Objective Retinoic acid receptor(RAR?) selective antagonist,Ro41-5253(Ro) was used in the counteract test,to confirm the role of RAR? in enhancement of IgM synthesis in cord blood lymphocytes(CBLs) by retinoic acid(RA).Methods CBLs were cultured in vitro and stimulated with or without RAR? agonist RA and(or) antagonist Ro.The cells were harvested at 24 hours and 48 hours of culture time to measure the levels of gene expression of RARa,IL-4 and IL-6.ELISA was used to measure the concentration of IgM in the supernatant of 5 days culture.Results Ro could inhibit the promotion of RA on IgM synthesis in CBLs.RA could up-(regulate) RAR? gene expression,which could be restrained by Ro.Ro also could counteract the up-regulation of RA on IL-4 and IL-6 gene expression.Conclusion The effect of RA on IgM synthesis in CBLs is modulated by RAR?.

AIM: To study the physiological function changes of the tears film after rebuilding ocular surface with corneal stem cells, and to discuss the validity and the estimate system of rebuilding ocular surface with the corneal stem cells. METHODS: The male New Zealand rabbits were used to establish the alkali burning model in the right eye. The corneal stem cells of the left eye were cultured on the amniotic membrane in vivo, and then transplanted to the right eye. Furthermore, the physiological function changes of the tear film were examined. RESULTS: Compared to the before alkali burning, the ocular surface cell morphology was similar after rebuilding ocular surface with the corneal stem cells, which were cultured on the amniotic membrane in vivo; The tear film breakup time test showed the a remarkable difference between after and before the alkali burning, but the cell modality after rebuilding had no remarkable difference compared to that before the alkali burning. CONCLUSIONS: It's an effective method to rebuild the ocular surface with the corneal stem cells in vivo, the cell framework and the physiological function of the tears film recover well after rebuilding. It may be a validity estimate system of rebuilding ocular surface to analyze framework and configuration of the ocular surface and test the tear film breakup time.

Objective: To investigate the effects of inhibition of microRNA-221 (miR-221) expression on the proliferation and apoptosis of bladder cancer cells. Methods: Has-miR-221 inhibitor and has-miR-221 inhibitor negative control were synthesized, and then transfected into bladder cancer T24 and J82 cells. The transfection efficiency was observed under a fluorescence microscope 5 h after transfection. The expression levels of miR-221 in T24 and J82 cells were detected by real-time fluorescence quantitative PCR at 24, 48 and 72 h after transfection; the proliferation of T24 and J82 cells was also detected by MTT assay. The expression levels of p53 upregulated modulator of apoptosis (PUMA), Bax and Bcl-2 mRNAs and proteins in T24 and J82 cells 48 h after transfection were measured by RT-PCR and Western blotting, respectively; the apoptosis of T24 and J82 cells was determined by flow cytometry (FCM) and acridine orange (AO)-ethidium bromide (EB) staining. Results: The efficiencies of has-miR-221 inhibitor transfection into T24 and J82 cells were 80% and 90%, respectively. The expression levels of miR-221 in T24 and J82 cells after transfection with has-miR-221 inhibitor were lower than those in the negative control group and the blank control group (only adding liposome) (P < 0.05). The proliferative inhibition rates of T24 and J82 cells after transfection with has-miR-221 inhibitor were higher than those in the negative control group and the blank control group (P < 0.05), and this effect was in a time-dependent manner. The expression levels of PUMA and Bax mRNAs and proteins in T24 and J82 cells after transfection with has-miR-221 inhibitor were higher than those in the negative control group and the blank control group (P < 0.05), and the expression levels of Bcl-2 mRNA and protein were opposite (P < 0.05). The apoptosis rates of T24 and J82 cells after transfection with has-miR-221 inhibitor were higher than those in the negative control group and the blank control group (P < 0.05). Conclusion: Inhibition of miR-221 expression can suppress the proliferation of bladder cancer cells and induce apoptosis.

Objective To investigate the association of the major histocompatibility complex class Ⅰ chain-related antigens A (MICA)-129 gene polymorphism and soluble MICA (sMICA) levels with ulcerative colitis (UC) in Hubei Han nationality. Methods The genetic polymorphism of MICA-129 was examined using a polymerase chain reaction-sequence based test (PCR-SBT) in 256 UC patients and 460 healthy controls. From the above subjects, 80 patients and 90 healthy individuals were randomly selected for determining serum sMICA concentrations by ELISA. Results The frequencies of variant allele (G) and genotype (GG) in MICA-129 gene were significantly higher in the UC patients than in the controls(76. 8%vs 72. 2%, P =0. 060; 55.9% vs 46. 3% ,P =0. 016). Serum sMICA levels were significantly elevated in the patients compared to the controls[(576. 47 ±279. 02) ng/L vs( 182. 17 ±73. 11 ) ng/L,P ＜0. 001]. In addition, the sMICA levels were higher in the patients carrying MICA-129 GG genotypes than in those carrying ( GA + AA) genotypes [( 638. 87 ± 347. 15 ) ng/L vs ( 507. 51 ± 152. 87 ) ng/L, P = 0. 035].Conclusions The genetic polymorphism of MICA-129 and sMICA levels are correlated with the UC patients in Hubei Han nationality. Our findings demonstrate that MICA-129 gene may contribute to the pathogenesis of UC.

· AIM: To study the in situ relative intraocular position of the iris-claw phakic intraocular lens (PIOL)for high myopia using an anterior chamber optical coherence tomography (AC OCT)prototype.· METHODS: Six PIOLs (11.50 to 22.00D lens powers) were implanted in phakic myopic eyes. Using AC OCT, tomography was taken in the anterior chamber to measure the preoperative anterior chamber depth, postoperative distance between the PIOL and the corneal endothelium (endothelial-optic distance), and the postoperative distance between the PIOL and the crystalline lens.· RESULTS: Preoperative anterior chamber depth ranged from 3.27 to 3.91 mm and the postoperative endothelial-optic distance measured 2,07 to 2,24 mm. The distance between the crystalline lens and the posterior surface of the IOL ranged from 0.82 to 1.32 mm. Several tomography revealed the position of the PIOL on the iris, The pigment layer of the iris did not seem to be disturbed by the presence of the PIOL.· CONCLUSION, The original anterior chamber depths were reduced by 36,1% to 44.6% after implantation. This study of 6 eyes revealed that tomography taken by AC OCT are useful in verifying the intraocular position of the PIOL within the anterior chamber. Adequate space was maintained between the iris-fixated phakic intraocular lens and the corneal endothelium, angle, and crystalline lens.
