1.Simultaneous Ditermination of Three Indicators in Chrysanthemi Flos by Quantitative Analysis of Multi-Components by Single Marker

Objective To establish a method for determination of chlorogenic acid, galuteolin and 3,5-O-dicaffeoyl quinic acid in chrysanthemums by quantitative analysis of multi-components by single marker ( QAMS ) . Methods Using chlorogenic acid as the internal standard substance, the relative correction factors ( RCFs ) of galuteolin and 3, 5-O-dicaffeoyl quinic acid were determined by HPLC.The contents of chlorogenic acid, galuteolin and 3,5-dicaffeoyl quinic acid in 12 samples of chrysanthemums were authentically determined by the external standard method, so as to to verify the accuracy of the QAMS method. Results The average relative error of contents of galuteolin and 3,5-O-dicaffeoyl quinic acid was 2.75% and 1.33%by the external standard method and QAMS, respectively. Conclusion The established RCFs have good reproducibility.In case of lack of standard substances, the QAMS method with chlorogenic acid as an internal standard for determination of 3 indicators in chrysanthemum is feasible.
2. Silencing PI3K by siRNA activated FoxO3a gene and inhibited the growth of xenografted breast carcinoma in nude mice

Objective: To investigate the effect of silencing PI3K by small interfering RNA (siRNA) on the expression of FOXO3a in human breast cancer MCF-7 cells and the growth of xenografted breast carcinoma in nude mice. Methods: Plasmids of siRNA for PI3K gene (Psilencer1.0-U6-PI3K) were constructed and xenografted tumor model was established in nude mice by subcutaneous inoculation of MCF-7 cells. Twenty-seven nude mice with xenografted tumor were divided randomly into 3 groups. Group 1 was intratumorally administered by Psilencer1.0-U6-PI3K plasmid. Group 2 and group 3 were intratumorally given Psilencer10-U6 plasmid and physiological saline (group 1), respectively. The tumor volume and survival time of nude mice were recorded. The expressions of PI3K and FOXO3a was measured by Western blotting. Results: The tumor size was significantly decreased and the average survival time was significantly enlongated in Psilencer1.0-U6-PI3K-treated group (P <0.01). Western blotting analysis showed that the protein expression of PI3K in xenografted tumor was markedly decreased and FOXO3a was significantly elevated compared with control groups (P <0.05). Conclusion: Psilencer1.0-U6-PI3K markedly silenced the expression of PI3K gene, up-regulated the expression of tumor suppressor gene FOXO3a, inhibited the growth of xenografted tumor, finally prolonged the survival time of nude mice bearing human breast carcinoma.
3.IMPROVEMENT OF NIGERICIN PRODUCTION IN STREPTOMYCES HYGROSCOPICUS NND-52 BY COMBINATION MUTANT

Based on the lethal rate, positive and negative mutation rate, the optimal doses of UV, HNO2, UV + AO treating S. hygroscopicus NND-52-29 were determined. The mutant strain A19 that was obtained showed the derepres-sive property to glucose and the nigericin yield of A19 could be increased by 128%. When it was subcultured combined with natural screening, its stability was rather satisfied.
4.The Clinic Study of Synthesize Imaging Diagnosis in Fibrocystic Disease of Breast

Objective To analyse the necessity and feasiblity about the diagnosis and classification of fibrocystic disease of breast,through using synthesize imaging methods , to increase the disease 's diagnosis rate. Methods Using imaging checks (molybdenum photography,ultrasonography and infra-red scanner) for 600 breast's cases and contrast to pathogenesis if they were done operations. Results 143cases were normal,166 cases got other disease of breast,291 cases were fibrocystic disease of breast(48.1%) ,155 cases of them were performed operations and pathogenesis. the diagnosis similar rate between performed operations check and pathogcnesis was 83.2%. Conclusion The sensitivity of three kinds imaging check method is different each other,but one method can give some help each other. The synthesize imaging check can increase the diagnosis rate and avoid mis-diagnosis and doing needless operations.

6.Research and application of urethral sling placement on neurogenic incontinence

BACKGROUND:The patients with neurogenic urinary incontinence could be cured to restore their urinary storage function through the implant of the suburethral sling made of appropriate materials in a reasonable way. Meanwhile the choice of materials and the safe and effective control of tension are crucial factors. OBJECTIVE:To review the implanted methods, effectiveness and safety of sling made of different materials in the surgical treatment of neurogenic stress urinary incontinence. METHODS:The PubMed database, English database, CNKI database and related English books were retrieved with computer from January 1986 to January 2012 for the articles on the urethral sling material, urethral sling placement methods, efficacy and safety. The key words were“urethral sling, neurogenic, urinary incontinence”in English and Chinese. Final y, 47 articles were included for review according to the inclusion and exclusion criteria. RESULTS AND CONCLUSION:Urethral sling is a safe, handy, and micro-invasive operation, by using autologous fascia or various synthetic materials to implant into the body. The mechanism of neurogenic urinary incontinence is mainly related with the change of bladder function or the insufficiency of urethral sphincter function. However, it is stil a big problem for us to assure the postoperative ability of such patients to store and control urine because of their complicated conditions.
7.Retroperitoneal laparoscopic ureterolithotomy for impacted ureteral calculi after radical cystectomy and ileal conduit (report of 5 cases)

Objective To evaluate the clinical efficacy and security of retroperitoneal laparoscopic ureterolithotomy(RLUL) for impacted ureteral calculi after radical cystectomy and ileal conduit.Methods 5 patients with unilateral impacted ureteral calculi after radical cystectomy and ileal conduit received RLUL were selected,and a retrospective study was performed for manner,duration of surgery,complications and length of stay and other indicators,and the treatment effect was evaluated.Results All procedures were successful and the mean operation time was (82.0±27.7)min,the extubation time was (5.4±1.1)d,hospitalization time was (10.0±2.9)d.Postoperative follow up for 6-48 months,there were no obvious complications.Conclusion The RLUL showed satisfactory availability and security for management ureteral calculi after radical cystectomy and ileal conduit.
10.Changes of fiver and kidney function and morphology of the renal artery of the metabolic syndrome rats

Objective Toinvestigate the changes of liver and kidney function and morphology of the renal artery of the metabolic syndrome (MS) rats.Methods The rats were respectively fed with ordinary or high-fructose diet for 10 weeks,and then took the blood and anatomical observation,to explore the changes of liver and kidney function and morphology of the renal artery of the rats.Results The serum levels of ALT,AST,UA,BUN,CR in the control group and MS group were gradually increased with the feeding time.But those in MS rats were significantly increased than the control rats.MS rats showed dysfunction of liver and kidney,which similar to the clinical MS cases.Compared with the control group,the renal artery of MS rats had the same pathological changes as MS cases,presented with intima uneven thickening and structural disorder,medial smooth muscle cells hyperplasia to intimal,the internal elastic membrane with different degrees of damage and fracture,the film elastic fibers thickening and structural disorder,smooth muscle cell proliferation apparently,medial obvious thickening.Conclusion MS rats have mild dysfunction of liver and kidney and abnormal changes in intima and tunica media of renal artery.
