1.Effects of rhCNTF on axoplasmic transport following incomplete injury of optic nerves of adult rats

Objective To observe the effects of ciliary neurotrophic factor (CNTF) on the recovery of axoplasmic transport following incomplete injury of the optic nerves (ON) of the adult rats. Methods Moderate injury of the optic nerves of 50 adult female rats was induced by crushing the optic nerves for 10 seconds using hemostatic forceps. Recombinant human CNTF (rhCNTF) was injected into the vitreous cavity of CNTF treated group at 0, 1, 2, 4, and 8 weeks after injury. However, the same volume of distilled water was injected into the vitreous cavity of the control rats. The changes of axoplasmic transport were assessed by anterograde labelling combined with horseradish peroxidase (HRP) at 1, 2, 4, 8, and 12 weeks after injury. Results The labelling density at ON distal to the site of injury by HRP obviously decreased at 1 and 2 weeks after injury, and gradually enhanced at 4 weeks and peaked at 8 weeks after injury. The recovery ratios of axoplasmic transport in CNTF treated group and control group at 8 weeks after injury were 31 86% and 12 87%, respectively. No HRP reaction product was observed in the superior colliculus at 1 and 2 weeks after injury. 2.Role of preoperative neutrophil to lymphocyte ratio, platelet to lymphocyte ratio, prognostic nutritional index in the prognosis of patients with hepatitis B virus-related hepatocellular carcinoma after radical resection

Objective To investigate the role of preoperative peripheral blood neutrophil to lymphocyte ratio (NLR),platelet to lymphocyte ratio (PLR),prognostic nutritional index (PNI) in the prognosis of patients with hepatitis B virus (HBV)-related hepatocellular carcinoma (HCC) after radical operation.Methods This is a retrospective study,involving 426 surgically resected hepatitis B related hepatocellular carcinoma cases in a single center from 2003 to 2012.Results Kaplan-Meier analysis showed patients in NLR ≤ 1.62 group achieve higher rate of recurrence-free and overall survival than that in the NLR > 1.62 group,the difference was statistically significant (P < 0.005);Also PNI > 49.42 group showed higher rate of overall survival significantly than PNI≤49.42 group (P < 0.005).The results of Cox regression multivariate analysis further suggested that both NLR > 1.62 (HR 1.74,P =0.007) and PNI ≤49.42 (HR 0.70,P =0.021) were independent risk factors for overall survival,NLR > 1.62 (HR 1.45,P =0.03) was also an independent risk factor for recurrence-free survival.Conclusion The preoperative NLR and PNI may be independent risk factors for prognosis of patients with HBV-related HCC after radical operation.
3.Protective effect of trichostatin a and 5-azacitidine on cytokine-induced toxicity in pancreatic β-cells

Objective To investigate the effect of trichostatin A (TSA) and 5-azacitidine (5-AzaC) on pancreatic β-cells impaired by cytokine,via measuring the proliferation,apoptosis,and function of pancreatic β-cells.Methods RIN-m5f was impaired by interleukin-1β and interferon-γin vitro,and treated with TSA and 5-AzaC.Experiment groups included blank control group,cytokine induction group,0.05/0.10 μmoL/L TSA group,0.63/1.25 μmoL/L 5-AzaC group,and0.10 μmol/L TSA plus 1.25 μmol/L 5-AzaC group.The viability of RIN-m5f cells was detected by MTT assay.Apoptotic rate was determined by Annexin V-fluorescein isothiocyanate (FITC) /propidium iodide flow cytometry.Insulin secretion was measured by enzyme-linked immunosorbent assay.Results The viability of RIN-m5f cells in 0.05/0.10 μmoL/L TSA group,0.63/1.25 μmol/L 5-AzaC group,and 5-AzaC plus TSA group was 70.1%/79.2 %,67.3 %/82.9 %,and 89.1% respectively,being higher than that in the cytokine group (33.9%,P<0.05) ; the apoptosis rate was 10.3%/10.5%,7.9%/9.6%,and 8.2%,being lower than that in the cytokine group (16.6%,P<0.05) ; the capacity of glucose-stimulated insulin secretion of all the treated groups was higher than that in the cytokine group (P<0.05).Conclusion TSA and 5-AzaC might promote the proliferation of pancreatic β-cells impaired by cytokines,inhibit its apoptosis and recover its insulin secretion.
4.Comparison among the effects of tiaogan recipe, jianpi recipe,bushen recipe and ginsenoside on the contents of amino acids in hypothalamus and hippocampus of rats under repeatedly psychic stress

BACKGROUND: When one is in a stress state, some amino acids as neurotransmitter in his brain are of important regulating action to his cerebral functions and his psychic behaviors,and some traditional Chinese drugs can regulate the stress state of the body.OBJECTIVE:To observe the content changes of glutamic acid,aspartic acid, γ-aminobutyric acid and taurine in the hypothalamus and hippocampus of rats under repeatedly psychic stress so as to investigate the effects of tiaogan recipe, jianpi recipe, bushen recipe and ginsenoside on them.DESIGN: A randomized grouping and controlled observation trial.SETTING:Department of Basic Theory of Traditional Chinese Medicine(TCM) of Basic Medical Science College, Guangzhou University of TCM.MATERIALS:The experiment was completed from March 2002 to January 2003 at the Animal Center of Guangzhou University of TCM.Totally60 Wistar rats were randomly divided into 6 groups: normal, model, tiaogan, bushen, jianpi and ginsenoside groups, with 10 in each group. Compositions and doses of tiaogan recipe: radix bupleuri 5 g, fruetus gardeniae 5 g,radix paeoniae alba 15 g,fructus lycii 15 g,fructus aurantii 6 g,radix rehmanniae 18 g, concha haliotidis 30 g. Compositions and doses of shenqi pill: radix rehmanniae 30 g, rhizoma dioscoreae 15 g, fructus corni 15 g,rhizoma alismatis 10 g,poria 10 g,cortex moutan radicis 10 g,ramulus cinnamomi 4 g, radix aconiti praeparata 4 g. Compositions and doses of sijunzi decoction:radix codonopsis pilosulae 20 g,rhizoma atractylodis macrocephalae 15 g, poria 15 g, radix glycyrrhizae preparata 6 g.METHODS:① The traditional Chinese medicines were conventionally decocted; and the tiaogan recipe condensed to the liquid containing1.69 g/mL crude drug, shenqi pill containing 1.76 g/mL crude drug,sijunzi decoction containing 1.01 g/mL crude drug.Ginsenoside was prepared as 7 g/L water solution.The rats in the normal and model groups were by gavage given 2 mL of 9.0 g/L sodium chloride injection.The rats in the tiaogan, bushen, jianpi and ginsenoside groups were respectively by gavage given 2 mL of tiaogan recipe, shenqi pill, sijunzi decoction and ginsenoside solution 1 hour before immobilization stress. ② Except for rats in the normal group, those in the rest groups were all conducted for establishment of psychic stress reaction model.The rats were put into an immobilization tube,their action space was gradually reduced by using a mobile insertion piece,they were regulated to a nervous state without prpduction of intense revolting, which was done once a day, starting with 4 hours immobilization on the first day, and later on increased by 30-60 minutes per day, for consecutive 14 days. ③ The whole brain of the rats in each group was collected by decapitation,OPA high-performance liquid chromatography was used for assays of the contents of glutamic acid, aspartic acid, γaminobutyric acid and taurine in hypothalamus and hippocampus of the rats.MAIN OUTCOME MEASURES: The content changes of glutamic acid,aspartic acid,γ-aminobutyric acid and tanrine in the hypothalamus and hippocampus of rats in each group.RESULTS: Totally 60 rats involved all entered into the result analysis. ①The content changes of glutamic acid in the hypothalamus and hippocampus of rats in each group:Compared with normal group,the contents of glutamic acid in hypothalamus of rats in the tiaogan, jianpi,bushen and ginsenoside groups were markedly decreased [(21.85±8.19), (15.76±1.80),(14.68±7.91), (21.46±5.45), (13.43±7.68) μmoL/g]; compared with model group,the contents in the tiaogan, bushen, jianpi and ginsenoside groups were obviously raised [(11.04±3.65), (11.78±2.17), (18.67±2.98), (20.91 ±3.96),(17.71±1.83) μ moL/g, P < 0.05, P < 0.01, P < 0.01, P < 0.05]. ② The content changes of aspartic acid in the hypothalamus and hippocampus of rats in each group:Compared with model group,the contents of aspartic acid in the hypothalamus in the jianpi and ginsenoside groups were obviously decreased [(8.65±1.18), (5.72±1.32), (4.67±1.88) μmoL/g, P < 0.01,P < 0.01], while the contents in the hippocampus of rats in the jianpi,bushen and ginsenoside groups were markedly raised[(2.58 ±0.87),(3.93±0.49), (4.52±0.98), (3.83±0.41) μmoL/g, P < 0.01, P < 0.01, P < 0.05].③ The content changes of γ-aminobutyric acid in the hypothalamus and hippocampus of rats in each group:Compared with model group,the contents of γ-aminobutyric acid in the hypothalamus of rats in the tiaogan,jianpi,bushen and ginsenoside groups were markedly decreased[(20.92±4.96), (15.87±2.90), (13.84±2.63), (14.94±3.98), (10.94±3.68) μ moL/g,P < 0.05, P < 0.05, P < 0.01, P < 0.01], while the contents in the hypothalamus were obviously raised [(4.12±1.66), (4.18±1.04), (6.67±1.29),(6.11±0.99), (6.37±0.78) μmoL/g, P< 0.05, P< 0.01, P< 0.01, P< 0.01].④ The content changes of taurine in the hypothalamus and hippocampus of rats in each group: Compared with model group, the contents of taurine in the hypothalamus of rats in the tiaogan, jianpi, bushen and ginsenoside groups were markedly decreased [(10.24±1.72), (7.82±1.14), (8.00±2.05),(6.42±3.17) μmoL/g, P < 0.05, P < 0.05, P < 0.01], while the contents in the hippocampus in the jianpi and ginsenoside groups were obviously raised [(12.61±3.51), (17.03±2.74), (18.04±2.14) μnoL/g, P< 0.01, P< 0.01].CONCLUSION:The central acting site of tiaogan recipe may mainly be in the hypothalamus,possibly being related with down-regulating amino acids.While the central acting sites ofjianpi recipe,bushen recipe and ginsenoside may include the hippocampus and hypothalamus, being mainly related with up-regulating amino acids,through enhancing the integration of the hippocampus on stress so as to gain the effect of anti-injury of stress.
5.Stability of akebia saponin D in gastrointestinal contents of rats.

In order to study the stability of akebia saponin D (ASD) in biological fluids in vitro, the determination methods of ASD were established in this study. Akebia saponin D was dissolved in artificial gastric juice, intestinal juice and gastrointestinal contents of rats, respectively, then thermostatically maintained at 37 degrees C. At time intervals after degradation, samples were withdrawn and the concentrations of ASD were determined by HPLC, from which stability of it at different biological specimen was evaluated. As a result, ASD was totally degraded in large intestinal contents of rats in 8 hours. ASD was very stable in artificial gastric juice, intestinal juice and gastric contents of rats. All of the above data proved that ASD was easily degraded by coliform bacteria but stable in acid environment and with the presence of digestive enzyme.

6.Application of computer aided design in the treatment of acetabular malignant tumor: One-case report

BACKGROUND: Acetabular malignant tumor reconstruction is to obtain pelvic stability and lower limb walking function to excise the tumor at safe margin.Excision range has been evaluated by MRI,CT,X-ray,which are subjective and lack preoperative design.Computer-aided three-dimensional reconstruction can evaluate tumor erosion range from all planes to accurately excise the tumor.OBJECTIVE: To evaluate the value of computer aided design in the treatment of acetabular malignant tumor.METHODS: One case with acetabular hemangiosarcoma was checked with lamellar CT scanning,which acquired some two-dimensional data in disease area.The three-dimensional reconstruction of anatomical model,design of cutting bone extent,design of individual prosthesis and sham operation were made by computer.Based on computer aided design proposal,acetabular tumor was resected,pelvic ring and right hip articulation were reconstructed with allogeneic semi-pelvis and individual total hip replacement.RESULTS AND CONCLUSION: The patient began to non-weight bearing walk with double crutches 2 months after operation.At6 months,the patient walked normally.The right hip joint motion was good with no pain.Postoperative X-ray film displayed individual prosthesis matched to pelvis.The patient fell a little numbness of skin in the lateral of right hip.No phlebothrombosis,prosthesis loosening or dislocation was found.Computer aided design has a good perspective of application in the treatment of acetabular malignant tumor.Individualized treatment can improve operation accuracy,reliability,convenience and curative effect.
7.Intestinal Function Changes in Mice after Spinal Cord Injury in Early Stage

Objective To investigate the influence of spinal cord injury on intestinal function in mice after a short period. Methods 105 Kunming mice were randomly assigned to normal group (group A, n=30), sham group (group B, n=30) and model group (group C, n=45). Group A received no treatment, group B was only exposed the spinal cord, while group C was induced by a constant compression with a modified arterial forcep at T10 to establish spinal cord injury model. 12 hours, 24 hours and 48 hours after modeling, myoelectric slow wave activity and tension activity of the ileum were tested, and HE staining was used. Results Compared with groups A and B, the slow wave activity was significantly weaker in group C at every time point (P<0.05), so was the amplitude of tension activity (P<0.05). Frequency of tension activity was obviously higher in group C than in groups A and B 24 hours and 48 hours after modeling (P<0.05). The injury scores were higher in group C than in groups A and B (P<0.05). Conclusion There is reduce in myoelectric slow wave activity, tension activity of the ileum and mild injury in intestinal mucosa in mice after spinal cord injury in the early stage.
8.Distribution and morphological characteristics of symptomatic atherosclerotic plaques in the middle cerebral artery using high?resolution magnetic resonance imaging

Objective To observe distribution and morphological characteristics of symptomatic atherosclerotic plaques in the middle cerebral artery (MCA) using high?resolution magnetic resonance imaging (HR?MRI), and to investigate HR?MRI characteristics of atherosclerotic plaques in the MCA in patients with acute cerebral infarction. Methods A total of 57 symptomatic patients with MCA atherosclerotic plaques recruited in the Affiliated Hospital of Yangzhou University from January 2014 to January 2016 were imaged with diffusion weighted imaging (DWI), three dimensional time of flight magnetic resonance angiography (3D TOF?MRA) and HR?MRI scanning for plaque on a 3.0 T MRI scanner. According to the results of DWI examination, the 57 patients were divided into transient ischemic attack (TIA) group (27 cases) and acute cerebral infarction group (30 cases). The distribution of the narrowest lumen plaque was evaluated by cross?section division into four equal arcs (superior, inferior, ventral, dorsal arcs). For quantitative analysis, lumen area (LAMLN), vessel area (VAMLN) at maximal lumen narrow (MLN) and LAreference, VAreference were measured, then wall area (WA), plaque area (PA), percentage of plaque burden, rate of lumen stenosis and remodeling index (RI) were calculated. The data of each group were compared and analyzed. Results The location and morphological analysis of the 57 patients with symptomatic MCA atherosclerotic plaques revealed that plaques were located in the ventral wall in 19 cases (33.3%), the upper wall in 15 cases (26.3%), the dorsal wall in 10 cases (17.5%), and the lower wall in 13 cases (22.8%). For the location variations in ventral wall, upper wall, dorsal wall and lower wall, the TIA group was shown as six cases (22.2%), five cases (18.5%), seven cases (25.9%) and nine cases (33.3%), and the acute cerebral infarction group was shown as 13 cases (43.3%), 10 cases (33.3%), three cases (10.0%) and four cases (13.3%), respectively. There was no statistically significant difference in the distribution of each side wall between the two groups (P>0.05). VAreference, LAreference, VAMLN and RI of the TIA group and the acute cerebral infarction group were (19.89 ± 1.34) mm2, (15.19 ± 2.04) mm2, (20.78 ± 1.78) mm2, 1.09 ± 0.11 and (19.70 ± 1.34) mm2, (14.60 ± 2.33) mm2, (21.53 ± 2.34) mm2, 1.10 ± 0.11, respectively. There was no statistically significant difference between the two groups (P>0.05). The remodeling patterns of both groups were mainly positive remodeling, with a total of 44 cases (77.2%). In the TIA group and the acute cerebral infarction group, the WAMLN, PA, stenosis rate and plaque load percentages were (8.85±1.92) mm2, (4.00±3.00) mm2, 20.92%± 9.18%, 19.05% ± 14.93% and (11.10 ± 1.88) mm2, (6.00 ± 2.25) mm2, 28.56% ± 8.67%, 27.30% ± 7.69%, respectively. The differences between the two groups were statistically significant (t=-4.466, t=-2.865, t=-3.231, t=-2.580, P<0.01). There were eight patients (29.6%) with unsmooth plaque surface in the TIA group and 19 patients (63.3%) in the acute cerebral infarction group. The differences between the two groups were statistically significant (χ2=6.475, P<0.05). LAMLN in the TIA group and the acute cerebral infarction group was (11.93±1.59) mm2 and (10.43±2.08) mm2 respectively, and the difference between the two groups was statistically significant (t=3.033, P<0.01). Conclusions Symptomatic atherosclerotic plaques in MCA in the acute cerebral infarction group have higher plaque load, thicker vascular wall at the maximum stenosis and more unsmooth plaque surface. This indicates the characteristics of high?risk plaques to a certain extent.
9.Calcifying fibrous tumor of the stomach: report of nine cases and review of literature

Objective:To investigate the clinicopathological features and pathological diagnosis methods of calcifying fibrous tumor (CFT) of the stomach.Methods:The clinicopathological data of 9 patients with gastric CFT in the Second Hospital of Hebei Medical University from August 2015 to May 2020 were retrospectively analyzed, immunohistochemistry was used to detect the immunophenotypic characteristics of gastric CFT, and the relevant literature was reviewed.Results:Among 9 patients with gastric CFT, 1 case was male and 8 cases were female, with a median age of 53 years old (18-63 years old). There were 7 cases occurred in the body of the stomach, while 2 cases in the fundus. All cases were single solid nodule with the cut surface gray-white or gray-red in colour and hard and tough in texture. Maximum diameter of tumor ranged from 0.6 cm to 1.5 cm. Observation under the microscope showed the spindle-shaped tumor cells were sparsely distributed in a large amount of collagenized stroma with scattered lymphocytes and plasma cells infiltration. Calcification or gravel formation was detected in all cases. The tumor cells in 9 cases expressed Vimentin, 3 cases expressed CD34, none of them expressed PDGFRA, SMA, SMMS-1, Desmin, ALK, DOG-1, CD117, S-100, β-catenin, CKpan and calponin; Ki-67 positive index was 1%-3%.Conclusions:Gastric CFT is more common in middle-aged and elderly people, which is characterized by gastric body with smaller volume. The diagnosis of gastric CFT mainly depends on pathological morphology and immunohistochemical examination. It needs to be differentiated from a variety of spindle cell tumors more commonly found in the stomach.
10.CT diagnosis of midgut malrotation.

OBJECTIVETo explore the diagnostic value of CT in midgut malrotation.
METHODSThe CT appearances of 16 patients with midgut malrotation were analyzed retrospectively.
RESULTSThe features of CT manifestation in 16 cases were as follows: (1) Horizontal part of duodenum could not reach medioventral line or could reach it but encircled right-down behind the superior mesenteric artery(SMA). (2) Ectopic ileocecal junction. (3) Jejunum located in right-middle abdomen while ileum in left abdomen. (4) A clockwise or counterclockwise rotation of the superior mesenteric vein (SMV) around the SMA. (5) Mid-gut volvulus.(6)Accompanied by other malformations.
CONCLUSIONAmbulation of duodenum, location of the small intestine and colon as well as anatomical position of mesenteric vessels should be intensively observed in order to exclude midgut malrotation.
