1.Relationship between formation of intestinal microflora and food allergy in infants

Objective To investigate the hypothesis that food allergy in infants may be associated with variation in their intestinal microflora. The formation of intestinal microflora in healthy infants and changes in food allergic infants were detected.Methods 16S rRNA gene sequences specific for bifidobacterium, lactobacillus and escherichia coli in fecal were quantitatively detected by real-time PCR. The three fecal floras were assessed in 71 healthy infants and 100 infants with food allergy. Results After birth,there were bifidobacteria colonized in infantile intestine,then the number increased rapidly up to 5 times at the sixth month, which was always the preponderant flora. Lactobacilli was also presented in infantile intestine 1 month after birth and augment gradually. The number of Escherichia coli was less than bifidobacteria and lactobacilli and appeared to decline during the early infants. The number of bifidobacteria and lactobacilli in the infants with food allergy were markedly less than that in the healthy infants, but escherichia coli was significantly more than that in the healthy infants.Conclusions During the first year of life,the intestinal microflora in infants is in a developing process. Compared with the healthy infants,bifidobacteria and lactobacilli decrease, but escherichia coli increase in the food allergic infants.These results indicate that the probiotics may be benefit to the prevention and treatment of food allergy.
2.System evaluation of health status and intervention in US Armed Forces

Objective To systematically evaluate the health status and intervention measures in the US Armed Forces, and to provide reference for the development of health promotion strategies in our army.Methods The PubMed, Medline, Springer, Elservier, HighWire and CNKI Database were searched electronically, with assigned search strategy for American military health status and intervention measures published from Jan.1997 to Dec.2015.Results There were 25 of pieces literature about training injury, low back pain, posttraumatic stress disorder, depression and cardiovascular disease that were screened.Conclusion The main health problems facing the US Armed Forces and the focus of the intervention measures involved training injuries, low back pain, mental illness and cardiovascular disease.Protection of military health is shifting from the single disease prevention to diversified comprehensive maintenance in the new era.
3.Network formulaology: a new strategy for modern research of traditional Chinese medicine formulae.

This paper briefly analyzed and discussed the current status and major scientific challenges of traditional Chinese medicine (TCM) formulaology research. To promote formulaology research, a new strategy and corresponding technology, network formulaology, were proposed to reveal the complex interaction between functional chemome and biological responses network. The research framework and directions of network formulaology were also summarized and prospected.

4.Changes in gene-expression profiling in kidney of rats with sepsis

Objective To investigate the gene-expression profile in kidney of rats during late sepsis (24hours) by using microarray technology in order to offer some clue to revealing the pathogenetic mechanism of sepsis at gene level. Method A total of 30 Wistar rats were selected and divided into model group and control group randomly(random number). The rats of control group were sham operated and the rats of model group received cecal ligature and puncture (CLP) operation. The biomarkers of renal function were assayed and the histopathological changes of kidney in rats were observed under transmission electron microscope 24 hours after operation. Gene chips containing 22 107 rat-genes cDNA were used to exmine gene-expression in kidney of septic rats to sieve the genes with different expressions with software. Data were analyzed by using SPSS version 11.0 software package.Statistical analyses of two independent samples carried out by using t -test. Results Compared with the control group, the levels of blood urea nitrogen (BUN) and creatinine (Cr) of sepsis group were higher (P < 0.01 ). The histopathological changes in kidney of rats demonstrated the establishment of sepsis model successful 24 hours later.Compared with the control group, there were 325 genes with differential expression in model group. Among the known-functional genes, there were 100 up-regulated and 64 down-regulated. Sorted by biological function, the genes were mainly related to metabolism, immunoresponse, cellular signal transduction, apoptosis, ion channel,growth factor and so on. Conclusions A sequence of genes expressed differentially in kidney of rats with late sepsis. Microarray technology played an important role in the research into sepsis mechanisms.
5.The relationship among IL-10, TNF gene polymorphisms, Helicobacter pylori infection and gastroduodenal diseases in Hubei Han ethnic

Objective To study the distribution of IL-10 and TNF gene polymorphisms in patients with gastroduodenal diseases in Hubei Han ethnic and their association with Helicobacter pylori (Hp) infection. Methods Six hundred and five patients with gastroduedenal diseases (196 chronic gastritis, 189 gastroduodenal ulcer and 220 gastric cancer) as well as 624 healthy controls were genotyped with PCR-RFLP method for IL-10-1082,-819,-592 and TNFα-308, lymphotoxin-α (LTα) Nco Ⅰ and AspH Ⅰ gene polymorphisms. Hp infection status was determined with ELLS& Results (1) There was significant difference of IL-10-1082 AG + GG genotype among the gastric cancer group with the non-malignant gastric diseases groups and healthy control group (P <0. 05). There was no significant difference of IL-10-592 and -819 gene polymorphisms among gastric cancer patients,non-malignant gastric disease patients and healthy controls (P>0. 05). The genotype frequencies of IL-10-819 were the same as those of IL-10-592. (2) Frequency of IL-10-1082 AG + GG genotype in gastric cancer patients with positive Hp was significantly higher than that in the other three groups (P < 0. 05). (3) Frequency of LTα Nco I AG genotype in gastric cancer patients with Hp infection was signiilcandy higher than that in Hp positive healthy controls (P < 0. 05). There were no other associations between TNFα-308, LTα Nco Ⅰ and AspH Ⅰ gene polymorphisms and Hp infection in gastroduodenal diseases. Conclusions (1) Allele AG + GG of IL-10-1082 was associated with gastric cancer in Han nationality of Hubei province. (2) IL-10-1082 AG + GG,LTct Nco ⅠAG heterozygous genotype may be associated with Hp infection in patients with gastric cancer in Han nationality of Hubei province.
9. Pu-erh tea's anti-atherosclerosis mechanism by reducing the expression and activity of NF-κB and promoting macrophage apoptosis

Objective: To explore the anti-atherosclerosis mechanism of pu-erh tea by reducing the expression and activity of NF-κB and promoting macrophage apoptosis. Methods: ① ApoE-/- mice were fed with pu-erh tea extract for 8 and 16 weeks to observe the positive rate of CD68 and the area of lipid plaque. ② Apoptosis detection kit was used to detect the apoptosis of macrophages. ③ P65 and IkB-β expressions in aortic plaque and macrophages were detected by Real-time quantitative PCR and Western blot; the expressions of inflammatory factors in aortic plaque were detected. Electrophoretic mobility change assay (EMSA) was used to analyze the binding activity of NF-κB DNA. Results: ① The area fraction of lipid plaque in ApoE-/- mice in the 16-week intervention group was significantly reduced (vs. control group, P<0.05). The area of lipid components in the 16-week intervention group was significantly reduced (vs. control group, P<0.05). ② The positive rate of CD68 in the plaque was significantly lower in the 8-week and 16-week intervention group than in the control group (vs. control group, P<0.05). The apoptosis rate of macrophages in aortic plaque (vs. control group, P<0.05). ③ The expressions of p65 gene and protein in macrophages of pu-erh tea extract intervention group 8 and 16 weeks decreased (vs. control group, P<0.05). The expressions of I-κBβ gene and protein in 16-week intervention group decreased (vs. control group, P<0.05). The relative expression levels of IL-6, IL-12 and TNF-α in aorta tissue of 16-week intervention group decreased compared with control group (vs. control group, all P<0.05). There was no significant difference in the expressions of IL-8, IL-18, MMP9, ICAM and VCAM between the two groups (vs. control group, all P>0.05). The average gray value of NF-κB DNA binding activity in the 8- and 16-week intervention groups decreased respectively (vs. control group, P<0.05). Conclusion: Pu-erh tea may play an anti-atherosclerosis role by inhibiting the expression and activity of NF-κB, promoting the apoptosis of macrophages and reducing the level of inflammation in vivo. This mechanism may not be mediated by I κB.
10.Effects of Paired Box2,Proliferation Cell Nuclear Antigen and Cell Apoptosis in Nephrotic Syndrome with Steroid-Resistance

Objective To investigate the expression of paired box2(Pax2),proliferation cell nuclear antigen(PCNA) and cell apoptosis in steroid-sensitive and steroid-resistant groups with primary nephrotic syndrome(PNS) and to find out the action of Pax2 expression in PNS with steroid-resistance.Method The expressions of Pax2,PCNA were evaluated by immunohistochemistry and cell apoptosis by fluorescence micoscope.Results Pax2 expression in renal tubule had a positive correlation with PCNA expression in steroid-sensitive group.In steroid-resistant group,Pax2 expression had no correlation with PCNA.Pax2 had a negative correlation with cell apoptosis.Conclusions Pax2 proper expression facilitate PCNA expression and repair tubulointerstitial lesions in steroid-sensitive group.Renal tubular epithelial cell proliferation coordinated with cell apoptosis.Pax2 overexpression in steroid-resistant group lead to the decrease of cell proliferation and cell apoptosis and lead to the severe tubule lesions,which made to glucocorticoid resistance.
