2.Effect of Acupuncture Combined with TAX on the Expression of Apoptosis Related Factor p53 and bcl-2 of Mice Lewis Lung Cancer Cells

Objective We adopt acupuncture associating with TAX,to investigate the anti-cancer effect and mechanism.Methods 48 mice with inoculated Lewis Lung Carcinoma were divided randomly into 4 groups:model group,acupuncture group,medicine group,combination of acupuncture and medicine group.Growth state of the mice and change curve of tumour.Protein expression of apoptosis related factor p53 and Bc1-2 in tumour were observed by Western Blotting method.Results Acupuncture,TAX and the combination of acupuncture and TAX separately had anti-tumor effect,with the combination was the best and improve growth state of the mice.p53 and bc1-2 can be detected in four groups.Compared with model group,the expression of other groups was lower.Compared with acupuncture group and medicine group, the exprission of combination group of acupuncture and TAX was lower.Conclution Combination of acupuncture and TAX can prohibit p53 Mutant and decrease Bcl-2,which have co-operating effect.
3.The design and clinical application of a new type of dynamic Hoffmann external fixator for tibial shaft fractures

Objective To explore biomechanical properties and clinical results of a new type of dynamic Hoffmann external fixator(NDHEF)for tibial shaft fractures.Methods Fifteen specimens of adult tibia were randomly divided into three groups and osteotomized obliquely in the mid part of the tibia.These specimens were fixed with NDHEF,improved Hoffmann external fixator(IHEF)and maltifunctional external fixator(MEF)re- spectively.The axial compression,bending,torsion and stress shielding tests were performed on the specimens in each group for biomechanical comparison.From 1995 to 2004,40 cases of open tibiofihular fractures were treated with NDHEF and 88 cases with IHEF and their clinical results were analyzed and compared.Results The biomechanical tests showed that the compressive rigidity,bending rigidity and torsion rigidity of NDHEF were similar to those of IHEF(P>0.05)but significantly stronger than those of MEF(P<0.05).The stress shielding of NDHEF was obviously weaker than that of other external fixators(P<0.05).All the 128 cases were followed up for 12 to 36 months(average,16.8 months).The mean substantial bone healing time for NDHEF treatment was 22.3 weeks and that for IHEF was 26.8 weeks.The difference was significant(P<0.05).The rates of delayed union and malunion were all 7.5% for NDHEF,but 15.9% and 4.5% respectively for IHEF.The difference was insignificant (P>0.05).Conclusions The biomechanical performance of NDHEF is superior to that of IHEF,for it can diminish the stress shielding and accelerate bone healing.It is a good external fixator for tibial fractures.

5.Rat prostate glandular epithelial cells cultured in vitro and their barrier function.

OBJECTIVETo culture rat prostate glandular epithelial cells and study their barrier functions in vitro.
METHODSRat prostate glandular epithelial cells were cultured in vitro. The expression of the tight junction protein claudin-1 was determined by immunohistochemistry, the structure and composition of the epithelial cells observed under the inverted microscope and transmission electron microscope. The transepithelial electrical resistances (TEERs) were monitored with the Millicell system. The permeability of the prostate glandular epithelial cells was assessed by the phenol red leakage test.
RESULTSCompact monolayer cell structures were formed in the prostate glandular epithelial cells cultured in vitro. Immunohistochemistry showed the expression of the tight junction protein claudin-1 and transmission electron microscopy confirmed the formation of tight junctions between the adjacent glandular epithelial cells. The TEERs in the cultured prostate glandular epithelial cells reached the peak of about (201.3 ± 3.5) Ω/cm2 on the 8th day. The phenol red leakage test manifested a decreased permeability of the cell layers with the increase of TEERs.
CONCLUSIONThe structure and function of rat prostate glandular epithelial cells are similar to those of brain capillary endothelial cells, retinal capillary endothelial cells, and intestinal epithelial cells. In vitro cultured prostate glandular epithelial cells have the barrier function and can be used as a model for the study of blood prostate barrier in vitro.

6.Bone marrow mesenchymal stem cells suppress E coli-induced bacterial prostatitis in rats.

OBJECTIVETo investigate the inhibitory effect of bone marrow mesenchymal stem cells (BMSCs) on E coliinduced prostatitis in rats.
METHODSBMSCs were isolated, cultured and amplified by the attached choice method. Fifty SD rats were randomized into five groups of equal number: normal control, acute bacterial prostatitis (ABP) , chronic bacterial prostatitis (CBP), ABP + BMSCs, and CBP + BMSCs, and the animals in the latter four groups were injected with E. coli into both sides of the prostate under ultrasound guidance for 1 - 14 days to induce ABP and for 4 - 12 weeks to induce CBP. The control rats were injected with the same amount of PBS. Two weeks after injection of BMSCs into the prostates, pathomorphological changes in the prostate were observed under the light microscope and the mRNA and protein levels of IL-1β and TNF-α determined by RT-PCR and ELISA, respectively, followed by statistical analysis with SPSS 18.0.
RESULTSHistopathological evaluation showed typical pathological inflammatory changes in the prostates of the rats in the ABP and CBP groups, including glandular structural changes, interstitial edema, inflammatory cell infiltration, and fibrous hyperplasia, which were all remarkably relieved after treated with BMSCs. The mRNA and protein levels of IL-β ([0.829 ± 0.121] and [271.75 ± 90.59] pg/ml) and TNF-α ([0.913 ± 0. 094] and [105.78 ± 19. 05] pg/ml) in the ABP and those of IL-1β ([0. 975 ± 0. 114] and [265. 31 ± 71. 34] pg/ml) and TNF-α ([0. 886 ± 0. 084] and [107. 45 ± 26. 11 ] pg/ml) in the CBP groups were significantly higher than those in the control rats ([0. 342 ± 0.087] and [45.76 17. 99] pg/ml, P <0. 05); ([0.247 ± 0.054] and ([19.42 ± 7. 75] pg/ml, P <0. 01) as well as than those in the ABP + BMSCs ([0. 433 ± 0. 072] and [51. 34 ± 22. 13] pg/ml, P < 0. 05 ) ; ( [0. 313 ± 0. 076] and [28. 38 ± 8. 78] pg/ml, P < 0. 01) and the CBP + BMSCs group ([0.396 ± 0.064] and [56.37 ± 21.22] pg/ml, P <0.05); ([0.417 ± 0.068] and [29.21 ± 10.22] pg/ml, P <0.01).
CONCLUSIONInjection of BMSCs can reduce E coli-induced prostatic inflammation reaction, which.may be associated with its reduction of inflammatory cell infiltration and the expressions of IL-1β and TNF-α in the prostate tissue.

7.Childhood trauma experience and its relation to the eye emotion recognition ability in patients with schizophrenia

ObjectiveTo explore the childhood trauma experience and its relation to the eye emotion recognition ability in patients with schizophrenia.MethodsSeventy-one schizophrenic patients and fifty-two healthy subjects were assessed with the Childhood Trauma Questionnaire (CTQ) and the Eye Basic Emotion Discrimination Task (EBEDT).ResultsThe CTQ total score in the patients group was significantly higher than those in the control group (47.86 ± 13.26 vs 34.27 ± 7.27,P < 0.01 ).The subscores in emotional abuse,physical abuse,sexual abuse,emotional neglect and physical neglect of CTQ in the patients group were also significantly higher than those in the control group.(all P< 0.01 ).Compared with controls,the patients had significantly lower total accuracy( (86.74 ± 6.16vs73.42 ± 11.86) %,P < 0.01 ) and longer total reaction time ( (2776.75 ± 703.42 vs 4321.40 ± 1956.83 )ms,P < 0.01 )of EBEDT.The significant difference were also found on the accuracy and reaction time of the EBEDT six basic emotion between the two groups ( all P < 0.01 ).Correlation analysis indicated that CTQ total score were negatively correlated with the EBEDT total accuracy ( r=-0.390,P<0.01 ),and its sub-accuracy of happiness,surprise,sadness,and anger ( respectively r =- 0.419,r =- 0.451,r =- 0.391,r =- 0.344,P < 0.01 ) in the patients group.The correlation between CTQ total score and EBEDT total accuracy was also found in the control group.ConclusionChildhood trauma is related to eye basic emotion recognition in schizophrenia patients,suggesting that the trauma during childhood can affect the social recognition ability.
8.Experimental study on anti-inflammation and analgesia effect and acute toxicity of extract from Pongamia pinnata roots

AIM: To study the effect of extract from Pongamia pinnata roots on anti-inflammation and analgesia and acute toxicity. METHODS: The models of mice ear edema induced by xylene and Cotton pellet granuloma in rats to observe the anti-inflammation effect of PRE via oral administration. The effect of PRE on analgesia was tested by measuring the latent period licking hind foot with the hot plate method and counting body twisting induced by acetic acid in mice. The acute toxicity of PRE was measured by the method of Bliss. RESULTS: PRE could significantly inhibit the ear edema caused by xylene in mice, granuloma hyperplasia caused by cotton in rats. It could significantly prolong the pain threshold on hot-plate in mice, reduce the writhing times in mice. The LD50 of PRE was 6. 371 8 g/kg, its 95% confident limit was 5. 408 4-7. 723 2 g/kg. CONCLUSION: PRE has obvious effect on anti-inflammation and analgesia and the lower acute toxicity.
9.The Clinic Study on the Treatment of Maxillary Protrusion Malocclusion with Micro-Screw Palatal Implant Anchorage

Objective To investigate the clinic application of micro-implant anchorage in the treatment of maxil-lary protrusion malocclusion. Methods Twenty-two patients,aged 18 to 25 years old,with maxillary protrusion were divid-ed into two groups:experimental group and control group with 11 patients in each group. All patients were treated with ex-traction. Micro-screw palatal implant was used in the cases of experimental group as orthodontic anchorage ,and traditional anchorage composed of extraoral arch used in the cases of control. The cephalometric films were measured before and after treatment. Statistical methods were utilized to analyze the morphological changes of facial profile and hard tissues in both groups. Results The values of U1-NA(mm:3.08±1.18 vs 8.15±3.05) and U1-SN(101.90°±3.50° vs 117.90°±6.05°) were sig-nificantly decreased after treatment compared with those before treatment in the experimental group ( P<0.01). The value of U1-L1(123.98°±5.78°vs 103.89°±8.95°) was significantly increased after treatment (P<0.01). In control group, the values of U1-NA (mm:5.01±1.34 vs 9.12±2.13) and U1-SN(101.90°±3.97° vs 114.87°±7.69°)were significantly decreased after treat-ment. The values of U1-L1(126.01°±3.12°vs 112.98°±5.98°) and U6-PtPNS(mm:21.45±2.43 vs 18.36±2.19)were significant-ly increased after treatment (P<0.05). The value of U1-L1(19.48°±8.90° vs 13.01°±5.90°) was significantly changed in exper-imental group than that of control group, but the value of U6-PtPNS(mm:0.90±0.29 vs 3.78±0.12)was significantly changed in control than that of experimental group (P<0.01). Conclusion The maxillary protrusion malocclusion with micro-im-plant anchorage can be used as treatment for patients with maxillary protrusion that needs strong anchorage.
10.Analysis of influence factors on the elasticity measurement of focal liver lesions with real-time shear wave elastography

Objective To study the influence factors of real-time shear wave elastography(SWE) on the elasticity of focal liver lesions (FLLs) measurement.Methods The mean Young's modulus of 160 FLLs were measured by SWE.The relation between elasticity with different aspects of FLLs were analyzed including postures (supine and left lateral position),respiratory phases (end of inspiratory and expiratory),the probe position (rib and intercostal space) and different operators.Results There were significant differences between different postures and respiratory phases in the elasticity measurement of FLLs by SWE (P <0.05).There was no significant difference between robe position and different operators in the elasticity measurement of FLLs by SWE(P >0.05).Conclusions The postures and respiratory phases can impact the elasticity measurement of FLLs by SWE.
