PLR ≥ 205.1 group,ORR,the median PFS and the median OS was ORR were 42.1%(8/19),3.21 months and 9.25 months respectively.Compared with group PLR ≥ 205.1,PLR<205.1 group had a higher ORR(P=0.045) and PFS(P=0.018),the difference was statistically significant.COX multivariate analysis showed that NLR was an independent prognostic factor for PFS(RR=1.091,95%CI:1.004-1.185,P=0.039) and OS(RR=0.986,95%CI:0.932-1.148,P=0.021).PLR was an independent prognostic indicator of PFS(RR=1.100,95%CI:0.996-1.005,P=0.024).Conclusion NLR and PLR may be independent prognostic factors in patients with advanced lung adenocarcinoma.
4.The correlation between air pollutants and outpatient visits in Zhoushan

Objective :
To explore the correlation between air pollution and outpatient visits,and to provide evidence for health risk assessment of air pollutants and intervention.
Methods :
The data of air pollutants and outpatient visits in 2016 in Zhoushan Hospital were collected,and the outpatient volume on the days when the concentration of air pollutants reached and exceeded the standard were compared. Spearman rank correlation analysis was used to explore the relationship between outpatient volume and the concentration of air pollutants.
Results :
In 2016,the median(inter-quartile range)of daily outpatient volume in Zhoushan Hospital was 3 304(1 638)person-times. O3,PM2.5 and PM10 were the primary air pollutants in Zhoushan in 2016. The average daily outpatient volume of internal medicine,circulatory system,other diseases and all when the air was polluted at light level or above were higher than those when the air quality was good(P< 0.05). CO concentration was positively correlated with the average daily outpatient volume of respiratory system and circulatory system(P< 0.05),was negatively correlated with the average daily outpatient volume of all(P< 0.05). O3-8 h concentration was positively correlated with the average daily outpatient volume of internal medicine,other diseases and all(P< 0.05),and was negatively correlated with the average daily outpatient volume of respiratory system and pediatrics(P< 0.05). SO2 concentration was negatively correlated with the average daily outpatient volume of respiratory system,skin and subcutaneous tissue(P< 0.05). The concentration of NO2,PM10 and PM2.5 were positively correlated with the average daily outpatient volume of respiratory system and pediatrics(P< 0.05).
Conclusion
The main air pollutants in Zhoushan were O3,PM2.5 and PM10. When they exceed the limits,the outpatient volume would increase.
5.Advances in sperm membrane antigens.

A series of sperm membrane antigens, the important molecular markers associated with the program of spermatogenesis and maturation of sperm, which also are for attachment of the physiological mechanism of the sperm-egg interaction and fusion and the pathological changes in the infertility, were reviewed. Some traditional crucial sperm membrane proteins, such as FA-1, PH-20, fertilin, have been cloned and sequenced, and some novel gene which encode new proteins associated with the sperm membrane also have been screened out successively. All of these provided a basement for lucubration of these proteins and the research of the contraception vaccine.

7.Anthraquinones and triterpenoids from roots of Knoxia roxburghii.

Nine compounds were isolated from an ethanol extract of the roots of K. roxburghii by using a combination of various chromatographic techniques including column chromatography over silica gel, MCI gel, Sephadex LH-20, and reversed-phase HPLC. On the basis of physical-chemical properties and spectroscopic data analysis, their structures were identified as munjistin (1), 1-methoxy-3,6-dihydroxy-2-hydroxymethyl-9,10-anthraquinone (2), 1,2,3-trihydroxy-9,10-anthraquinone (3), arjunolic acid (4), hyptatic acid-A (5), hyptatic acid-B (6), 2α,3β,24-trihydroxyurs-12-en-28-oic acid (7), 2α,3β,23-trihydroxyurs-12-en-28-oic acid (8), and daucosterol (9). Compounds 1-9 were obtained from this genus for the first time.

8.Sexual hormone and sperm cytological test in patients with aspermia.

OBJECTIVESTo test the serum sexual hormone and sperm cytology in aspermia patients.
METHODSRadioimmunoassay (RIA) and Wright-Giemsa stain were used to detect serum sexual hormone and sperm cytology in 45 aspermia patients.
RESULTSAmong the 45 patients, spermatogenetic cells were detected in 16 patients(35.6%). The level of serum testerone were decreased in 16 patients, and FSH, LH were increased in 13 cases as well as that of PRL in 4 cases.
CONCLUSIONSThese results suggested that the testis function was demaged, the ratio of T/LH could further reflect the function of Leydig cells, the detection of PRL were significant only in diagnosis of aspermia induced by hypermia. It is an important index in identifying obstructive and non-obstructive aspermia and has important significance in estimating the extention of testis demage and commanding the clinical treatment of serum hormone detection and sperm cell analysis.

9.Study on the risk factors and characteristics on childhood sexual abuses among female students in a college.

OBJECTIVETo study the risk factors and abusers' characteristics on childhood sexual abuses among female students to provide reference data for preventive measures on child sexual abuse.
METHODSAnnonymous questionnaire survey of 701 adult female students in a college about their personal experience on childhood sexual abuses, their home conditions and abusers' characteristics.
RESULTSA total number of 701 female students were surveyed. 115 (22.11%) experienced non-physical contact sexual abuses before the age of 18, including 26 (3.71%) who suffered from attempted and 8 (1.14%) were forced with genital or anal sexual intercourse. 117 (16.69%) said they had suffered from sexual abuse before the age of 16. Most abusers were males (99.4%) and only a small percentage of them (3.2%) used violence. Most non-physical contact sexual abuses were from strangers (78.7%), while 71.3% of physical contact abuses were from acquaintances including 12.5% of them were teachers, 17.5% were neighbors and 21.3% were relatives. The risk factors of child sexual abuses were: frequent use of violence, bad hobbies and poor health conditions of parents, tight family relationships, and living in remarried families. Having well-educated fathers was a preventive factor.
CONCLUSIONStudies on childhood sexual abuses among females revealed that the incidents were not rare, and family environment played an important role.

10.Advances of the study of sarcosaphagous insects in forensic medicine.

The study of sarcosaphagous insects is a subspecialty in forensic medicine based on the knowledge of entomology. It could help to determine the time of death, especially the postmortem interval in decomposed cases. This paper explores its history, species and erosion process of sarcosaphagous insects. It reviews the species identifying methods with molecular biology and entomological morphology. Details of its application in estimating postmortem interval in recent years and study of sarcosaphagous insects in the field of forensic medicine are summarized.
