1.Effects of different concentrations of ox-LDL on the proliferation of rat theca cells and the expression of steroidogenesis related genes LXR-α and StAR

Objective · To investigate the effects of ox-LDL on the proliferation of rat theca cells and expression of LXR-α and StAR, two genes associated with androgen biosynthesis. Methods · The expression of LXR-α in the ovarian tissue of rats was determined by immunohistochemistry. Primary theca cells were isolated and collected from rat ovary and cultured in vitro. Furthermore, the theca cells were treated with 25, 50, 100, 150, 200, 300 and 400 mg/L ox-LDL, respectively. The variations in LXR-α mRNA were identified using real-time PCR. MTT assay was performed to detect cell viability. The expression of LXR-α and StAR was measured by Western blotting analysis. Results · The effect of ox-LDL on the proliferation of rat theca cells and the levels of LXR-α and StAR in theca cells was in a concentration-dependent manner. Following exposure to various concentration of ox-LDL for 24 h, the proliferation of theca cells was induced by low concentration of ox-LDL (25-150 mg/L), and 100 mg/L ox-LDL showed the most significant inducing effect. Moreover, the cell survival rate was diminished considerably following with ox-LDL concentration increasing, especially lowered by 400 mg/L ox-LDL. The mRNA level of LXR-α was increased with low concentration of ox-LDL (25-150 mg/L) and the impact of ox-LDL on the induced expression of LXR-α mRNA was considerably distinct at the concentration of 150 mg/L. On the other hand, the expression of LXR-α mRNA was reduced with high concentration of ox-LDL, and the impact of 400 mg/L ox-LDLwas substantially distinct. The protein expression levels of LXR-α and StAR were increased with 150 mg/L ox-LDL, but StAR protein level in 150 mg/L ox-LDL group revealed no significant difference when compared with control group. The expression of LXR-α and StAR protein was significantly inhibited with 400 mg/L ox-LDL in the rat theca cells. Conclusion · Low concentrations of ox-LDL can induce the proliferation of theca cells, and promote the expression of StAR and LXR-α. Whereas, high concentrations of ox-LDL can reduce the cell viability and inhibit the expression of StAR and LXR-α.
2.Diagnosis and surgical treatment of pulmonary sequestration:A report of 18 cases

[Abstract ] Objective Pulmonary sequestration (PS) is a congenital development malformation of the lungs .This study aimed to explore the diagnosis and surgical treatment of PS . Methods We retrospectively analyzed the clinical data on 18 PS pa-tients (8 males and 10 females, aged 20-68 [42.34 ±14.63] years) treated in our department from September 2009 to September 2014.Medical imaging manifested tumors in 11 cases and cystic lesion in the other 7.All the patients underwent surgical resection . Results The diseased pulmonary lobes were removed for the 17 cases of the intralobar type and local resection was performed for the 1 case of the extralobar type .Fourteen of the cases were confirmed to be PS , 3 misdiagnosed, and 1 case missed at diagnosis .The pa-tients were followed up for 8-18 months after treatment , all recovered without surgery-related complications . Conclusion Pulmona-ry sequestration is a rare congenital malformation of the lungs and the lack of specific clinical manifestations makes it easy to be misdi -agnosed.Surgery is the main strategy for its treatment .
3.Effects of the Pup-proteasome system on the growth of Mycobacterium smegmatis strains

Objective To study the effects of prokaryotic ubiquitin-like protein ( Pup)-proteasome system on the growth of Mycobacterium strains.Methods The genes encoding Pup ( pup gene) and protea-someβsubunit ( prcB gene) were respectively knocked out from Mycobacterium smegmatis ( M.sm) strains by homologous recombination.The growth and viability of the wild-type and mutant strains of M.sm were an-alyzed under normal culture condition and under hypoxia as well as anaerobic conditions.Results The pup and prcB genes were completely and precisely knocked out from M.sm strains and the mutant strains were named △SM-Pup and△SM-prcB, respectively.The△SM-Pup strains grew faster than the wild type ( WT) and△SM-prcB strains.No significantly differences in the growth of M.sm were found between the WT and△SM-prcB strains.Conclusion The Pup-proteasome system was involved in the growth of M.sm, espe-cially the pup gene.There was difference between pup and prcB genes in regulating the growth of M.sm.The functions and influences of Pup-proteasome system still need further investigation.
4.Long-term effects of pelvic autonomic nerve preservation on anorectal function after radical hysterecto-my

Objective To investigate the long?term effect of nerve?spring radical hysterectomy( NSRH) on anorectal function after radical hysterectomy. Methods Fifty?six cases of uterine cervical carcinoma patients who met the criteria were selected for the study and were randomly divided into RH group and NSRH group. Defecation functional and anorectal manometry were compared at 1 year after surgery. Results There were 2 patients were excluded both in the two groups, and 26 cases were included in the follow up of each group. 5.Influence of Environment Stimulation on Learning and Memory Ability and Hippocampal Pathology of Neonatal Rats with Hypoxic-Ischemic Brain Damage

Objective To observe the influence of environment stimulation on learning and memory ability and hippocampal pathology of neonatal rats with hypoxic-ischemic brain damage (HIBD).Methods The models of HIBD SD rats were established by the method of Rice, and were divided randomly into three groups: enriched environment stimulation group (EE), impoverished environment stimulation group (IE), and standard environment stimulation group (SE). The sham-operation rats were served as control group. Different environment stimulation was administrated to the rats since day 1 after HIBD.On the day 28,Morris water maze was used to evaluate the learning and memory ability. HE staining and nissl stain were employed to observe the pathological change and the number of neurons in hippocampus of rats.Results The learning and memory ability of EE group was significantly higher than that of SE group (P0.05), and the ability of SE group was higher than that of IE group ( P0.05). 6.Analysis of anxiety,depression, sleep quality and quality of life in patients with primary pruritus ani

Objective:
To learn anxiety, depression, sleep quality and quality of life among patients with primary pruritus ani, and to analyze their correlation with anal itching symptom, so as to provide insights into the prevention and treatment of primary pruritus ani.
Methods:
Patients with primary pruritus ani were selected from anorectal department of the First Affiliated Hospital of Zhejiang Chinese Medical University. The Visual Analogue Scale, pruritus frequency score, Hospital Anxiety and Depression Scale, Pittsburgh Sleep Quality Index and Dermatology Life Quality Index (Chinese version) were used in the survey. Spearman rank correlation analysis was employed to analyze the correlation between the degree and frequency of pruritus ani with, anxiety, depression, sleep quality and quality of life.
Results:
Sixty patients with primary pruritus ani were investigated, including of 7 males and 53 females, with an average age of (34.37±10.30) years. The patients with mild, moderate and severe pruritus accounted for 28.33%, 48.33% and 23.34%, respectively. The patients with pruritus frequency of grade 1, 2 and 3 accounted for 51.67%, 38.33% and 10.00%, respectively. The patients with anxiety, depression, and coexistence of anxiety and depression accounted for 18.33%, 13.33% and 10.00%, respectively. The patients with very good, good, average and poor sleep quality accounted for 15.00%, 46.66%, 30.00% and 8.34%, respectively. No, mild, moderate and severe impacts on quality of life among the patients accounted for 10.00%, 46.66%, 33.34% and 10.00%, respectively. The degree of pruritus was positively correlated with anxiety (rs=0.405, P=0.001), depression (rs=0.343, P=0.007), sleep quality (rs=0.293, P=0.037) and quality of life (rs=0.338, P=0.008). However, there was no significant correlation between the frequency of pruritus with the above factors (all P>0.05).
Conclusion
The degree of pruritus in patients with primary pruritus ani is related to anxiety, depression, sleep quality and quality of life.
7.Comparison analysis of clinical evaluation with hemodynamic monitor in the hemodynamic assessment of critically ill patients

Objective To investigate the accuracy of careful clinical evaluation in hemodynamic status and guidance of PiCCO monitor in clinical treatment.Methods A total of 96 hemodynamic unstable cases were evaluated prior to the insertion of the PiCCO catheter.The attending physician in charge of the patient was required to complete a questionnaire to predict the range of key hemodynamic variables for CI,GEDI,SVRI and EVLWI.Additionally,the attending was also asked to indicate a plan for therapy based on the predicted hemodynamic profile and decide if the predicted therapy plan was altered after the the first measurement of hemodynamic variables.Results The accurate prediction of hemodynamic variables was CI (55.2%),GEDI(60.4%),SVRI(63.5%) 和 EVLWI (78.1%),among which EVLWI had a higher accuracy(P < 0.05).49% doctors altered their planned therapy according to the result of the PiCCO information.Doctors had more difficulty in accurately predicting hemodynamic values in critical patients which APACHE Ⅱ scored 15 ~25 (42.3% vs 67.9% and 42.3 % vs 75.0%,x2 =4.755,5.231,P < 0.05).The prediction of patients with acute myocardial infarction was more accurate than those of without acute myocardial infarction,and less to alter the planned therapy(21.1% vs 55.8%,x2 =7.382,P =0.007).The patients of impaired oxygenation had less accurate predictions and less therapy alterations(32.3% vs 56.9%,x2 =5.110,P =0.024).Attending was able to predict the hemodynamic status more accurately(63.9% vs 40%,x2 =5.152,P =0.023) and alter the predicted therapy less(39.3% vs 65.7%,x2 =6.189,P =0.013) in patients who were enrolled later.Conclusions Clinical evaluation in hemodynamic status of critically ill patients had a lower accuracy,the information obtained by PiCCO often instruct clinical doctors to choose the optimal treatment.
8.UFLC-PDA fingerprint of Tibetan medicine Pterocephalus hookeir.

This study was aimed to establish an UFLC fingerprint of Tibetan medicine Pterocephalus hookeir samples from different habitats. UFLC-PDA was adopted to analyse 21 batches of P. hookeir samples from different habitats. The chromatographic condition was as follow: Agilent proshell 120 SB-C18 column (4.6 mm x 100 mm, 2.7 microm) eluted with the mobile phases of acetonitrile and 0.2% phosphoric acid water in gradient mode. The flow rate was 1.0 mL x min(-1), and the detection wavelength was set at 238 nm. The fingerprints of 21 batches P. hookeir were carried out by similarity comparation, and 15 chromatographic peaks were extracted as the common peaks of fingerprint, of which 5 peaks were identified as chlorogenic acid, loganin, sweroside, sylvestroside III, triplostoside A. The similarity degrees of 18 batchs of samples were above 0.9, and the other 3 batchs of samples were below 0.9. This is the first established fingerprint of P. hookeir by using UFLC-PDA. This method has good precision, stability and repeatability that it could provide basis for quality control and evaluation of P. hookeir.

9.Effect of stress-induced hyperglycemia on myocardial perfusion and prognosis in elderly patients with acute myocardial infarction undergoing primary percutaneous coronary intervention

Objective To investigate the effect of stress-induced hyperglycemia (SHG) on myocardial perfusion and clinical prognosis in elderly patients with acute myocardial infarction (AMI) who underwent primary percutaneous coronary intervention (PCI).Methods 348 elderly patients with first-time occurrence of acute ST-elevation myocardial infarction (STEMI) who underwent primary PCI within 12 hours from June 2008 to June 2010 were enrolled and followed up.All patients were divided into three groups according to serum glucose (SG) on admission:normal group (SG< 7.0 mmol/L,n=112);SG elevation group (7.0 mmol/L≤SG≤11.1 mmol/L,n=128) and (steady high blood glucose) SHG group (SG>11.1 mmol/L,n=108).Myocardial perfusion indexes,including ST segment resolution (STR),TIMI myocardial perfusion grade (TMPG),peak value of creatine kinase CK-MB,left ventricular ejection (LVEF),and major adverse cardiac events (MACE) of patients in three groups,were measured and compared after emergency PCI.Results The blood glucose levels were increased,ST-elevation 2 h after PCI were well declined,the percentages of patients with TMPG 2-3 were decreased and peak values of CK-MB were increased in the three groups (all P<0.01).After 12 months of follow-up,Kaplan-Meier survival analysis showed that patients of three groups had significantly different cumulative non-events survival rates [89.3% (100/112) vs.85.9% (110/128),76.3% (83/108),P<0.05].Multivariate Cox regression analysis showed that steady high blood glucose were the independent predictor for the occurrence of MACE in patients undergoing PCI after adjusting for age and gender,and the risk of MACE was increased by 5.811 folds in SHG group as compared with normal group (P<0.01).Conclusions Stress induced hyperglycemia in elderly patients with STEMI can decrease myocardial perfusion level after primary PCI,which will lead to high incidence of MACE.
10.Effect of PM2.5 on sympathetic innervation by stimulating the secretion of cardiac nerve growth factor in healthy rat hearts

ObjectiveTo assess the effect of PM2.5 on sympathetic innervation and the relationship with cardiac nerve growth factor in healthy rat hearts,identify the ability to the sympathetic nerve reconstruction,and explore the possible arrhythogenic mechanism of PM2.5.MethodsForty healthy SD rats were instilled into trachea with two different solutions twice per week for four weeks:control group with saline and experimental group with PM2.5 25 mg/kg.Then these rats were killed,and biatrial appendages,two ventricular anterior walls were left.Immune cytochemical staining of cardiac nerves was performed using anti-tyrosine hydroxylase antibodies and cardiac nerve growth factor was detected by western blotting.ResuitsCompared with the control group,both the density of sympathetic nerves and the expression of cardiac NGF protein in the experimental group were much higher in the left ventricular anterior wall ( P<0.01 and P<0.05) and in the right ventricular anterior wall ( P<0.01 and P<0.05).ConclusionsPM2.5 can induce regional sympathetic hyperinnervation in both ventricular anterior walls in healthy rats by stimulating the NGF protein secretion,and the sympathetic nerve reconstruction effect of PM2.5 was confirmed.
