OBJECTIVE To design the detection system of the leaky surface acoustic wave(LSAW) biosensor in(liquid) phase and detect human papilloma virus(HPV) target sequence with the LSAW biosensor.METHODS A detection delay line and a reference delay line were constructed, the phase of the two delay line changes was(evaluated) in liquid phase, and real time monitoring of HPV reaction was performed by using the LSAW biosensor system.RESULTS The phase of the two delay lines had different changes in liquid phase;HPV could lead to a great change of the phase in the detection delay line, and reference delay line had no change.CONCLUSIONS The(detection) system of the LSAW biosensor in liquid phase is successfully constructed and real time monitoring of(detection) of HPV is realized.

Objective To investigate the value of 99Tcm-methoxyisobutylisonitrile (MIBI) scintigraphy in assessing the preoperative chemotherapy response and multidrug resistance of osteosarcoma. Methods From January 2007 to October 2008, 12 patients (female:4, male:8; mean age:16.3 years,range:8-27 years) underwent early (10min) and delayed (120 min) 99Tcm-MIBI scintigraphy before and after preoperative chemotherapy.Seven cases had osteosarcoma at the distal femurs, 2 at the proximal tibias, 2 at the upper end of humerus and 1 at the fibula. The tumor-to-background ratio (T/B) and washout rate (WR) were calculated.Tumor necrosis was classified according to Huvos criterion after limb salvage surgery.Immunohistochemical staining for P-glycoprotein(gp) was examined.Spearman correlation analysis and t-test were performed. Results According to Huvos criterion, 7 patients were classified as good responders with more than 90% of tumor cell necrosis and 5 as poor responders with less than 90% of tumor cell necrosis.R value (ratio of early phase T/B after and before chemotherapy) was significantly lower in good responders than that in poor responders (0.473 ± 0.21 vs 0.998 ± 0.06, t= 5.342, P= 0.000 ).R value was significantly correlated with the degree of tumor cell necrosis ( rs=- 0.87, P= 0.000 ).WR was significantly higher in patients with positive P-gp expression than that in patients with negative P-gp expression ((38.36 ±18.64)% vs (6.40±5.87)%, t= -3.278, P=0.008).There was significant correlation between the WR and P-gp expression (rs = 0.91, P= 0.001 ).Conclusion 99Tcm-MIBI scintigraphy is a feasible non-invasive technique to assess the chemotherapy response and to detect P-gp expression of osteosarcoma.

Objective To investigate the features of familiar facial palsy, ophthalmoplegia and dysphagia characterized by autosomal dominant inheritance in a family and to discuss the classification and pathogenesis of the disease. Methods Clinical, electrophysiological, pathological examinations were performed and blood samples were obtained from 5 patients and 26 family members.PCR protocol was used to identify a certain gene. Results In the 5 patients receiving physical examination, all had ptosis, external ophthalmoplegia, facial paralysis, dyphagia, hoarseness, decreased pharyngeal reflex;4 had amyotrophy of muscle of tongue, temporal nuscle, masseter and muscles of distal lower limbs;3 had proximal limb asthenia and distal limbs amyotrophy.Compared to those of oculopharyngeal muscular dystrophy(OPMD)with similar symptoms and signs, both electrophysiological manifestation and pathological findings of the family members supported the diagnosis of muscular dystrophy, but the(GCG)6(GCA)3GCG in the first exon of PABPN1 mutated neither in normal family members nor in patients. Conclusions This family presents clinical manifestations somewhat resembling to those of OPMD and distinctive to other disorders, but has a totally different genetic background from OPMD.It may be a new subtype of muscular dystrophy.

Objective To establish the model of acute spinal infarction, to evaluate the relative factors affecting results in spinal embolization, and to provide the theorial basis with the preoperative embolization of spinal tumors. Methods Through the SAE of the lumbar arteries, the neuro function of the posterior legs of dogs, MRI findings, and pathologic changes of the spinal specimen were observed in 12 dogs. The embolizing agents was gelfoam(GF). Results The significant ischemia changes of spinal column and the corresponding muscles at the occluding spinal after embolizing more than one segmental arteries occurred in 9 dogs, but there were no paraplegia or obvious changes in 3 dogs having been embolized single lumbar arteries no matter they sent out the radiculomedullary artery(RA) or not. Paraplegia occurred in one dog after embolizing the multisegmental arteries. Conclusion (1) The method of SAE in dog can be used to set up the experimental model of the acute ischemia of spine. (2)The occlusion in single segmental arteries can not result in the infarction of the whole spine. (3)The serious complication may result from embolizing multisegmental spinal arteries (especially sending out RA) (4) The protecting embolization should be carried out in order to decrease the reaction during SAE in spine.

Objective To study the immunophenotypic features of NK/T cell lymphoma in children and its association with Epstein-Barr virus (EBV) infection. Methods Five cases of children′s NK/T cell lymphoma were studied. CD45RO, CD3?, CD56, CD20, TIA-1 and granzyme B were detected by immunohistochemistry staining for investigating immunophenotype. The expression of EBV-latent membrane protein (LMP-1) were detected by immunohistochemistry. EBV-encoded RNA (EBER1/2) were detected by in situ hybridization (ISH). Results CD45RO, CD3?, TIA-1 and granzyme B were positive in 5 cases, CD56 was positive in 2 cases, while CD20 negative in 5 cases.EBER1/2 positive in 4 cases and LMP-1 positive in 3 cases. Conclusions NK/T cell lymphoma in children is strong associated with EBV infection, and EBV infection may play an important role in the pathogenesis of children NK/T cell lymphoma.
