1.Effects of Iodine Deficiency and Hypothyroidism during Pregnancy on the Protein Expressions of cAMP Response Element-Binding Protein in the Entorhinal Cortex of Pups

Objective To observe the effect of iodine deficiency and hypothyroidism on the protein expressions of CREB in the entorhinal cortex of pups.Methods Twenty-eight female Wistar clean rats after pregnancy were randomly divided into control group,hypothyroid 1,2 group and iodine deficient group(7 in each group).From GD6 till PN28,iodine-deficient group was administered with iodine-deficient diet [iodine content:(14.11 ?1.96) ng/g] and tap water;hypothyroid 1 and 2 group were administered with 5 mg/L and 15 mg/L PTU in the drinking water and fed with normal diet [iodine content:(470.50?46.52) ng/g].Control group received tap water and normal diet during the experiment.Five pups from each group were sacrificed and intracardiac perfused at PN7,PN14,PN21,PN28 and PN42.Brains were removed,fixed and sectioned coronally.All sections were observed and were analyzed for the protein expression of CREB by immunohistochemistry in the entorhinal cortex.Results At PN7,there was no significant difference in the expression of Ng in all four groups.At PN14 and PN21,the expression of CREB in the entorhinal cortex in hypothyroid 1,2 group and iodine deficient group were significantly lower than those of controls(P
2.THE EXPRESSION OF RETINOIC ACID RECEPTOR GENES IN LYMPH NODES OF YOUNG CHILDREN AND THE DEVELOPMENT OF B CELLS

Objective: To investigate the influence of the expression of retinoic acid receptor genes on the development of B cells in lymph nodes of young children. Method: In situ hybridization was performed on the frozen section of lymph node of young children (≤age 5 ). Six digoxin labeled antisense RNA probes for retinoic acid receptors mRNA(RAR?、?、?、RXR?、?、?)were used. The expression and distribution of retinoic acid receptor genes in lymph nodes and their influence on the differentiation and maturation of B cell were observed. RT-fluorescent PCR was also used to observe the expression levels of retinoic acid receptor genes and their relation to the maturation of B cells. Results: In situ hybridization showed all the 6 retinoic acid receptor genes were expressed in lymphocytes and reticular cells of lymph node, and the distribution was widespread. RT fluorescent PCR also showed a varying expression of 6 retinoic acid receptor genes in lymph node among different age children, but lower in children younger than 1 year, and then increased gradually with the development of immune system. Conclusion: The expression and regulation of retinoic acid receptor genes may take part in the ontogenesis of B cells, and play a key role in the regulation of retinoic acid and enforce the anti-infective immunity in children.
3.THE INFLUENCE OF RETINOIC ACID ON THE DEVELOPMENT OF LYMPH NODE B CELLS IN CHILDREN

Objective: To investigate the effect of retinoic acid on the differentiation and development of B lymphocytes, and explore the mechanism of vitaminA in increasing the production of antibodies. Method: In vitro cultured cells from children’s normal mesentery lymph nodes, before and after administration of retinoic acid or retinoic acid antagonist, the changes of cell surface markers were analyzed by flowcytometry to observe the differentiation and maturation of B cells. Results: During culture in vitro, the percent of mature CD19+IgM+ B cells increased and relatively immature CD19+IgM- B cells decreased gradually, and the changes were especially obvious at 48 h. Objective To evaluate the clinical efficacy and security of retroperitoneal laparoscopic ureterolithotomy(RLUL) for impacted ureteral calculi after radical cystectomy and ileal conduit.Methods 5 patients with unilateral impacted ureteral calculi after radical cystectomy and ileal conduit received RLUL were selected,and a retrospective study was performed for manner,duration of surgery,complications and length of stay and other indicators,and the treatment effect was evaluated.Results All procedures were successful and the mean operation time was (82.0±27.7)min,the extubation time was (5.4±1.1)d,hospitalization time was (10.0±2.9)d.Postoperative follow up for 6-48 months,there were no obvious complications.Conclusion The RLUL showed satisfactory availability and security for management ureteral calculi after radical cystectomy and ileal conduit.
8.Application of combined detection of enterovirus nucleic acid and antibody in early etiological diagnosis for hand-foot-and-mouth disease

Objective To assess the value of combined detection of enterovirus nucleic acid and antibody in early etiological diagnosis for hand-foot-and-mouth disease ( HFMD).Methods A case-control study was conducted.A total of 1 066 cases of children clinically diagnosed with HFMD from Hangzhou Children′s Hospital were involved into the research group from January to June 2014, consisting of 401 common cases and 665 severe cases; Throat swabs and serum samples from these children underwent combined detection for EV71/CA16/EV of enterovirus nucleic acid by fluorescence quantitative RT-PCR and for EV71/CA16-IgM by ELISA.All data were analyzed with SPSS 16.0.Results The total positive rate of enterovirus nucleic acid EV71/CA16/EV by fluorescence quantitative RT-PCR in the 1 066 cases of children clinically diagnosed with HFMD was 75.52%( 805/1 066 ) ( 95%CI: 72.80%-78.05%).But the total positive rate of combined detection was 91.46%( 975/1 066 ) ( 95%CI:89%.58-93.04%).The total positive rate of combined detection is higher than that of RT-PCR test(χ2 =98.338,P=0.000).The positive rate of EV71 type of combined detection was 64.63%(689/1 066)(95%CI:61.67%-67.49%),which is 15.38%higher than that of RT-PCR test 49.25%(525/1 066)(95%CI:46.21%-52.29%)(χ2 =51.453, P=0.000).In 665 severe cases of HFMD, the total positive rate of combined detection was 96.69%(643/665)(95%CI:94.95%-97.87%), which is higher than that of RT-PCR test 79.25%(527/665)(95%CI:75.92%-82.22%)(χ2 =95.607, P =0.000).In the severe cases, the positive rate of EV71 type of combined detection was 87.52%( 582/665 ) ( 95%CI:84.71%-89.89%) , which is 18.95% higher than that of RT-PCR test 68.57%(456/665) (95%CI:64.87%-72.06%) (χ2 =69.665, P=0.000).In the fatal cases, the positive rate of EV71 type of combined detection was 95.92%(94/98) (95%CI:89.28%-98.68%).Conclusions The combined detection of enterovirus nucleic acid and specific IgM antibody can significantly increase the positive rate of HFMD, especially for severe cases.The combine detection increases both the total positive rate and EV71 positive rate.Thus it has a high potential for becoming a new guidelines for laboratory diagnosis of HFMD.

Objective To observe the intervention with electroacupuncture(EA) through regulating protein kinase(MAPK/ERK) pathway for cerebral ischemia rats.Methods SD rats were randomized into pseudo-operation group,model group and EA group.And the three groups were divided into 2-hour,one-day and 3-day subgroups according to the observation time.Focal cerebral ischemia(FCI) rat models were established by middle cerebral artery occlusion(MCAO) with heat-coagulation method.EA group received EA on Baihui(GV20) and Dazhui(GV14),qd,for 2 hours,one day and 3 days respectively.The learning and memory abilities of the rats in Y maze test and their neuroethological score were observed.Immunohistochemical method was used to observe the expression of phosphorylated extracellular signal-regulated kinase(p-ERK) after ischemia in CA1 area and CA3 area.Results In the model group,the neuroethological score was markedly increased,time for arriving the safe area in Y maze was prolonged(P
