1.Microwave ablation for the treatment of hepatic cavernous hemangiomas:its serious complications and their prevention and treatment

Objective To analyze the complications caused by microwave ablation for hepatic cavernous hemangiomas, and to discuss the prevention and treatment of the complications. Methods During the period from July 2009 to May 2011 at authors’ hospital, a total of 34 times of microwave ablation procedure were carried out in 30 patients with hepatic cavernous hemangiomas. The microwave ablation-related complications were recorded and the prognosis was evaluated. Results Serious complications occurred in 2 patients (6.7%), including acute hemolysis leading to acute renal failure (n = 1) and abdominal wall abscess (n = 1). Postoperative fever was seen in 18 cases (60%), transient hemoglobinuria in 4 cases (13.3%), nausea with vomiting in 5 cases (16.7%), marked pain at the surgical area in 8 cases (26.7%), right pleural effusion in 4 cases (13.3%), hepatic dysfunction in 24 cases (80%) and leukocytosis in 11 cases (36.7%). No treatment-related death occurred. Conclusion For the treatment of hepatic cavernous hemangiomas, microwave ablation is minimally invasive with higher safety and satisfactory effect, although this technique carries somewhat higher risks when the hemangioma is larger or the location of the hemangioma is anatomically complicated. Some of these complications can be early detected and proper management should be actively adopted in order to avoid as well as to minimize the adverse results.
3.The mechanism study of Chinese traditional medicine mixture on protection of vascular endothelial cell from apoptosis in cerebral ischemia-reperfusion injury

Objective To investigate the mechanisms of Chinese traditional medicine mixture protection of vascular endothelial cell from apoptosis in cerebral ischemia-reperfusion injury. Methods Healthy, clean SD rats were randomly divided into 4 groups: Sham opera-tion group (SOG), cerebral ischemia reperfusion injury group (IRG), cerebral ischemia preconditioning group (IPG) and Chinese tradi-tional medicine mixture preconditioning group (CPG). Furthermore, IRG, IPG and NPC were divided into 4 sub-groups: 1 d, 7d, 14d, 21d subgroup, according to the different time point since ischemia-reperfusion took place. And in CPG, Naotai formula extract was used. Cere-bral vascular endothelial cells of rats were removed and Hoechst 33258 staining and the DNA gradient bands were used to detect the apoptosis of these cells. Then the influence of Naotai formula extract on caspase-3, 8 and 9 activation, and Bid lysis was examined by Western-blot, and the mechanisms of Chinese traditional medicine mixture protection of vascular endothelial cell were investigated from apoptosis signal pathway. Result Naotai formula extract can inhibit the apoptosis of endothelial cells in ischemia-reperfusion injury, and it also can inhibit the activation of caspase-3, 8 and 9, thus inhibit the lysis of Bid into tBid. Conclusion Naomi formula extract inhibit the apoptosis of endo-thelial ceils in ischemia-reperfusion injury via apoptosis signal pathway.
4.The study on the total oxidative and antioxidant status and lipid levels in patients with COPD

Objective To explore and evaluate the total oxidative and antioxidant status and lipid levels in patients with COPD. Methods Measurement and comparative analysis were preformed between 51 cases of COPD patients and 45 healthy people in serum TOS,TAS,IMA concentration,blood lipid,and the concentration of ox-LDL. Results In the COPD group,serum IMA concentration was significantly higher than those in the control group(0.94 ± 0.31 vs. 0.62 ± 0.25,P<0.001). The serum TOS was higher than that in the control group(6.51 ± 4.85 vs. 4.53 ± 3.81,P<0.05). The serum ox-LDL concentration in COPD group increased significantly compared with the control group(5.91 ± 3.61 vs. 4.51 ± 2.05,P<0.05). The serum HDL concentration in the COPD group was significantly lower than that in the control group(36.49 ± 9.60 vs. 44.19 ± 10.14,P<0.001). There was no sig-nificant difference in TAS,triglyceride and low LDL between the two groups. Conclusions The serum levels of TOS,ox-LDL and IMA are significantly increased in COPD patients compared with the control group,while HDL-C concentration is significantly decreased. Increase of IMA and ox-LDL in serum of patients with COPD may be relat-ed to imbalance of TOS and TAS due to chronic inflammation and oxidative stress.
5.Experimental study on inhibitory effects of diallyl sulfide on growth and invasion of human osteosarcoma MG-63 cells.

The inhibitory effects of diallyl sulfide (DAS) derived from allicin on in vitro and in vivo proliferation of human osteosarcoma MG-63 cells and the action mechanism, and the influence of DAS on invasive capability of MG-63 cells were investigated in order to search for the novel medicines for osteosarcoma. In the in vitro experiment, MG-63 cells were treated with different concentrations of DSA, and the morphological changes of MG-63 cells were observed under an inverted phase microscope. MTT method was used to assay the proliferation of MG-63 cells. Semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) was used to detect the VEGF mRNA expression level in MG-63 cells. By using Transwell invasion assay, the influence of DAS on invasive ability of MG-63 cells was tested. In the in vivo experiment, the nude mice MG-63 cells tumor-bearing model was established, and different concentrations of DAS were injected beside the tumor. Twenty-one days after treatment, the mice were killed, the tumor size and tumor inhibition rate were calculated. The microvessel density (MVD) was determined by using immunohistochemistry. In the in vitro experiment, different concentrations of DAS could obviously inhibit proliferation of MG-63 cells in a time- and concentration-dependent manner. RT-PCR revealed that the expression levels of VEGF mRNA in DSA groups (different concentrations) were significant reduced as compared with those in control group (all P<0.05). Transwell invasion assay indicated that in 20 and 40 μg/mL DAS groups, the number of migratory cells was 91.4±8.3 and 81.8±7.4 respectively, which was significantly declined as compared with that in control group (150.4±14.7, both P<0.05). In the in vivo experiment, DAS could significantly suppress the growth of MG-63 tumor-bearing tissue. Immunohistochemistry demonstrated that different concentrations (20 and 40 μg/mL) of DAS could significantly decrease MVD of MG-63 tumor-bearing tissue (all P<0.05). It was suggested that DAS could inhibit the growth of MG-63 cells probably by suppressing the expression of VEGF mRNA.
6.Detection and correlation analysis of sperm apoptosis rate and level of reactive oxygen species in seminal plasma among infertile males

Objective To detect the sperm apoptosis rate and level of reactive oxygen species (ROS) in seminal plasma and explore their correlation among infertile males. Methods Ninety-two inferitile males were divided into varicocele (VC) group (n=32), leukocytospermia group(n=30) and the other cause group (n=30), and another 24 in vitro fertilization sperm samples were sereved as controls. The routine sperm parameters including seminal pH, sperm viability and sperm density were examined by computer assisted sperm analysis, the sperm apoptosis rate was asseseed using Annexin V/PI staining, and the ROS level in seminal plasma was detected by TBA method. The differences in seminal parameters between three infertile groups and control group were compared, and the correlation of sperm apoptosis rate with level of ROS in seminal plasma was explored in each group. Results The sperm viability of three infertile groups was significantly lower than that of control group (P<0.01). The sperm apoptosis rates and levels of ROS in seminal plasma in VC group and leukocytospermia group were significantly higher than those in control group (P < 0.05 or P < 0.01). The sperm apoptosis rate was positively correlated with the level of ROS in seminal plasma in leukocytospermia group(r=0. 573, P < 0.05). Conclusion The increased sperm apoptosis rate and level of seminal plasma ROS may be related to the infertility of patients with VC and leukocytospermia. The increased level of seminal plasma ROS may be one of the causes of increased sperm apoptosis rate in patients with leukocytospermia.
9.Gastrointestinal absorption and pharmacodynamic research of insulin-loaded poly(lactic acid) nanoparticles for oral administration

AIM To investigate the effect of DO C(deoxycholic acid) on the absorption of INS-PLA-NP[insulin-loaded poly(lactic acid) nanoparticles] in different sites of gastrointestinal tracts. METHODS After INS-PLA-NP that contained or did not contain DOC was adminitered to different sites in gastrointestianl tracts(stomach, small intestine and colon)of normal rats, the hypoglycemic effect was observed. RESULTS The hypoglycemic effect did not exist after intragastric administration of INS-PLA-NP whether or not DOC was added. The alleviatory hypoglycemic effect was evident after intraintestinal absorption of INS-PLA-NP. After DOC was added, the absorption of INS-PLA-NP was accelerated obviously and the hypoglycemic effect was strengthened significantly. Glucose levels hardly changed after INS-PLA-NP was administered to colon. With the use of DOC, a little hypoglycemic effect appeared. CONCLUSIONS The absorption of INS-PLA-NP in small intestine was accelerated and enhanced by DOC. DOC could be used as absorption enhancer of INS-NP in the future.
10.Chemical constituents of Illicium micranthum

AIM: To study the chemical constituents of Illicium micranthum. METHODS: Various column chromatographic techniques were applied to isolation and purification and the structures were elucidated by NMR and MS. RESULTS: Seven compounds were isolated from the part of ethyl acetate and identified as Neomajucin(1),2-oxo-3,4-dehyhroxyneomajucin(2),Transcinnamic acid(3),3,3′,4′,5,7-pentahydroxyflavan(4),Shikimic acid(5),Kaempferol(6),Kaempferol-3-O-?-L-rhamnopyranosyl-(1→6)-?-D-glucopyranoside(7). CONCLUSION: The above compounds are obtained from this plant for the first time.
