Journal of Experimental Hematology 2009;17(4):986-989
This study was purposed to investigate the angiogenesis-promoting activities of human mesenchymal stem cells (hMSCs) modified by hepatocyte growth factor (HGF) and the underlying mechanisms. The hMSCs were transfected by recombinant adenoviral vector carrying human HGF gene and seeded onto the chicken chorioallantoic membrane. Three days later, the number of blood vessels was counted and their angiogenic response was compared with those of hMSCs of same generation, recombinant basic fibroblast growth factor (bFGF) and alpha-MEM as control. The expression levels of bFGF, VEGF, angiopoietin-1 and angiopoietin-2 were evaluated by RT-PCR assay. The results showed that gene-modified hMSCs exhibited greatest activity to promote angiogenesis while the angiogenic response was nearly same between groups treated by hMSCs and bFGF, all of which were significantly higher than that observed in control (p < 0.01). RT-PCR analysis revealed that hMSCs constitutively expressed multiple angiogenesis-associated growth factors and their levels seemed up-regulated by HGF gene transfer. It is concluded that HGF gene-modified hMSCs show a potent angiogenesis-promoting function and may be useful in the treatment of ischemic disorders.

OBJECTIVETo investigate the protective effect of Ophiopogonis polysaccharide (MDG-1) on isolated myocardium ischemia/reperfusion injury (IRI) and subcutaneous injection of isoprenaline induced acute myocardial ischemia.
METHODSIn ex vivo heart experiment: Langendorff guinea pigs were randomly divided into the IRI group, the fructose sodium diphosphate (FDP) group, treated with FDP 10(-6) - 10(-4) g/mL for positive control and the MDG groups treated with MDG-1 10(-6) - 10(-4) g/mL. The amplitude and frequency of cardiac contraction, coronary blood flow at different time points after ischemia reperfusion were measured. In integral animal experiments: acute myocardium ischemia model rats established by subcutaneous injection of isoprenaline were used, they were administered with MDG-1 in dosage of 10, 20 and 40 mg/kg respectively, and controlled with propranolol. Besides, a normal control group and an untreated model group for control were set up. The ST segment shift in ECG and lactate dehydrogenase (LDH) activity in serum were observed.
RESULTSEx vivo heart experiment showed that different doses of MDG-1 can increase IRI caused abnormal coronary blood flow, quickly resume the heart contraction and restrain the quickened heart rate (all P < 0.01). The integral animal experiment showed that oral administration of 40 mg/kg can reduce the increased activity of LDH in serum (P < 0.05) induced by isoprenaline, but almost had no effect on ST-segment shift in ECG.
CONCLUSIONMDG-1 can alleviate IRI isolated myocardium of guinea pigs, and oral administration of MDG-1 showed a definite protection on isoprenaline caused rats' myocardial ischemia damage.

OBJECTIVETo explore the alterations of apoptosis factor Bcl-2/Bax in the early Parkinson's disease (PD) rats and the protective effect of scorpion venom derived bioactive peptide.
METHODSHealthy male SD rats (180-220 g) were randomly divided into 4 groups (n = 10): early PD model group, sham operation group, scorpion venom derived bioactive peptide control group, scorpion venom derived bioactive peptide therapy group. 6-hydroxydopamine (6-OHDA) was used to prepare the early PD rat model. The immunohistochemistry was used to detect the expression of Bax and Bcl-2 and further explore the mechanism of anti-apoptosis regarding the neuroprotective effect of scorpion venom derived bioactive peptide.
RESULTSThe results indicated that compared with the control rats, the immunostaining of Bax in the brain increased significantly while that of Bcl-2 decreased significantly in the lesion side of 6-OHDA treated rats. Interestingly, scorpion venom derived bioactive peptide could attenuate the above abnormal changes.
CONCLUSIONUp-regulation of Bax and down-regulation of Bcl-2 could participate in the early stage of PD and the anti-apoptotic mechanism could be involved in the neuroprotective effect exerted by scorpion venom derived activity peptide regarding the dopaminergic neuron in the early stage.
