2.The research of peripheral blood regulatory T cell in patients with idiopathic thrombocytopenic purpura

Objective To investigate the change of peripheral blood CD4+CD25+ regulatory T cells (Tr) in patients with idiopathic thrombocytopenic purpura (ITP), and to analyze its role in the pathogenesis of ITP.Methods Anticoagulated venous blood was collected from ITP patients (ITP group,n=35) and healthy controls (healthy control group,n=35). T lymphocytes were isolated and purified with human CD3+ T cell enrichment columm. The percentage of peripheral blood CD4+CD25+ regulatory T cells was detected by immunofluorescence staining (PE-anti-CD4 monoclone antibody and FITC-CD25 monoclone antibody) and bicolor flow cytometry by CD25/CD4 gating.Results The number and constituent ratio of CD4+CD25+T cell were significantly lower in ITP group than those of healthy control group (P<0.05).Conclusion There is peripheral blood celluar immunological function disorder in ITP patients, and decrease of CD4+CD25+ T cell population may be involved in the pathogenesis of ITP.
3.The relationship between C-terminal telopeptides of type I collagen in GCF and periodental indexes

Objective:To detect the correlativity between clinical periodental indexes and C-terminal telopeptides of type I collagen (CTX)concentration in gingival crevicular fluid(GCF).Methods:77 teeth with periodontitis in 60 patients from the international Bethune peace hospital were included.The concentration of CTX in GCF was detected by enzyme-linked immunosorbent assay.The plague indexe(PL),gingival indexe(GI),probing depth(PD)and attachment loss (AL)level were recorded at clinic.The correla-tion of CTX concentration and clinic indexes was analysed by Kruskal-Wallis test using SPSS 1 1 .0 software.Results:The Spearman correlation coefficient of PL,GI,PD and AL with CTX was 0.386(P <0.01 ),0.366(P <0.05),0.382(P <0.05)and 0.31 4(P <0.05)respectively.Conclusion:PL,GI,PD and AL are positively correlated with the concentration of CTX in GCF(P <0.05).
4.Clinical evaluation of Nickel-Titanium rotary instruments in preparation of root canals

Objective To study and assess clinical application of two Nickel-Titanium(NiTi)rotary instru- ments,namely ProTaper and Hero 642,in preparation of root canals.Methods 125 teeth were divided into three groups and respectively instrumented by stainless K-files,ProTaper or Hero 642 rotary instruments.All teeth were obturated with lateral condensation method.The efficiency of preparation and obturation was analyzed with radio- graphs.Results NiTi rotary instruments were better in keeping the curvature and flow of curved canals than stain- less K files.There was no transportation,apical blockage and ledge in NiTi groups.The operative time was shorter and endodontic flare-up seldom occurred in NiTi groups.Conclusion The NiTi rotary instrumentation technique could be used to prepare curved root canals effectively and quickly.The future use of NiTi engine-driven rotary in- strument appeared to be promising.
5.Symptomatic middle cerebral artery stenosis:stroke recurrence,clinical prognosis and vascular change

objective To investigate the recurrence of stroke,clinical prognosis and vascular changes in patients with ischemic stroke due to middle cerebral artery stenosis. Methods The ischemic stroke patients with symptomatic middle cerebral artery stenosis were enrolled continuously and followed up prospectively for six months. The recurrence of ipsilateral stroke,clinical prognosis and dynamic changes of vessels were analyzed. Results Eighty patients were included,and 20.0% of the patients(16 cases)presented with recurrence of ipsilateral ischemic stroke and 56 cases (70.0%)with a good outcome(modified Rankin scale[mRS]≤1)during the 6 months follow-up;38.6% patients (27 cases) presented with significant vascular changes with progression in 12 cases (17.1%)and regression in 15 cases (21.4%). Conclusion The patients with simple symptomatic middle cerebral artery stenosis have an high rate recurrence of ipsilateral stroke but have good prognosis;Lesioned artery of the majority of patients in the short period after stroke was stable,but vascular stenosis in some patients could appear progression or remission.
6.Effect of coenzyme Q10 on the expression of interleukin-17 and interleukin-23 in gingival tissue of diabetic rats with periodontitis

BACKGROUND:Coenzyme Q10 participates in the electron transport of respiratory chain and possesses antioxidant, anti-apoptotic and anti-inflammatory properties. It has achieved good outcomes in cardiovascular disease, diabetes and cancer. Coenzyme Q10 may also have a certain application value in the fields of diabetes and periodontitis. OBJECTIVE:To observe the effect of coenzyme Q10 on the expression of interleukin-17 and interleukin-23 in gingival tissue of type 2 diabetic rats with periodontitis.METHODS:Forty-eight healthy male Wistar rats were randomly divided into 3 groups: control, periodontitis+ diabetes+physiological saline and periodontitis+diabetes+coenzyme Q10. Rats in the control group were fed with normal diet and water. Rats in the periodontitis+diabetes+physiological saline and periodontitis+diabetes+ coenzyme Q10 groups were subjected to induction of periodontitis using the method of silk ligation and type 2 diabetes by feeding a high-fat and high-sugar diet and intraperitoneal injection of streptozotocin. After successful modeling, rats in the periodontitis+diabetes+coenzyme Q10 group were intragastricaly administered coenzyme Q10 for 8 successive weeks. Rats in the periodontitis+diabetes+physiological saline group were administered equal amount of physiological saline. At the end of 2nd, 4th and 8th weeks after drug administration, four rats were randomly selected and sacrificed. The expression levels of interleukin-17 and interleukin-23 in gingival tissue were detected by immunohistochemistry. RESULTS AND CONCLUSION: At the end of 8th week, interleukin-17- and interleukin-23-positive expression in the periodontitis+diabetes+physiological saline group was significantly higher than that in the periodontitis+ diabetes+coenzyme Q10 group (P < 0.05). Coenzyme Q10 can reduce the expression levels of interleukin-17 and interleukin-23 in gingival tissue of type 2 diabetic rats with periodontitis, and aleviate periodontal tissue inflammation of type 2 diabetic rats with periodontitis.
7.Correlation of plasma heme oxygenase-1 level and type 2 diabetes mellitus

Objective To investigate the correlation of plasma heme oxygenase-1 (HO-1) level and type 2 diabetes mellitus (T2DM). Methods The outpatient with type 2 diabetes mellitus (T2DM) undergoing oral glucose tolerance test and healthy individuals with physical examination were divided into T2DM group and healthy control group, the differences between the two groups in HO-1 and methane dicarboxylic aldehyde (MDA), mean fluorescence intensity (MFI), homeostasis model assessment of insulin resistance (HOMA-IR) and fasting blood glucose (FBG) were comparatively analysed , and analyzed the correlations between HO-1 and reactive oxygen species (ROS) MFI, MDA, HOMA-IR and FBG. Results The type 2 diabetes group in MDA and MFI, the expression rate of HO-1 were higher than those of control group (P<0.05), correlation analysis of expression of HO-1 was positively correlated with MFI, MDA (r=0.489, 0.763, P<0.05) in the T2DM group, HO-1, HOMA-IR and FBG were significantly higher than the healthy control group, the difference was statistically significant(P<0.05). The expression of HO-1 and HOMA-IR and FPG levels were positively correlated in the T2DM group (r=0.271, 0.426, P <0.05). Conclusion T2DM patients with hyperglycemia and oxidative stress, plasma HO-1 expression is significantly increased, HO-1 is related to oxidative stress, insulin resistance and hyperglycaemia, which has certain value on clinical assessment of T2DM and therapeutic efficacy.
9.A genetic adaptive pattern-low hemoglobin concentration in the Himalayan highlanders.

Mean hemoglobin (Hb) concentration of about 3 500 subjects derived from 17 studies of Himalayan highlanders (Tibetans, Sherpas, and Ladakhis) was compared with lowlanders (Chinese Han, Indian Tamils) lived in the Himalayas, and European climbers during Everest expeditions as well as Andean natives. The results found that Hb concentration in Himalayan highlanders was systemically lower than those reported for Andean natives and lowland immigrants. These comparative data demonstrated that a healthy native population may successfully reside at high altitude without a significant elevation in Hb, and the lower Hb levels of Himalayan highlanders than those of migrated lowlanders and Andean natives are an example of favourable adaptation over the generations. In addition, excessive polycythemia has frequently been used as a marker of chronic mountain sickness (CMS). Altitude populations who have a higher Hb concentration also have a higher incidence of CMS. The low Hb in Himalayans suggested as showing adaptation over many generations in Tibetan stock. Recent work in Tibet, suggested that Tibetans there may have adapted to high altitude as a result of evolutionary pressure selecting for genes which give an advantage at altitude. All of the population genomic and statistical analysis indicated that EPAS1 and EGLN1 are mostly likely responsible for high altitude adaptation and closely related to low Hb concentration in Tibetans. These data supported the hypothesis that Himalayan highlanders have evolved a genetically different erythropoietic response to chronic hypoxia by virtue of their much longer exposure to high altitude.

10.Optimization of extraction technology for salidroside, tyrosol, crenulatin and gallic acid in Rhodiolae Crenulatae Radix et Rhizoma with orthogonal test.

The extracting technology of salidroside, tyrosol, crenulatin and gallic acid from Rhodiolae Crenulatae Radix et Rhizoma was optimized. With extraction rate of salidroside, tyrosol, crenulatin and gallic acid as indexes, orthogonal test was used to evaluate effect of 4 factors on extracting technology, including concentration of solvent, the dosage of solvent, duration of extraction, and frequency of extraction. The results showed that, the best extracting technology was to extract in 70% alcohol with 8 times the weight of herbal medicine for 2 times, with 3 hours once. High extraction rate of salidroside, tyrosol, crenulatin and gallic acid were obtained with the present technology. The extracting technology was stable and feasible with high extraction rate of four compounds from Rhodiolae Crenulatae Radix et Rhizoma, it was suitable for industrial production.
