1.Trans-meridional-point therapeutics and transdermal therapeutic system

AIM: To compare difference and similarity of transdermal therapeutics between traditional Chinese medicine(TCM) and Western Medicine, to investigate the merit and shortage of the trans-meridional-point therapeutics in TCM, and to find out the problems urgent to be resolved so as to expand external remedy of TCM.METHODS: A part of literatures of TCM about transdermal therapeutics were reviewed, and PubMed database and relevant articles in Chinese medical journals were retrieved to analyze the relevant information RESULTS: The trans-meridional-point therapeutics of TCM had a long his tory, with rich experience and definite effect. The key of it was the amplification and integrative effect produced during the process of absorption of drugs through meridional point. The research of transdermal therapeutics combined with transdermal therapeutic system(TTS) had been in a starting stage, but it was limited in the research of a disease and a prescription, being of shortage in evidence of effective mechanism and morphology. Now one of the hot points in effective mechanism of TTS was to resolve rate-limiting process for medicine transportation through the corneal layer so as to promote entering of enough medicine into the body.CONCLUSION: It is urgent to solve the essence of the morphological basis of acupoint structure, establish experimental animal mode of transdermal therapeutics, and popularize clinical application on the basis of a great deal of medicinal experiments on animals so as to bring new ideas for transdermal remedy of Chinese drug.
2.Clinical study of urinary microprotein as predictors of outcome in critically ill patients

Objective To evaluate the feacibility of microalbumin(MA),alpha-1 -raicroglobulin (?1-MG) ,N-acetyl-beta-D-glucosaminidase (NAG), retinol-binding protein (RBP)as predictors of outcome in critically ill patients. Methods A prospective study was underwent in 30 critically ill patients.Urinary samples were collected at ICU admission and on the third,seventh ICU day for MA,?1-MG,NAG,RBP measurement.The severity of illness was assessed by APACHE II score calculated on the first ICU day,and the degree of organ dysfunction was assessed using SOFA score calculated on the first,third,seventh ICU day. Results MA,?1-MG,the durations of ICU stay and mechanical ventilation,APACHE II score, SOFA score were relative factors of MODS and death in ICU. There was significant association of APACHE II with MA (r=0.397),?1-MG (r=0.448), and RBP (r= 0.465), respectively. The area under ROC curve of APACHE II score,SOFA score,MA,?1-MG, RBP,NAG to predicate the death in ICU were 0.875 ( P

4.Cell Senescence and the Enzyme System for Surveillanceand Repair of DNA Damage

Senescence is one of the most important phenomena in cells' life. It is hold by one of hypothesis for cell senescence that residue DNA damages of a cell will accelerate its senescence.The normal function of surveillance and repair system for DNA damage is highly related with the senescence regulation of a cell. As a result, research of senescence regulation role of enzymes related for surveillance and repair of DNA damage, such as PARP, DNA-PK, ATM, p53, etc., will discover the inner relation between stress response of cell to DNA damage, regulation of DNA damage repair and cell senescence. That may be helpful for research of anti-aging and treatment of tumor by regulation of senescence of tumor cells.
5.Determination of TanshinoneⅡ_A in Ganxinkang Capsules by HPLC

OBJECTIVE:To establish HPLC method for the determination of tanshinoneⅡ A in Ganxinkang capsules.ME_ THODS:The separation was carried out on C 18 column.The mobile phase was composed of methanol-water(80∶20)with a fl_ ow rate at0.8ml/min and detection wavelength at269nm.RESULTS:The linear range of tanshinoneⅡ A was0.16?g~0.80?g (r=0.9989)with average recovery at96.3%(RSD=1.21%).CONCLUSION:This method was convenient,accurate,rapid and reproducible,and suitable for the quality control of this preparation.
6. Role of protease activated receptor-2 in rheumatoid arthritis: Recent progress

Protease activated receptor-2 (PAR-2) is a G-protein-coupled receptor. Recent studies indicate that PAR-2 is mainly expressed in leukocytes and activated by pancreatin and (or) tryptase, which subsequently induces inflammation through degranulation of leukocytes. Activation of PAR-2 in leukocytes is possibly involved in the pathogenesis of rheumatoid arthritis.
7.Microembolic signal monitoring in patients with symptomatic carotid artery stenosis

Objective To investigate the relationship of microembolic signals (MESs) between the degree of symptomatic carotid artery stenosis, ultrasonic characteristics of plaques, peak systolic velocity at the stenotic site and risk factors for stroke. Methods A total of 52 patients with symptomatic carotid artery stenosis were enrolled. MESs of bilateral middle cerebral arteries were monitored and detected by carotid color Doppler flow imaging. Results The positive rate of MESs on the symptomatic sides was significantly higher than that on the asymptomatic sides (28. 8% vs. 4. 5%, P < 0. 05). The positive rate was not significantly correlated with the degree of stenosis, ultrasonic characteristics of plaques, peak systolic velocity on the stenotic sides, and risk factors for stroke. Conclusions MESs mainly occurred on the symptomatic sides of carotid artery stenosis, and they were more closely correlated with unstable plaques.

9.Chromosome Location and Elongation of Radiation-induced Expressed Sequence Tag by the Aid of Bioinformatics

Regulation of gene expression is one of the most importa nt responses of cells to DNA damage induced by radiation. A novel expressed seq u ence tag (EST) fragment had been cloned from human embryo lung cells induced by 50cGy radiation and named RIG1. To clone the full-length cDNA of RIG1, a non-c loned cDNA library of human embryo lung cells induced by low dose irradiation ha d been established. This library was used as template in enchanced nest RACE PCR and biotin-labeled probe was used for further purification. The 3′ flanking s equence of this EST was cloned and sequenced with this set of technology. It was illuminated by homology analysis that this 3′ flanking sequence and the origin al EST are well aligned with a BAC clone of 20th chromosome and the predic ted exons sequence of this chromosome is well consisten ce with the real EST. Thus the RIG1 can be roughly located in 20th chromos ome. By use of the exons sequence predicted from chromosome sequence by GENSCA N, full-length of RIG1 gene has been cloned. Chromosome location of RIG1 gene i s further determined by this successful verification of Bioinformatics predictio n by experiment. By the same step, genome sequence of RIG1 has been determined. Therefore,by the combined use of Bioinformatics analysis,the full-length cDNA sequence and genome sequence of RIG1 gene are obtained and the predicted protei n sequence is determined.
10.Effects of ginseng fruit saponins(GFS)on coronary circulation and myocardial oxygen metabolism in dogs with acute myocardial infarction

Objective To study the effects of ginseng fruit saponins(GFS)on the coronary circulation and cardial oxygen metabolism in dogs with acute myocardial infarction.Methods The acute myocardial infarction model was induced by ligating the left anterior descending coronary of dogs for 6 h.90 min after treatment with GFS,the blood flow of coronary artery was determined by using electromagnetism flowmeter with MP-27 type.At the same time,the blood oxygen contents of artery and vein were determined by using analysis instrument of blood gas with CORNIHG 178 type.Results After treated by GFS(in a dosage of 5 and 10 mg?kg~(-1) iv drip for 90 min),the myocardial blood flow(MBF) was increased and coronary vascular resistance(CVR) was decreased significantly.Meanwhile the cardiac oxygen consumption and myocardial oxygen utilization rate were also decreased,but cardiac oxygen consumption index had not changed significantly.Conclusion GFS has protective effects on acute myocardial ischemia through improving the coronary circulation,increasing myocardial blood supply and decreasing cardiac oxygen consumption and myocardial oxygen utilization rate.
