1.Content Comparison of 18 Amino Acids in Plancenta Histolysate Determined by Post-column Derivatization Cation-exchange Chromatography and Pre-column Derivatization HPLC

Objective:To compare the content differences of 18 amino acids in plancenta histolysate determined by pre-column de-rivatization HPLC and post-column derivatization cation-exchange chromatography ( AARO) . Methods: The HPLC method was per-formed on a C18 column and 2, 4-dinitro chlorobenzene ( DNCB) was used for pre-column derivatization, and then the determination was carried out after adding 0. 1 mol·L-1 borax buffer (pH=9. 1), and the AARO method was used for the direct determination with a strong acid cation-exchange chromatographic column and post-column derivatization. Results: The RSD for reproducibility of the AARO method was 1. 84%-0. 91%, while that of the HPLC method was 1. 87%-1. 04%. Conclusion:Both AARO method and HPLC method can be used for the quantitative determination of 18 amino acids in plancenta histolysate with the similar results. However, pre-column derivatization HPLC method may cause incomplete derivatization and instable derivatives.
2.Development of Circulation System for Disinfection of Sickbeds

Objective To receive disinfection messages of sickbeds from nurse workstations and to reduce the periods of the request transfer.Methods The system transferred the request for replacing bed between wards and disinfection units by UDP messages,and informed the staff by active speaker and LCD screen.Results The time of replacing beds decreased significantly,so the wards were very satisfied with the disinfection units.Conclusion The system can reduce the time of replacing beds obviously and make the requests for replacing beds satisfied.
3. Effect of melatonin on hormone secretion and tissue ultrastructure of hypothalamus-pituitary-thyroid axis in rats

Objective: To investigate the effect of melatonin (Mel) on hormone secretion and tissue ultrastructure of hypothalamus-pituitary-thyroid axis(HPTA) in rats. Methods: A total of 60 male Sprague-Dawley rats were evenly randomized into four groups: control, low-dose Mel(0.5 mg·kg -1·d-1), middle-dose Mel(10 mg·kg -1·d-1), and high-dose Mel(50 mg·kg -1·d-1) group. The rats in the latter 3 groups received intraperitoneal injection of different doses of Mel daily. At 1 st, 2nd and 3rd weeks after treatment, rats were sacrificed and blood serum samples, hypothalamus, pituitary and thyroid tissues were obtained. Serum T3, T4, FT3, FT4 and TSH levels were estimated by radioimmunoassay (RIA). Expression of proTRH mRNA in hypothalamus tissues were detected by real time RT-PCR. The ultrastructures of hypothalamus, pituitary and thyroid were observed under transmission electronic microscope. Results: The serum levels of T4, FT3, FT4 and TSH in the middle-dose and high-dose Mel groups were significantly decreased compared with those in the control and low-dose Mel group at 1st, 2nd and 3rd weeks(P<0.01). T4 levels decreased in the middle-dose and high-dose Mel groups at a weekly basis, with significant difference found between different weeks (P<0.01). TSH level was deceased at the 2nd week, and that at the 3rd week was significantly lower than those at the first two weeks (P<0.01). The expression of proTRH mRNA was lower in high-dose Mel group compared to that in the control group at 1st, 2nd and 3rd weeks. With the prolongation of Mel treatment, expression of proTRH mRNA had a decreasing tendency first and an increasing tendency later. No obvious pathological ultrastructure changes were found in the hypothalamus, pituitary and thyroid tissues. Conclusion: Melatonin may inhibit Hypothalamus-Pituitary-Thyroid axis in a time- and dose-dependent manner in rats.
5.Asymmetric index analysis on the orthodontic-orthognathic treatment of facial asymmetry patients in skull positioning posterior-anterior radiographs.

OBJECTIVETo analyze the asymmetry of measurements in facial asymmetry patients with skull positioning posterior-anterior radiographs after orthodontic-orthognathic treatment.
METHODSPosterior-anterior cephalometric radiography and cephalometric analysis were performed in forty-five patients with different degrees of facial asymmetry. A single sample t-test was conducted to compare the asymmetry of measurements before treatment and individual normal occlusion. The measurement and reference values in facial asymmetry patients who underwent orthodontic-orthognathic treatment were summated by analyzing the correlation coefficient. The paired t-test was employed to compare the difference between the two groups before and after the treatment.
RESULTSFourteen measurements were significantly different before the treatment and individual normal occlusion (P < 0.05). Me[X] exhibited a higher correlation index of Ag[X] in forty-five cases. Twenty-eight patients showed the main asymmetry in the mandibular body, whereas fifteen patients in the ramus. Two patients showed the main asymmetry in both mandibular body and ramus. Fifteen measurements were significantly different before and after the treatment (P < 0.05).
CONCLUSIONFacial asymmetry is mainly concentrated in one-third of the surface, primarily manifested in the mandibular body. Orthodontic-orthognathic treatment is preferred when Ag[X] is greater than the minimum reference value of 11.31%, Go[X] is greater than 9.79%, and Me[X] is greater than 5.2 mm.

6.The relationship between distribution of different subtypes of human immunodeficiency virus-1 and the transmission routes in Shenzhen

ObjectiveTo investigate the distribution of human immunodeficiency virus-1 (HIV-1)subtypes and the relationship with transmission routes in Shenzhen city. MethodsThe epidemiological survey was conducted in 429 subjects with HIV-1 infection.Vein blood was collected and prepared with anticoagulation.Thereafter,the viral nucleic acid was extracted and subsequently amplified for HIV-1 env,gag genes by reverse transcription nested polymerase chain reaction. The viral sequences were analyzed using Mega software and compared with those in HIV database. The HIV-1 subtypes were confirmed and evolutionary tree was established. The gene discrete rate of different subtypes was calculated.ResultsFour hundred qualified samples were identified as CRF01_AE (n=207),CRF07_BC (n=115),CRF08 _BC (n=14),CRF02_AG (n=1),B (n=49) and C(n=14),respectively.CRF01_AE was the predominant subtype that was detected in the subjects with different transmission routes,such as homosexual contact,heterosexual contact,intravenous drug use and hospital-acquired infection. Among these,sexual contact was the most common route.There were nine cases infected with subtype B HIV-1 in 10 case who were blood borne. The gene discrete rate was more significant in subtype B HIV-1 compared to the other subtypes.According to the evolutionary tree analysis,the viruses under the same subtype formed the relatively independent clusters by different transmission routes. But some overlaps were observed as well. Conclusions Tbere are many HIV-1subtypes in HIV-1 infected subjects in Shenzhen, while the recombinant viruses present as the predominant form.Transmission routes are various with sexual contact as the most common route.Every subset distributes unevenly and forms its own characteristic.Overlap can be observed among the gene distribution of different HIV-1 subtypes.
7.Anatomical variation in the circle of Willis and cerebral ischemia

The circle of Willis is the major cerebral collateral circulation.Its anatomical variation is in populations higher.The previous studies showed that anatomical variation in circle of Willis was an independent risk factor for cerebral ischemia.This article reviews the relationship between the various anatomical variation types of the circle of Willis and cerebral ischemia.
8.Correlation study of blood uric acid level and carotid atherosclerosis in perimenopausal women

[Objective]To observe the relationship between blood uric acid(UA)level and carotid atherosclerosis(CAS)in perimenopausal women.[Methods] Two hundred and thirty-three perimenopausal women were selected.Intima media thickness(IMT)of bilateral carotid arteries were measured by colour Doppler ultrasound.They were divided into normal group(162 cases)and CAS group(71 cases)according to the results of colour Doppler ultrasound.The level of blood UA between two groups was compared and the influence factors of CAS we re analyzed.[Results] The level of blood UA in CAS group was significantly higher than that in normal group[(341 ± 86)μ mol/L vs.(293±78)μ mol/L](P < 0.05).The detection rate of hyperuricemia in normal group was 10.49%(17/162),while the detection rate in CAS group was 40.85%(29/71),which had statistical significance between two groupe(P<0.01).Pearson correlation analysis showed that CAS had positive correlation with age(r=0.233,P=0.028),body mass index(BMI)(r=0.215,P=0.044),bloodUA(r=0.249,P=0.019)and fasting plasma ghcose(r=0.214,P=0.046).AndCASwas negatively correlated with high density lipoprotein cholesterol(r=-0.224,P=0.036).Logistic regression analysis showed that after the factors of age and BMI were corrected,CAS still had significantly positive correlation with the level of blood UA(OR=4.805,95% CI 1.700-11.673,P < 0.05).[Conclusion]Hyperuricemia is a related risk factor of the incidence of carotid atherosclerosis in perimenopausal women.
9.Comparison of the preparation of high purity collagen type I from four biomaterials

[Objective]To compare the purity and extraction rate of four purecollagen type I products prepared from various biomaterials by a limited enzyme digestion method for the use in tissue engineering.[Method]Bovine cortical bone, bovine achilles tendon, porcine achilles tendon and porcine skin were splitted into pieces of 0.2-0.5 mm. After being immersed in glacial acetic acid, they were extracted with pepsin. Then the crude products were dissolved, centrifuged, dialyzed and freeze drying to prepare pure collagen type I. The final products were confirmed by absorbance, amino acid analysis and'SDS-PAGE electrophoresis comparing them with the products of Sigma Company.[Result]The wave length of maximum absorbance of the final products was 230 nm, and the amino acid analysis and SDS-PAGE electrophoresis confirmed that the final products were collagen type I. The purity of product extracted from bovine cortical bone was the highest (96.12%) and higher than that from Sigma Company. The extraction rate of bovine achilles tendon collagen was the highest (75.34%).[Conclusion]Collagen type I of higher purity and higher extraction rate can be prepared using a limited enzyme digestion method.And the product from bovine cortical bone is better than the others,which has a promising prospect.
10.OBSERVATION OF ULTRASTRUCTURAL AND ALKALINE PHOSPHATASE LOCALIZATION OF THE PERITUBULAR TISSUE OF SEMINIFEROUS TUBULES IN MAN AND SEVERAL RODENTS

The ultrastructure and AlP localization of the peritubular tissue of seminiferoustubules in man,rabbits,guinea pigs,rats and mice were observed.Usually,there are three layers of the cell which contains slender and branchcytoplasmic processes in human peritubular tissue.Cells of the inner most layerappear to have the characteristic of myoid cells.The outer layers looked the sameas fibroblasts.The myoid cells are frequently separated by wider intercellular spaces.AlP is localized in the basal lamina,innermost layer of collagenous fibrils zone,cytoplasm and inward plasma membrane of the myoid cells.The ultrastructure of the peritubular tissues of the four rodents are basicallythe same,there are only small differences among them.The myoid cells of rabbit andguinea pig are arranged in one layer and the ends of the adjacent cells may beoverlapped,which have a two-layers appearance.The myoid cells of rat and mouseform just one layer surrounding the tubule.The basal lamina in rabbit appears oneto two layers and in the other three rodents is only one layer.There are tightjunctions,open spaces of 100-200 (?) and wider intercellular spaces between theadjacent myoid cells in these four rodents.The AlP is localized in the basal lamina,inner non-cellular layer,cytoplasm and inward plasma membrane of the myoid cellsas well as endothelial cells of lymphatic sinusoid of all four rodents,but theowtward plasma membrane of myoid cells and outer non-cellular layer have theAlP only in guinea pig,rat,and mouse not in the rabbit.The physiological significance of the peritubular tissue and AlP enzymic sheathwere discussed.
