2.Analysis of amplification and bioinformatics on mycobacterium tuberculosis protein higA

Objective To amplify the higA gene from the Mycobacterium tuberculosis,and to analyze the structure and function of their encoded proteins by using bioinformatics.Methods Total DNA was extracted from Mycobacterium tuberculosis.PCR of higA was performed and the products were sequenced.The biological features of the higA protein including,its physical and chemical properties,signal peptide,spatial structure and epitopes were analyzed by using software online.Results The PCR products of higA were 450 bp in length,which were consistent with the expected size.The higA protein consisted of 149 amino acids and had the following characteristics:a theoretical isoelectric point of 7.93,a fat-soluble factor of 94.30,and instability coefficient of 36.57.The higA protein had no signal peptide,containing 10 phosphorylation sites and multiple potential epitopes.Conclusion Mycobacterium tuberculosis higA gene can be amplified by PCR and the characteristics of higA protein is identified.

Articular cartilage defects are commonly found in clinics. It is always a great challenge for the repair of cartilage defects due to the limited self-regeneration after injury. Tissue engineering,a newly emerging biotechnique to regenerate cartilage with chondrogenic potential cells and biodegradable scaffolds in vivo and in vitro,provides a promising method to solve the challenge in cartilage defects repair. To regenerate cartilage with favourable structure and function,it is essential to gain a deep insight into the biochemical structure,biomechanical property and their relationship of articular cartilage. This article gives an introduction to the biochemical structure,biomechanical property and their relationship of both native and tissue-engineered cartilage.
4.Sympathetic Responses in Caloric Stimulation:Role in the Cardiovascular Control of Anesthetized Rats

Objective To explore the potential connection between sympathetic response, heart rate and blood pressure modulation after caloric irrigation in order to study the role of vestibular stimuli in cardiovascular control. Method Efferent splanchnic nerve firing rates, ECG and blood pressure were recorded simultaneously during caloric stimuli on intact anesthetized(CON) rats(n=5), sinoaortic denervated (SAD)rats(n=5) and bilateral vestibular destroyed (VD) rats(n=5). Result It was found that after caloric stimulation with ice water mean blood pressure CON rats with intact reflex became lower and the mean heart rates became slower, splanchnic sympathetic nerve activities increased for a moment and then dropped significantly. SAD rats had significant stronger splanchnic sympathetic nerve activities VD rats after caloric stimulation, and their blood pressures changed to apposite directions. The coupled respiratory component on splanhnic sympathetic nerve activities were strongly affected by the caloric stimulation. Conclusion It is suggested that semicircular canal stimulation participate at least in the short-term blood pressure control mechanism and the role of central nervous system on respiratory drive may also be involved. Baro-reflex and vestibular afferent may play different role in the control of blood pressure they may work synergically in some physiological control processed.
5.Primary Schwann cell growth on electrospun polymethyl methacrylate nanofiber scaffolds

Objective To evaluate the value of electrospun polymethyl methacrylate (PMMA) nanofibers with different topological structures as scaffolds for growth of Schwann cells (SCs).Methods Electrospun PMMA nanofibers with random or aligned topological structures were fabricated and measured with biocompatibility.Lentivirus-transfected green fluorescent protein was used as the reporting gene to monitor form and growth manner of SCs on different substrates and dependency of cell body and process with fiber structure,with PMMA thin films served as the control.Results Electrospun PMMA nanofibers revealed good biocompatibility and could exert contact guidance to the growth of SCs.Topological structures of the electrospun nanofibers influenced cell morphology.SCs were aligned with the orientation of substrate fibers and form longer cell process when growing on aligned nanofibers (P <0.01).Primary SCs preferred to follow the cue of aligned nanofibers compared to random fibers.Conclusion Aligned electrospun PMMA nanofibers have the potentiality as transplantable scaffolds for loading SCs after neural injury.
6.The effects of JARID1B siRNA on proliferation and apoptosis in HL-60 cell.

OBJECTIVETo study the effect of small interfering RNA(siRNA) targeting JARID1B gene on the proliferation and apoptosis in HL-60 acute promyelocytic leukemia cell line, and to explore its mechanisms.
METHODSThe JARID1B siRNA was transfected into HL-60 cells using Lipofectamine(TM) 2000(Lipo) vector. The proliferation inhibition by siRNA targeting JARID1B was detected by MTT, cells apoptosis by flow cytometry, the mRNA expression of JARID1B by RT-PCR, the protein expression of JARID1B, Bcl-2, procaspase-9, procaspase-3, c-myc and P27 and histone methylated H3K4 by Western blot.
RESULTSsiRNA targeting JARID1B upregulated histone methylated H3K4 level, inhibited the proliferation of HL-60 cells, and induced the cells apoptosis. After transfection of siRNA targeting JARID1B at 0, 30, 60, 120 nmmol/L for 24 hours, the apoptotic rate were (11.0 ± 3.6)%, (35.2 ± 5.1)%, (52.7 ± 3.8)%, and (62.0 ± 5.7)% respectively (F = 70.27, P < 0.01). The protein expression of P27 was upregulated, and Bcl-2, procaspase-9, procaspase-3, c-myc was down regulated.
CONCLUSIONSJARID1B siRNA upregulates histone methylated H3K4. It reduces HL-60 cells proliferation and apoptosis by up regulating the p27 expression and down regulating the Bcl-2, procaspase-9, procaspase-3, c-myc expression. It might be a new therapeutic targeting for human leukemia.

7.Study on effects of Tripterygium wilfordii polycoride in resisting macrophage inflammation and regulating inflammation via TLR4/NF-κB.

To investigate the effect of Tripterygium wilfordii polycoride (TWP) on LPS-induced macrophage inflammatory response, particularly the inhibitory effect on inflammatory factors TNF-α and IL-1β and the regulatory effect on inflammation via TLR4/NF-κB. The MTT method was adopted to test the effects of tested drugs, TWP, dexamethasone (DXM) and azathioprine (AZA) on cell growth to define the appropriate concentration. LPS was used to induce the inflammatory reaction in mouse RAW264. 7 cell lines. The Elisa kit was adopted to test the release level of TNF-α and IL-1β. The Western blotting was applied to test the protein expressions of TNF-α and IL-1β. The RT-PCR was adopted to test the expressions of TLR4 and NF-κB. According to the results, TWP could inhibit the release of macrophage inflammatory factors TNF-α and IL-1β in a dose dependent manner. All of TWP groups showed a weaker efficacy than that of the DXM group. But the TWP high dose group revealed a better effect on TNF-α and equal effect on IL-1β compared with the AZA group. TWP show an equal or better effect in down-regulating TLR4 and NF-κB p65 expressions in a dose dependent manner than DXM and AZA. In conclusion, TWP could inhibit TLR4 and NF-κB p65, which may be related to the down-regulation of TLR4 and NF-κB p65 receptor expressions.

8.Expression of Bcl-2,Bax and proliferating cell nuclear antigen in the liver of rats with hepatic fibrosis and the effect of transforming growth factor-?1 vaccine on them

Objective To observe the expression of Bcl-2,Bax and proliferating cell nuclear antigen(PCNA) in liver of rats with hepatic fibrosis and the effects of transforming growth factor (TGF)-?1 vaccine on them.Methods Thirty healthy male Sprague-Dawley rats were assigned into 3 groups,named healthy control group(n=10),hepatic fibrosis group(n=10) and TGF-?1 vaccine treated group(n=10).The animal model with hepatic fibrosis was established by injecting solution dimethylnitrosamine (DMN) into abdominal cavity with concentration as 0.5% and dose as 0.2 mL/ 100 g.In TGF-?1 vaccine treated group,every rat was not only injected with DMN but also 150?g TGF-?1 vaccine protein.On the 42nd day,all rats were sacrificed.Then the blood and the liver tis- sues were collected.The expression levels of Bcl-2,Bax and PCNA in liver tissues were detected by S -P immunohistochemistry and observed by routine pathological evaluation.Alanine aminotransferase (ALT),aspartate aminotransferase(AST) and albumin(Alb) were determined by auto biochemical analytical tool.Serum levels of hyaluronic acid (HA),laminin(LN) were detected by radioimmunoas- say (RIA).Results The expression of Bax,which promoted apoptosis,directly correlated with pathological grade in liver of rats,while the expression of Bcl-2 and Bcl-2/Bax,which protected a gainst apoptosis,inversely correlated with pathological grade in liver of rats.The expression levels of TGF-?1 and Bax in healthy control group were significantly lower than those of fibrosis group,how ever,the expression levels of Bcl-2 were comparable between these two groups.As compared with fi- brosis group,the expression of TGF-?1 was significantly lower while the expression of Bcl-2 was sig nificantly higher in TGF-?1 vaccine treated group.However,the expression of Bax was comparable between these two groups.The expression level of PCNA of fibrosis group was significantly higher than that of healthy control group but dramatically lower than that of TGF-?1 vaccine treated group (Both P
9.Pathological features of cerebral gliomas after stereotactic radiosurgery and radiotherapy: a comparative study

Objective To study the histopathological changes and ultrastructure features of human gliomas after receiving stereotactic radiosurgery (SRS) and stereotactic radiotherapy (SRT), and observe the changes of tumor-associated factor Ki-67 level, endothelial growth factor (VEGF) level and microvessel density (MVD).Methods A comparative, retrospective study of pathological and immunohistochemical changes of 25 gliorna specimens without radiotherapy (controls) and 25 patients with glioma received with SRS and SRT (treatment group), admitted to our hospital from November 1995to July 2008, were carried out. Eight specimens from both groups, fixed with glutaraldehyde, were chosen for electron microscope observation; the ultrastructure changes in the tumor center, tumor margin and peripheral edema brain tissues in each specimen were observed. Immunohistochemical staining was employed to detect the expressions of Ki-67 and VEGF, and the MVD. And these data were statistically analyzed. Results The square and extent of tumor necrosis and liquation were positively significantly correlated with the tumor grade (r=0.649, P=0.001). Electron microscope indicated that the organdles,capillaries and blood-brain barrier in the tumor center, tumor margin and peritumoral edema cortex of the treatment group occurred different degrees of brain tissue degeneration and necrosis; while these were without damage in the controls. Immunohistochemistry showed that the protein expression of VEGF, the Ki-67 positive cells, and the MVD in the control group and treatment group were positively correlated with the glioma grade (P<0.05). Of the glioma with the same grade, the Ki-67 protein expression and MVD in the treatment group were significantly lower than those in the control group (P<0.05).Conclusion SRS and SRT may lead tumor cells to different degrees of degeneration, necrosis, and apoptosis, as well as the three-tier structure damage of capillaries in the blood-brain barrier, which provides a laboratory and theoretical foundation to take individualized comprehensive treatment of different gliomas.
10.Clinicopathologic study of distal 2 cm rectal carcinoma intramural spread over dentate line.

OBJECTIVETo study the extent of distal intramural spread of distal 2 cm rectal carcinoma over the dentate line, and to improve quality of life for these patients through providing pathological evidence of operation mode choice.
METHODSSpecimens of thirty patients with rectal carcinoma, operated with ISR(intersphincter resection) or Miles procedure from May 2005 to July 2007, were collected, and their large and histology pathologic slices were examined. Specimens were dissected distally to the dentate line per 2 mm within 1cm and per 5 mm exceed 1cm. The length of distal intramural spread to rectal carcinoma was measured under light microscope.
RESULTSAmong the 30 specimens, distal intramural spread over the dentate line was observed only on one case and the invasion length over the dentate line was more than 2 cm, the invasion lengths of the other 29 cases were within the dentate lines.
CONCLUSIONDistal 2 cm rectal carcinomas seldom spread over dentate lines. Anus discomfortableness of these patients and other complications could be reduced through reserving more skin over the dentate line, which are important for the improving of quality of life in these patient.
