OBJECTIVETo construct a cDNA library of osteosarcoma SAOS-2 cells and screen the proteins interacting with the bone morphogenetic protein-2 (BMP-2) to explore the role of BMP-2 in the development of prostate cancer.
METHODSThe total RNA was extracted from SAOS-2 cells, from which poly(A)(+)RNA was purified to generate the cDNA using RT-PCR with primer SMARTIII and primer CDSIIIoligo(dT). The library was constructed and screened by cotransformation of the double-stranded cDNA (dscDNA) and linearized pGADT7-Rec with the bait plasmid to the yeast AH109. The proteins interacting with BMP-2 were screened by AH109 mating with the bait strain Y187, and the interaction was confirmed using far-Western blot analysis.
RESULTSThe cDNA library of human osteosarcoma SAOS-2 cells was constructed, which was characterized by high diversity and sufficient capacity. The dscDNA size range was between 250-5000 bp, with a cotransformation efficiency of 4.3 x 10(5) and recombination efficiency of 1.9 x 10(6), and 4.3 x 10(5) clones were screened. The mating efficiency was 32% and 1.0 x 10(6) clones were screened by the bait of BMP-2, and 4 positive clones was obtained and sequenced.
CONCLUSIONThe diversity and capacity of the cDNA library meet the needs for screening genes related to prostate cancer.

Objective To compare the stability effect of the fixation segment using the new test system based robotics to simulate the complex human movement at natural state about the three-level fixa-tion by using four, five or six pedicle screws in treatment of thoracolumbar burst fractures. Methods Six human cadaveric spines were dissected from T_(11)-L_3. The inferior half part of L_1 vertebral bodies and L_1-L_2 dises were resected to mimie an unstable L_1 burst fracture with loss of anterior column support. Specimens were tested in accordance with the following order:intact, 4,5 and 6 pedicle screws fixation at the three-level fixation. The range of motion (ROM) of the fixation segment (T_(11)-L_3) was measured with the six-freedom degree robotics system controlled by mixed force and displacement during flexion, exten-sion, lateral bending and axial torsion, when the stiffness was calculated. One-way statistieal analysis was used for analyzing the collected data. Results With increased number of screws, the ROM of the fixa-tion segment (T_(11)-L_3) was gradually decreased and the stiffness gradually increased. The ROM under ax-ial rotation of six and five screws group became smaller than four screws group (P < 0.05). The stiffness under axial rotation of six screws group was higher than four screws group (P < 0.05). There was no sta-tistical difference between five screws group and four screws group in regard of the stiffness in axial rota-tion (P > 0.05). There were no significant differences in ROM and stiffness under six different loading directions between six screws group and five screws group (P > 0.05). No statistical difference was observed for three fixation modes in aspects of ROMs and stiffness under flexion-extension or lateral ben-ding (P > 0.05). Conclusions Three-level fixation of burst fractures with five or six screws offers im-proved biomechanical stability compared with traditional four screws fixation. But the difference is insig-nificant between six and five screws fixations.

8.Effect of fenofibrate on the diversity of intestinal flora in a mouse model of nonalcoholic fatty liver disease
Journal of Clinical Hepatology 2020;36(4):829-834
ObjectiveTo investigate the effect of fenofibrate on the diversity of intestinal flora in mice with nonalcoholic fatty liver disease (NAFLD). MethodsA total of 30 mice were randomly divided into normal group, high-fat group, and fenofibrate treatment group, with 10 mice in each group. The mice in the high-fat group and the fenofibrate treatment group were given high-fat diet intervention for 14 weeks, and those in the normal group were given normal diet for 14 weeks. After 10 weeks of dietary intervention, the mice in the fenofibrate treatment group were given fenofibrate by gavage and high-fat diet for another 4 weeks, and the change in body weight was monitored during the whole process. Fecal samples were collected after 14 weeks, and high-throughput sequencing 16S rRNA was used to investigate the diversity and difference of intestinal flora. Liver tissue samples were collected, and HE staining and oil red O staining were performed to observe the degree of steatosis. A one-way analysis of variance was used for comparison between multiple groups, and the least significant difference t-test was used for further comparison between two groups. ResultsAfter 10 weeks of intervention, the fenofibrate treatment group had a significantly lower body weight than the high-fat group (P<0.05). The results of HE staining and oil red O staining showed that the fenofibrate treatment group had a significantly lower degree of fat deposition than the high-fat group. There was a significant difference in intestinal flora between the fenofibrate treatment group and the high-fat group, while there was no significant difference in intestinal flora between the fenofibrate treatment group and the normal group. The fenofibrate treatment group had significant increases in the abundance of Bacteroidetes, Verrucomicrobia, Faecalibaculum, Muribaculaceae_norank, and Akkermansia and significant reductions in the abundance of Firmicutes, Actinobacteria, Clostridium, Turicibacter, and Bifidobacterium. ConclusionThe increases in the abundance of Bacteroidetes and Verrucomicrobia and the reductions in the abundance of Firmicutes and Actinobacteria suggest that fenofibrate may have positive significance in the treatment of NAFLD.

OBJECTIVETo investigate the biomechanical effects of micro-implant anchorage technique with sliding mechanics on maxillary anterior teeth retraction under different implant insertion heights and different retraction hook heights.
METHODSThe three dimensional finite element model of maxillary anterior teeth retraction force system was constructed with CT scanning and MIMICS software and the relationships between brackets, teeth, wire and micro-implant were simulating the clinical factions. Then the initial tooth displacement was calculated when the insertion heights were 4 mm and 8 mm and the retraction hook heights were 1, 4, 7, 10 mm respectively.
RESULTSWith retraction hook height added, the anterior teeth movement changed from lingual crown tipping to labial crown tipping and the intrusion movement was more apparent when the micro-implant was inserted in a higher location.
CONCLUSIONThe ideal teeth movement control could be achieved by different insertion heights of micro-implant and different retraction hook heights in straight wire retraction force system.
