Objective To investigate the mechanism of recombinant human growth hormone(rhGH) in articular cartilage defect repair in vivo. Methods The cylindrical,full-thick articular cartilage defects,(3.5 mm) in diameter, were made in the knee joints of 25 rabbits.rhGH of 0.1 U/kg was injected into the right knee joints 3 times a week for 4 weeks and equal volume of physiological saline to the left knee joints as control. The animals were killed respectively in 4,6,8,12,24 weeks, and the macroscopic, histologic and ultrastractural examinations were performed. Results The repairing process in experimental group was faster and better than that in the control. In the experimental groups, the defect was filled on week 4,the regenerated cartilage mainly in the shallow region and fibrous tissue in the deep region;on week 24,the regenerated tissue was morphologically close to the neighbour normal cartilage, hard to differentiate by macroscopy;Light microscopy showed the defect recovered to normal cartilage structure and the electron microscopy showed a large quantity of mature cartilage cells. In the control groups, the defect was of shallow introcession, mainly of fibrous tissue. The histological examination showed the significant difference between experimental and control groups(P

Chinese Journal of Applied Physiology 2013;29(2):147-152
OBJECTIVEThis study was performed to determine whether recombinant human angiopoietin-1 (rhAng-1) decreases the permeability of the blood-brain barrier (BBB) in focal cerebral ischemia and reperfusion rats, whether RhAng-1 opens the BBB by affecting tight junction associated proteins zonnula occludin-1 (ZO-1), occludin and adherens junction protein vascular endothelial (VE)-cadherin.
METHODSThe rats were divided into eight groups randomly( n = 10): (1) sham-operated group; (2) ischemia group; (3)-(5) ischemia/reperfusion (middle cerebral artery occlusion and reperfusion (MCAO/R) 12 h, 48 h, and 7 days) and 0.9% saline groups; (6)-(8) ischemia/reperfusion (MCAO/R 12 h, 48 h, and 7 days) and rhAng-1 groups. The Bee permeability was assessed by Evans blue extravasation. The messenger RNA and protein expressions of ZO-1, occludin, and VE-cadherin were determined by reverse transcription-polymerase chain reaction, Western blot, and immunohistochemistry assays.
RESULTSThe BBB permeability and the brain infarct volume were significantly decreased after rhAng-1 injection. The expressions of ZO-1, occludin, and VE-cadherin were increased after rhAng-1 injection.
CONCLUSIONrhAng-1 may decrease the permeability of BBB in MCAO/R rats by upregulation of ZO-1, occludin, and VE-cadherin.

Chinese Journal of Tissue Engineering Research 2014;(25):3980-3986
BACKGROUND: Numerous evidence has demonstrated that the magnesium aloy with excelent mechanical properties can degradein vivo, and can be used as vascular stent materials, bone fixation materials, porous materials for bone repair. But it is not reported in the biliary stent. OBJECTIVE:To observe the degradation procedures and evaluate the changes of mechanical characteristics of biliary stents made of magnesium aloy (AZ 31B) in human bilein vitro. METHODS:The baloon-expandable biliary stents were made of magnesium aloy with laser sculpture. Then, 15 stents treated with micro-arc oxidation on the surface served as experimental group and another 15 stents without micro-arc oxidation as control group. A dynamic numerical simulation system was establishedin vitro to simulate the internal environment of human biliary tract. Al of the biliary stents were placed into this system. Their shapes were observed, and their qualities and radical forces were measured every 20 days. At the same time, their surfaces were scanned by scanning electron microscope. RESULTS AND CONCLUSION:(1) The degradation speeds of the stents in the experimental group were obviously slower than those in the control group. The fracture of the connecting rods in control group and experimental group appeared 20 days and 40 days later, respectively. The peak time of degradation in the control group and experimental group were 30 days and 50 days, respectively. The stents were fuly biodegraded within 70 days in the control group while within 90 days in the experimental group. With time, the stent surface became more rough, and the density, area and depth of etch pits were al increased in the two groups. At the same stage, the degradation speed of the control group was more rapid than that in the experimental group. (2) The qualities and radical forces of the stents were gradualy reduced with time in both groups. However, the degradation speed of stents in the experimental group was much slower than that in the control group. In summary, the degradation speed of the biliary stents made of magnesium aloy treated with micro-arc oxidation is appropriate and can meet the clinical requirement for the degradation time of biliary stents. This novel material could potentialy be used for the preparation of biliary stents.

Chinese Journal of Tissue Engineering Research 2014;(1):88-93
BACKGROUND:The rabbit adipose-derived stem cells are mostly isolated by type I col agenase digestion, but rarely by explants culture method.
OBJECTIVE:To isolate rabbit adipose-derived stem cells for adipogenic and osteogenic differentiation.
METHODS:The rabbit adipose-derived stem cells were isolated from rabbit adipose by explants culture method, and cultured in vitro fol owed by morphological observation. The grow curve and cellsurface markers CD29, CD44, CD45 of passage 3 cells were analyzed respectively by 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide and flow cytometry;cells from the third passages were induced for adipogenic and osteogenic differentiation by different revulsants, and cells were examined by oil red O staining and alizarin red staining .
RESULTS AND CONCLUSION:The rabbit adipose-derived stem cells cultured in vitro exhibited a spindle-shaped appearance and could rapidly expand. Flow cytometry analysis revealed that the third passage of rabbit adipose-derived stem cells was positive for CD29, CD44, but negative for CD45. Rabbit adipose-derived stem cells were positive for oil red O staining at 14 days of adipogenic induction, and positive for alizarin red staining at 14 days of osteogenic induction. In conclusion, we could successful y isolate rabbit adipose-derived stem cells using explants culture method.
