1.Expression of lentivirus-mediated short hairpin RNA (shRNA) targeting collagen type Ⅰ in rat mesangial cells

Objective To investigate the expression of lentivirus-mediated short hairpin RNA (shRNA) targeting collagen typeⅠ (Col Ⅰ) in rat mesangial cells.MethodsThe Col Ⅰ shRNA lentivirus vector was constructed with three-plasmids packaging approach which encoding human/rat homology Col Ⅰ gene.Rat mesangial cells were infected by infection enhancer (control group),the empty lentiviral vector (pSC-GFP group),and lentiviral expression vector (pSC-GFP/ColⅠ group),respectively.Infection efficiency in the three groups was detected with flow cytometry,and the expression of Col Ⅰ mRNA and protein in the three groups was respectively detected by RT-PCR and Western blotting.ResultsRat mesangial cells were efficiently infected in pSC-GFP group and pSC-GFP/Col Ⅰ group.The infection efficiency was 72.42% in pSC-GFP group and 75.42% in pSC-GFP/Col Ⅰ group.RT-PCR showed that the expression of Col Ⅰ mRNA in pSC-GFP/Col Ⅰ group was significantly lower than that in control group and pSC-GFP group (P
2.Clinical effect of orthokeratology for juvenile with myopia astigmatism and its effects on corneal endothelial cells

Abstract?AIM:To investigate the clinical effect of orthokeratology for 400 juvenile with myopia astigmatism and its effects on corneal endothelial cells.?METHODS:Four hundred patients(800 eyes), of whom the average age was 11.5 ±2.3 years old, 239 male, 161 female, were divided into two groups: orthokeratology group and spectacles group. Parameters including efficacy data ( uncorrected visual acuity, corneal curvature, axial length and diopter ) and corneal endothelial cell data ( count of endothelial cell, endothelial cell density, fluorescein staining and central corneal thickness) were observed at 1d, 1, 6, 12 and 24mo after wearing.? RESULTS: The visual acuity of spectacles group recovered to normal after wearing, that of orthokeratology group recovered to normal at 1mo after wearing.At 2a after wearing, the corneal curvature, diopter of orthokeratology group decreased significantly (40.09 ±0.31D, 0.23 ±0.06D respectively); while those of spectacles group increased, the differences between the two groups were significant (P<0.05).The axial length of the two groups increased slightly at 1mo after wearing ( P>0.05 ) compared to those before wearing. At 2a after wearing, the axial length of the two groups were 23.96 ± 0.38mm, 26.49±0.88mm respectively (P<0.05).At 2a after wearing, central corneal thickness was 527.33 ± 27.69mm, 526.98±26.89μm(P>0.05).The count of endothelial cell and endothelial cell density both decreased after wearing without significant differences (P>0.05).?CONCLUSION: Orthokeratology has less effect on the corneal endothelial cells, no obvious adverse reactions and can control the prognosis of myopia.

Stem cells are characterized by self-renewing, multipotent differentiation, and high proliferation and receiving more and more attention for their roles in the development and management of various diseases. There are epithelial stem cells and mesenchymal stem cells in the prostate. The markers of the epithelial stem cells include cytokeratin, stem cell antigen-1, and integrins alpha2beta1, CD49f, CD133, CD117, and CD44. The markers of the mesenchymal stem cells include CD30, CD44, CD133, neuron-specific enolase, and vascular endothelial growth factor receptor-1. Prostate stem cells are involved in the development and treatment of prostatic diseases. This review focuses on the latest progress in the studies of prostate stem cells.

4.THE PRIMARY STUDY ON RABBIT BONE MARROW MESENCHYMAL STEM CELLS IN CONSTRUCTING TISSUE-ENGINEERED CARTILAGE

Objective To study the feasibility of constructing tissue engineered cartilage by differentiated rabbit bone marrow mesenchymal stem cells(MSC) cultured in vitro and in vivo. Methods The MSC were isolated from the nucleated cells fraction of autologous bone marrow by density gradient centrifuge, and then induced into chondrogenic differentiation by adding dexamethasone, transforming growth factor-β1(TGF-β1) and ascorbic acid in vitro. After 3 weeks, some cells turned to round shape and secreted metachromatic matrix. The cartilaginoid grafts composed of chondrogenic MSC. Bovine type Ⅰ collagen and human fibrin were cultured within the chondrogenic medium for 2 weeks in vitro or transplanted subcutaneously adjacent to the knee joint for 3 weeks in vivo. Results The most cells in the grafts were degenerated and disappeared after cultured in vitro. But the residual cells were survival and secreted metachromatic staining proteoglycan with toluidine blue, which was characteristic cartilage matrix. The grafts developed into matured cartilage tissue assessed by histological examination after 3 weeks of transplantation in vivo. Conclusion MSC can be used as functional cells to constructing tissue engineered cartilage.
5.Study on the Innovation System Synergy Degree of Pharmaceutical Manufacturing Industry in Shanghai Based on the Composite System Synergy Degree Model

OBJECTIVE:To study the innovation system synergies of pharmaceutical manufacturing industry in Shanghai and lay the foundation for further enhancing the competitiveness of pharmaceutical enterprises of Shanghai. METHODS:Based on the composite system synergy degree model,order variables(G)of innovation environment(S1),innovation technology research and development(S2),innovation technology absorption(S3)and innovation output(S4)in the pharmaceutical manufacturing industry innovation system in Shanghai during 2006-2014 were selected,synergy degree model was built,and order parameter,order de-gree,synergy degree of each order variable were calculated in turn. RESULTS & CONCLUSIONS:S1-S4 obtained 4,4,4,2 or-der variables respectively. According to the calculation,synergy degree of pharmaceutical manufacturing industry in Shanghai dur-ing 2006-2014 ranged in -0.195-0.202,average value was -0.068,overall in the level ofinverse synergyandnon-synergy. Thus,the internal development of the pharmaceutical manufacturing innovation system in Shanghai is more disorderly. It is suggest-ed to reform from the two core influencing factors of the stability of innovative environment and promoting technology absorption, including accelerating the transformation and upgrading of the pharmaceutical industry to create a stable and innovative environ-ment,increasing the innovation investment of the pharmaceutical industry to improve the level of technology absorption and inde-pendent innovation,as well as strengthening cooperation within and outside the domain to effectively enhance the system synergy degree level of pharmaceutical manufacturing innovation system in Shanghai.
6.The microscopic cavernous sinus surgery via the extended transsphenoidal approach: an anatomical study of cavernous sinus

Objective This study aimed to establish an anatomic basis for the microscopic surgical anatomy of the cavemous sinus via the extended transsphenoidal approach.Methods Simulated surgery via extended transsphenoidal ap-proach was performed on seven adult cadaver heads with red--colored latex injected arteries.The cavernous segment of the ICA and its branch vessels and its relationship with cranial nerve were exposed and its anatomic parameters were measured under microscope.Results The tuberculum sellae,clivus,sellar base,ICA prominence,and optic nerve prominence are the important bone landmarks to define the surgical area.The average of the extent of bone removal of extended transsphe-noidal approach is 37.6 mm(range:28.7 mm~44.0 mm).Conclusions The bone removal from sellar base to the media edge of the foramen rotundum and over the ICA prominence can effectively expose the entire unilateral cavernous sinus.The microscopic cavernous sinus surgery via the extended transsphenoidal approach is an optimal surgical approach for the le-sions that invade the cavernous sinus from sella.
7.Repair of articular cartilage defects with tissue-engineered cartilaginous grafts in rabbits

Objective To observe the effect of repairing the articular cartilage defects with tissue-engineered cartilaginous grafts in rabbits. Methods A total of 60 rabbits were divided into the cartilage graft group, the pure carrier control group and the blank control group. In the cartilage graft group, the bone marrow mesenchymal stem cells (MSCs) of the rabbits were obtained by isolating and culturing the bone marrow aspirates in vitro. The culture system that facilitates the chondrogenous differentiation of MSCs in rabbits was established. The tissue engineering cartilaginous graft was composed of chondrogenetic MSCs, bovine type Ⅰ collagen and human fibrin. Then transplantations of the cartilaginous grafts were performed to repair articular cartilage defects in rabbits. Results Hyaloid cartilage was formed within the defects repaired with the grafts at 12 weeks after transplantation by analyzing the content of type Ⅱ collagen and metachromatism. In the control groups, the fibrous cartilage repair was observed first, then the fibrous tissues and bone repairs were found. Conclusions The cartilaginous graft through tissue engineering is a promising graft for repairing articular cartilage defects.
8.Effect of Different Media on MPN Estimation of Ammonia-oxidizing Bacteria in Lakes

The MPN method was used to enumerate ammonia-oxidizing bacteria (AOB) in water and sediments of several shallow lakes. The suitable incubation time, medium types and substrate (ammonium sulphate) concentrations were studied. The results showed that, MPN values increased with the incubation time, reaching a stable maximum at some time stages, which was 40 days in all the samples for MSF medium. Among the three media used (XZ-AOB、MSF、SW), MSF give the highest MPN value. In addition, am- monium sulphate concentration in medium was an important factor affecting MPN estimation of AOB. Compared to AOB in lake sediments, AOB in lake water was more sensitive to ammonium sulphate concentration.
9.Biological markers of ischemic stroke subtypes according to TOAST classification

The identification of the specific cause in every patient has important clinical implications, because ischemic stroke is an etiologically heterogeneous disease. The Trial of Org 10172 in Acute Stroke Treatment (TOAST) classification can be used to define the etiology of stroke. However, TOAST classification can not be completed timely on admission of patients with acute stroke, which has impacted early guidance of clinical treatment. This article reviews the biological markers of early differential-diagnostic significance of the TOAST classification.
10.Hepatectomy for primary liver cancer without hepatic blood flow occlusion

This study was to evaluate the feasibility of hepatectomy for primary liver cancer (PLC) without hepatic blood flow occlusion. Methods 194 PLC patients admitted between 1988~1998 underwent hepatectomy without hepatic blood flow occlusion including nonanatomical hepatectomy (100 cases),hepatolobectomy (41 cases), combined adjacent organ resection (30 cases), hepatic segmentectomy (22 cases) and left hemihepatectomy (3 cases). Results Operative time was 2 4 hr, intraoperative blood transfusion averaged at 649 ml. Operative complication rate was 18 0%, and there was no mortality. Conclusion Hepatectomy without hepatic blood flow occlusion for PLC patients can be performed safely, so it is a useful technique for hepatectomy.
