1.Losartan in the Treatment of Essential Hypertension

To investigate the efficacy and safety of losartan in the treatment of essential hypertension, 81 patients were randomly assigned to two groups. Patients in the first group(n =41) were given losartan 50 mg to 100 mg once a day; those in the second group( n = 40) were given benazepril 10 mg to 20 mg once a day. The treatment course lasted for a weeks. Ambulatory blood pressure, hepatic and renal functions, blood glucose were examined before and after therapy. Our results showed that the total efficacy rate in losartan group was 93% while that in benazepril group was 90%. The efficacy was similar between the two groups. However, effect of 24 h blood pressure control by lossrtan was superior to than of benazepril. The adverse reaction of losartan was milder that that of benazepril. It is concluded that losartan is a safe, long-acting antihypertensive agent for mild and moderate hypertension with good patient tolerance and less adverse reactions.
2.Prolapse of Fallopian Tube into Vaginal Vault after Hysterectomy: A case report.

The prolapse of a fallopian tube into the vagina is a rare complication of hysterectomy. We recently experienced a case of tubal prolapse after a laparoscopic hysterectomy in a 44-year-old woman. The vaginal examination showed a polypoid mass in the vaginal apex. Microscopically, the mass had the typical appearance of a fallopian tube with chronic inflammation. The cytologic finding of a vaginal vault smear was also described.

6.Treatment of murine hypoparathyroidism with transgenetic hemopoietic stem cell transplantation

Objective To investigate the gene therapy of hypoparathyroidism (HPT) with hemopoietic stem cell s(HSCs) integrated with parathyroid hormone (PTH) gene.Methods Human PTH gene was amplified by polymerase chain reaction from pcDNA 3.1 -PTH templates and inserted into murine stem cell virus (MSCV) vectors. The plasmids recombinated with PTH gene were transfected into PA317 packaging cells and screened for positive clones with Geneticin. The condensed recombinant retroviruses were obtained and used to infect HSCs coming from umbilical cord blood. The infected HSCs were injected into the mice suffering from HPT. The change of symptoms and serum level of PTH and calcium in each group of mice was observed.Results Human PTH gene was proved to be inserted into MSCV vectors and the titres were up to 2?10 7 colony forming unit (CFU)/ml in condensed retroviral solution. The secretion of PTH reached 15 ng per 10 6 cells per 48 h. The wild type viruses were not detected by PCR amplification. The mice suffering from HPT recovered quickly and the serum level of calcium and PTH remained normal for about 3 months after the HSCs recombinated with PTH gene were injected into them. In the groups of mice subject to the injection of the recombinant retroviruses containing PTH gene, the serum level of PTH and Calcium was significantly increased in a short time, and then declined slowly. And the mice took on the manifestation of hypoparathyroidism again. The mice only subject to the implantation of HSCs were all dead within 20 days.Conclusion A long term therapeutic effect could be obtained by infusing the HSCs which were integrated with PTH gene into the mice suffered from hypoparathyroidism.
7.Reactivation of human herpesvirus 8 by hepatocyte growth factor/scatter factor similar to that produced by HIV-1-infected T cells via ORF50 promoter of HHV-8

Objective To investigate a potential role for ORF50 promoter of human herpesvirus 8(HHV 8)in reactivation of HHV 8 in primary effusion lymphoma(PEL)BC 3 cells by HIV 1 related inflammatory cytokine,hepatocyte growth factor/scatter factor(HGF/SF). Methods The persistent stimulation of BC 3 was performed by recombinant HGF/SF and treated BC 3 cells were collected at the 3 rd and 7 th day after persistent stimulation respectively. Northern blot, quantitative PCR(real time PCR), and electron microscopy(EM)were carried out to detect the expression of mRNA of minor capsid protein ORF26 and the presence of viral particles of HHV 8 from treated BC 3 cells. Co transfection of BC 3 and human umbilical veil endothelial cells (HUVECs) with HHV 8 ORF50 promoter driven luciferase construct built previously by us and recombinant expression plasmid named pEV containing HIV 1 Tat coding gene was performed and stimulation of co transfected cells with recombinant HGF/SF and/or HIV 1 recombinant gp120 was conducted at 24 hours after transfection to induce luciferase gene expression. Then relative luciferase activity unit(RLU) was calculated at 24 hours after stimulation. In the meantime, stimulation of co transfected cells with 12 O tetradecanoylphorbol 13 acetate(TPA) was performed to be used as positive control. Results It was found that HGF/SF induced an 4.1 folds increase in mRNA expression of ORF26 when it was added individually to BC 3 cells at the 7 th day after persistent stimulation. Viral particles of HHV 8 were readily identified in BC 3 cells stimulated with HGF/SF at the 7 th day with EM analysis. Recombinant HGF/SF could upregulate promoter activity of HHV 8 ORF50 promoter in BC 3 and HUVECs cells and however, HIV 1 Tat and gp120 failed to act synergistically with HGF/SF to enhance ORF50 promoter activity in HUVECs. Conclusions The results show that HIV 1 related inflammatory cytokine HGF/SF is responsible for the induction of HHV 8 lytic cycle replication and the induction may be, at least partly mediated by ORF50 promoter of HHV 8.
8.Ultrasonic Diagnosis in Upper Extremity Deep Venous Flow Obstruction and Superior Vena Cava Syndrome

Objective To explore the pathogeny, pathology, ultrasonic and clinical expression of upper extremity deep venous flow obstruction and superior vena cava syndrome. Methods Retrospective analysis and summarize the disease changes of 10 cases in recent 5 years. Results 7 patients of 10 cases were superior vena cava syndrome, in which 4 cases caused by upper right lung malignant tumor, 3 cases caused by upper and media mediastinal malignant tumor. 2 patients of 10 cases were found thrombosis in right subclavian vein caused by duct of artificial pacemaker. The left 1 patient was no reason thrombosis in right axillary vein. Conclusion Ultrasonography combine with color and spectral Dopper image can evaluate the clinical development and therapeutic efficiency, which is a useful and effective non-invasive examination.
