1.Intra-arterial recanalization for acute ischemic stroke

2. Optimal extraction of indirubin from Isatidis Folium based on Plackett-Burman design combined with central composite design-response surface methodology

Objective: To investigate the significance of each influencing factor and optimize the process of extracting indirubin from Isatidis Folium by Plackett-Burman design combined with central composite design-response surface methodology (CCD-RSM). Methods: Plackett-Burman experimental design was used to screen the main influencing factors, and CCD-RSM was used to optimize the extraction process of indirubin. With the concentration of ethanol, the ratio of material to liquid, and the extraction time as independent variables and the extraction amount of indirubin as dependent variable, the optimum extraction process of indirubin from Isatidis Folium was predicted and analyzed by multiple linear regression and binomial fitting models with independent and dependent variables and the three-dimensional surface graph. Results; The optimal extraction process of indirubin was as follows: ethanol concentration 62%, solvent/sample ratio of 26, and extraction time 9 min. Under these conditions, the maximal extraction rate of indirubin was 4.37 mg/g which was consistent with model predictions. Conclusion: The optimal process is simple and convenient for extracting indirubin from Isatidis Folium with high precision, reproducibility, and predictability.
3.Purification of a Recombinant Human GST/MCP-1 Fusion Protein and Its Antitumor Effection

GST/MCP-1 fusion protein was overexpressed in Escherichia coli as inculstion bodies. After isolation of the inclusion bodies, the optimum conditions of denaturation and renaturation were studied. The renatured produts were purified to be electroporetically pure by affinity chromatography on immobilised glutathione. The purified product remains biological activities and can react specifically with MCP-1 antibodies by western blot analysis. The in vitro antitumor effect showed that GST/MCP-1 could activate monocytes and lymphocytes to inhibit the growth of a lung adenocarcinoma cell line A549. In vivo, GST/MCP-1 could inhibit the tumor growth in nude mice. These results suggested that MCP-1 had antitumor effect.
4.Effect of platelet-rich fibrin on proliferation and adipogenic differentiation of adipose-derived stem cells

Objective To study the effect of autogeneic platelet-rich fibrin (PRF) on proliferation and adipogenic differentiation of human adipose-derived stem cells (ADSCs) in vitro.Methods ADSCs were isolated from adipose tissue obtained from donors undergoing liposuction and were cultured,and underwent identification.ADSCs at passage 3 were divided into three groups:test groups were cultured with 1PRFM and 2PRFM,and control group was cultured without PRF membrane.Then the growth of the cells was observed by inverted microscope.MTT method was used to observe cell proliferation activity at days 1,2,3,4,5,6 and 7 after culture.Adipogenic differentiation of ADSCs was observed and quantified by oil red O staining at days 3,5,7,9,11 and 14.Results Cell proliferation and adipogenic differentiation would be increased with the PRFM,There were significant differences among three groups.Conclusions PRF could significantly promote proliferation and adipogenic differentiation of ADSCs.
5.Antimicrobial resistance in a class A tertiary hospital in Baise, Guangxi Province: 2015 surveillance report

Objective? To?understand?the?distribution?and?antibiotic?resistance?profile?of?clinical?isolates?in?the?Affiliated?Hospital?of Youjiang Medical University during 2015. Methods The pathogenic microorganisms were isolated from inpatients from January to?December,?2015?in?the?Affiliated?Hospital?of?Youjiang?Medical?University.?Antimicrobial?susceptibility?testing?was?carried?out?according?to?a?unified?protocol?using?Kirby-Bauer?method?or?VITEK?2-Compact?systems.?The?results?were?interpreted?according?to CLSL breakpoints released in 2014, and analyzed by WHONET 5.6 software. Results A total of 4004 strains of bacteria were collected, including 1146 (28.6%) strains of gram-positive bacteria, and 2858 (71.4%) strains of gram-negative organisms. The?prevalence?of?MRSA?and?MRCNS?was?25.8?%?and?79.4?%,?respectively.?MRSA?and?MRCNS?strains?were?significantly?more?resistant to most antibiotics than MSSA and MSCNS except trimethoprim-sulfamethoxazole. Most of the S. pneumoniae isolates were from non-meningitis patients, showing high resistance rate to macrolides and tetracycline, but very low resistance rate to quinolones. Enterococcus isolates were mainly E. faecium and E. faecalis. More E. faecium were resistant to high-level gentamicin and high-level streptomycin than E. faecalis. E. faecium isolates were generally more resistant than E. faecalis to most of the antimicrobial agents tested except clindamycin and tetracyclines. But no gram-positive cocci were found resistant to vancomycin, linezolid or tigecycline. ESBLs-producing strains accounted for 53.1% of the E. coli strains and 28.5% of K. pneumoniae isolates. Enterobacteriaceae isolates were still very susceptible to carbapenems. E. coli isolates were more resistant to most of the antimicrobial agents tested than other Enterobacteriaceae except to piperacillin-tazobactam and carbapenems. Enterobacteriaceae showed higher resistance to trimethoprim-sulfamethoxazole than the other antibiotics tested. Majority of P. aeruginosa isolates were susceptible to all the antibiotics tested (<10% resistant). A. baumannii?strains?showed?significantly?higher?resistance rate than P. aeruginosa to all the antibiotics tested. Conclusions Most of the data in this report are consistent with the national?data?in?terms?of?antimicrobial?resistance?profile.?These?data?are?useful?for?rational?use?of?antibiotics.
6.Effect of miRNA interference to annexinA3 gene on growth of human gallbladder cancer cells

Objective To construct a recombinant interfering RNA(miRNA)plasmid vector targeting annexinA3 and observe its impact on the growth and apoptosis of human gallbladder cancer cell line SGC-996.Methods Four small fragments of miRNA and one negative control were sequenced and cloned into vector pcDNATM 6.2-GW/EmGFPmiR.The recombinant plasmids were then transfected into gallbladder cancer cell line SGC-996 by positive liposomal transfection.The transcription and translation level of annexinA3 expression were detected by RT-PCR and Western Blot.Meanwhile, the proliferation and apoptosis of transfected tumor cells were determined by MTT and flow cytometry.Results The recombinant plasmids containing annexinA3 miRNA were successfully constructed.AnnexinA3 expression was significantly knocked down after miRNA plasmid transfection.Transfection of annexinA3 gene miRNA could effectively inhibit the proliferation of gallbladder cancer cell and promote apoptosis.Conclusion AnnexinA3 gene silenced by miRNA can induce the apoptosis of human gallbladder cancer cell line SGC-996.The process might serve as a potential approach for cancer gene therapy.
7.Control and Thinking of LIS System Management

Objective To introduce management of control and thinking from LIS System for providing some advice on the realization of clinical laboratory informatization. Methods Combined with our practical work, the general requirements, the overall structure, the design principles, the charging and the data security management of current information systems were analyzed. Results Under this model, LIS system was stable and secure, the laboratory workflow was also reasonable and the charging system is well in order. Conclusion The reasonable design and an overall co -ordination are required by the clinical laboratory information system, which will be better for our laboratory work and contribute much to the automation of laboratory information administration.

10.Correlation analysis on plasma D-dimer level with deep venous thrombosis after spinal surgery.

OBJECTIVETo analyze the relation of plasma D-dimer levels and incidence of deep venous thrombosis after spinal surgery.
METHODSThe clinical data of 63 patients underwent spinal surgery from October 2009 to October 2010 were retrospective analyzed. There were 40 males and 23 females with an average age of 48 years old(21 to 76) in operation. Operation levels of 15 cases were in cervical vertebrae, 4 cases were in thoracic vertebrae,and 44 cases were in lumbar vertebrae. Thirty patients with spinal fracture were caused by trauma and 33 patients without trauma, 11 patients combined with nerve injury. The patients were divided into two groups according to plasma D-dimer levels, more than or equal to 500 microg/L was D-dimer positive group and less than 500 microg/L was D-dimer negative group. Venous blood of all patients early morning with empty stomach were testd on admission, and at 2 h, 1 d, 2 d, 3 d, 4 d, 6 d, 8 d, 10 d, 15 d after operation,respectively.
RESULTSThere was no statistically significant differences in sex, operative segments, implants, operative posture, age, bleed volume, body weight, peroperative D-dimer levels between two groups. After operation, plasma D-dimer of 19 patients were more than or equal to 500 microg/L, with persistent or progressive increasing. Two cases occurred deep venous thrombosis in D-dimer positive group, they respectively were found at 3 days and 8 days after operation. Both of them underwent posterior decompression and internal fixation. However,no deep venous thrombosis was found in D-dimer negative group.
CONCLUSIONPostoperative D-dimer assay can effective predict deep venous thrombosis occurrence. D-dimer level more than or equal to 500 microg/L will be considered as a risk factor for deep venous thrombosis after spinal surgery.
