1.Expression of T cell factor-4 gene in dermal papilla cells of hair follicles

Objective To investigate the expression of T cell factor-4 (TCF4) gene in dermal papilla cells of hair follicles.Methods The expression of TCF4 gene was examined by in situ hybridization in scalp tissues of patients with alopecia areata and normal human controls,The protein and mRNA exprcssions of TCF4 were detected by immunochemistry and RT-PCR method,respectively,in aggregated and non-aggregated human dermal papilla cells.ResultsAs shown by in situ hybridization,TCF4 gene was expressed in the dermal papilla cells from healthy controls,but not in those from patients with alopecia areata.Both cell immunochemistry and RT-PCR showed that TCF4 gene expressed in aggregated dermal papilla cells,but not in non-aggregated dermal papilla cells.ConclusionsTCF4 gene is expressed in dermal papilla cells.The growth cycle Of follicles may be related to wnt signal.
2.Construction and expression of TCF4/pcDNA3.0 expression vector

ObjectiveTo clone TCF4 (T cell factor 4) gene and construct its eukaryotic expression vector. MethodsThe total RNA was extracted from the aggregated human dermal papilla cells. The full length cDNA encoding TCF4 was obtained by RT- PCR, digested by restriction enzyme, then inserted in the eukaryotic expression vector pcDNA3.0. The sequence and reading frame were confirmed by two restriction enzymes and sequencing. The recombinant vector TCF4/pcDNA3.0 was stably transfected into dermal papilla cells, and the expression changes of TCF4 gene were detected. ResultsTCF4 gene was cloned from dermal papilla cells and its eukaryotic expression vector was constructed. After the identification and sequencing, the reconstructed plasmid was confirmed containing the correct and full nucleotide sequence of TCF4 gene. After stable transfection, the mRNA and protein level of TCF4 gene were up-regulated in dermal papilla cells and the proliferation of dermal papilla cells was promoted. ConclusionThe expression vector TCF4/pcDNA3.0 was constructed successfully and could be expressed in the dermal papilla cells. TCF4 gene can promote the proliferation of the dermal papilla cells.
3.Enalapril Decreased Inducible Atrial Fibrillation in Old Rats

Objective: To study the effect of enalapril on inducible atrial fibrillation(AF) in old rats. Methods: Old male Wistar rats were randomly divided into control group(n = 12) and experimental group(n = 13). Rats in control group were fed routinely. Rats were fed with enalapril besides normal diet in experimental group for three months. Rats were then anesthetized, thoracotomy was performed and pericardium was opened to expose heart. Right atrium effective refractory period(ERP) was measured. Sinus conduction time (SCT) and sinus recovery time (SRT) were measured for evaluating sinus function. Interatrial conduction time(IACT) and atrium response to burst pacing were evaluated in vivo. Plasma angiotensinⅡ level and atrial tissue angiotensinⅡ level were determined by radioimmunoassay. Sections were cut from the tissue of atrium and stained with Masson trichrome. The ratio of the area occupied by interstitial to the total area was measured. Results: Contrast to control group,IACT and SRT were shorter in experimental group(P < 0.01 and P < 0.05 respectively). AF were induced in 9 rats in control group and 4 rats in experimental group(P < 0.05). AngiotensinⅡconcentration was significantly decreased in right and left atrium tissues of experimental group compared with that in control group(P < 0.01). A significant decrease in interstitial atrial fibrosis was presented in experimental group compared with that of control group(P < 0.01). Conclusion: Inducible atrial fibrillation rate was decreased in old rats after treatment with enalapril. This effect maybe resulted from the inhibited local atrium renin-angiotensin system and improved sinus node function by enalapril.

6.Retinal Function after Laser in Situ Keratomilensis

ObjectiveTo observe the retinal function and nerve fiber layer (RNFL) thickness after laser in situ keratomileusis (LASIK). MethodsLASIK was performed in 15 cases (30 eyes) with myopia after strict preoperative examination. All examinations such as vision, correction vision, diopter, intraocular pressure, corneal thickness, ocular axis, topography scan and fundus of eye examination were performed before and 1 day, 1 week, 1 month,3 months and 6 months after operation, as well as electroretinography (ERG), visual evoked potential (VEP), optical coherece tomography at same time. ResultsThere was not significant difference in the intraocular pressure, ERG and VEP 1 day after LASIK. The thickness of RNFL decreased 1 week after LASIK (P<0.05) and recovered 1~6 months later. ConclusionLASIK does not disturb the retinal function and RNFL thickness irreversiblely.
7.Long-term effects of early-life disaster exposure on mental health during the whole life cycle

Early nutritional status, lifestyle and life experiences are critical to the health and development of one’s whole life. Previous studies on the effect of earthquake, flood, ice storm and hurricane, famine on population health were reviewed and sorted out from a perspective of life cycle. It turned out that catastrophic events exposure during the fetus period could have some adverse effects on the growth and development of fetuses, newborns and children, and also on mental health during the life cycle from newborns to adults. Based on the research results at home and abroad, future research on the long-term impacts of early-life adverse events on mental health could be further explored from such aspects as expanding research objects, improving research methods, and extending research content.
8.Minimal invasive elastic intramedullary nails and external fixation for treatment of comminuted closed fracture of tibia-fibula shaft.

OBJECTIVETo investigate the clinical results of external fixation and AO titanium elastic intramedullary nailing for treatment of tibia-fibula comminuted closed fractures.
METHODSFrom June 2010 to June 2012,58 patients with tibia-fibula comminuted closed fractures were treated with external fixation and AO titanium elastic intramedullary nailing, including 31 males and 27 females with an average age of 38.5 years old ranging from 21 to 57 years old. According to the system of AO Classification, the fractures were classified as type B1 in 9 cases,type B2 in 7 cases, type B3 in 10 cases, type Cl in 14 cases, type C2 in 12 cases,and type C3 in 6 cases. According to the system of Winquist-Hanson,the fractures' comminuted were classified as grade 1 in 23 cases, grade 2 in 17 cases, grade 3 in 12 cases, and grade 4 in 6 cases. According to the system of Johner-Wruhs, clinical results were compared between different type and grade groups by the time of last followed-up.
RESULTSAll 58 patients were followed up with an average time of 6.8 months (ranged from 18 to 36 weeks). All fractures had clinical healing with an average time of 28 weeks (ranged from 24 to 32 weeks). The total rate of good to excellent results was 91.4%. The rate of good to excellent in the group of grade 1 was higher than that of other grades. The complication rates and fracture healing time would increase respectively with higher Winquist-Hanson's grade. The complication rates in the group of type C3 was higher than that of other types, but the rate of good to excellent was lower than that of other types. The complication rates in the group of type B1 was lower than that of other types,but the rate of good to excellent was higher than that of other types.
CONCLUSIONMinimal invasiveusing AO titanium elastic intramedullary nailing combined with external fixation for treatment of tibia-fibula fractures especially for the multiple-segment,long spiral mild-to-moderate comminuted with hidden fracture can get satisfactory reduction and reliable fixation,it conformes to the principle of BO completely, protects the fracture end blood supply,reduces the external fixation time, has less skin soft tissue complications, postoperative function recovered satisfactorily.

9.Clinical effect of staged repair and reconstruction of multiple ligament injuries in knee joints.

OBJECTIVETo evaluate clinical outcomes of anterior cruciate ligament (ACL) and posterior cruciate ligament (PCL) reconstruction under arthroscopy combined with limited open repair of medial collateral ligament (MCL) for the treatment of multiple ligament injuries of knee joints.
METHODSFrom March 2006 and June 2012,the data of 14 patients (14 knees) with multiple injuries of ACL, PCL, and MCL were collected. There were 8 males and 6 females with an average age of (31.8 +/- 8.1) years old (ranged, 20 to 49 years old). All the patients were performed with X-ray and MRI examination, and the results showed that 10 patients had combined with injuries of anterior cruciate ligament (ACL), posterior cruciate ligament (PCL) and medial collateral ligament (MCL); 4 patients had ALC,PCL and posterolateral corner (PLC) injuries. Four patients had medial meniscus injuries and 2 patients had lateral meniscus injuries. The MCL,PLC and meniscus injuries were treated with operation on the first stage, and functional exercises were performed 3 weeks after fixation. The reconstruction operation of ACL and (or) PCL was performed at the second stage under arthroscopy 3 to 6 months later when the movement range of knee joint recovered to the normal level with obvious relaxation.
RESULTSAll incisions healed by primary intention. All the patients were followed up with a mean duration of 48.9 months (ranged, 24 to 80 months). The Lysholm score was improved from preoperative 19.6 +/- 0.9 to the latest follow-up 87.1 +/- 2.8 (t=12.3, P<0.01). The International Knee Documentation Committee (IKDC) rating: 9 cases nearly recovered to normal, 5 cases were abnormal.
CONCLUSIONFor multiple ligament injuries in the knee, staged repair and reconstruction can effectively restore knee joint stability and function.

10.Cross-sectional study and influencing factors of self-management ability in patients undergoing continuous ambulatory peritoneal dialysis

Objective To investigate the status quo and influencing factors of self-management ability in patients undergoing continuous ambulatory peritoneal dialysis (CAPD).Methods Totally 339 CAPD patients with clinical follow-up regularly were investigated for the situation of self-management with general information questionnaire and Self-management Scale for CAPD.Results The total score of patients' self-management ability was (66.03±15.57).Multiple linear regression analysis showed that education level,marital status,occupation and duration of dialysis were influencing factors of patients' self-management ability.Conclusions Patients undergoing CAPD have certain degree of self-management ability.Nurses should promote their self-management ability according to different education level,marital status,occupation and duration of dialysis.
