Objective To explore the relationship between serum levels of unconjugated estriol (uE3)and human placental lacto-gen (hPL)and preeclampsia.Methods 123 cases of pregnant women diagnosed as preeclampsia were enrolled into observational group,and 1 50 cases of healthy pregnant women were enrolled into control group.Blood specimen were collected from the two groups in 28-<32 gestational weeks and 32-36 gestational weeks.Serum levels of uE3 and hPL were determined and compared between the two groups.Results The pregnancy outcomes of the control group were better than those of the observation group. With the increase of gestational age,there was increasing trend of serum levels of uE3 and hPL in the two groups.However,com-pared with the observation group,the serum levels of uE3 and hPL were higher in the control group,and the differences were statis-tically significant(P <0.05).Conclusion The decrease of serum levels of uE3 and hPL in pregnant women with preeclampsia may be associated with the pregnancy outcomes.

Induced pluripotent stem cells (iPSC) technology play an important role in studying diseases in vitro. iPSC disease model was based on the iPSC technology, and has achieved important results in many diseases. For those spe-cific genetic diseases that are relatively easy to simulate, iPSC disease model is especially useful. This technology can direct-ly obtained required material from patients, which is free from ethical constraints and rich in cell sources . iPSC disease model as widely used in studying disease mechanism, drug screening, safety inspection, cell therapy, and so on. It is expect-ed to be used in gene correction, gene repair and to obtain the required target cells for the treatment. Cells modification caused by reprogram, lacking of optimum experimental scheme for cell differentiation and difference in genetic backgrounde limit the application of iPSC disease model. More advanced nonintegrated programming technology, improvement of culture conditions, stricter quality control and setting up the standard for iPSC disease model will help to solve these problems.

Objective To assess whether lysyl oxidase like-2 (LOXL2) has a predictive or prognostic value in pulmonary fibrosis mice,and identify the relationship between LOXL2 and transforming growth factor (TGF)-β 1.Methods Male C57BL/6 mice (n=50) were randomly divided into the experimental group (n=40) and controls (n=10).Each mouse in the experimental group was induced by injecting bleomycin into trachea and mice were sacrificed on day 3,7,14 and 28,respectively.The concentrations of LOXL2,TGF-β 1 of the serum and lung homogenates were measured by enzyme-linked immunosorbent assay.One-factor analysis of variance (ANOVA) and nonparametric rank sum test was used for the comparisons between the two groups.Associations between these factors were analyzed by Spearman's test.Results ① The levels of serum and lung homogenates LOXL2 in the experimental group were obviously increased as compared to those in the controls [serum:(56±48) vs (25±23),P＜0.05;lung homogenates:(61±32) vs (22±16),P＜0.05].And in the experimental group,a positive correlation was found between the level of LOXL2 and the radiological index,the pathological score of inflammation and fibrosis.(Serum:r=0.435,0.533,0.335,P＜0.05;lung homogenates:0.675,0.736,0.526,P＜0.05).② The levels of lung homogenates TGF-β 1 in the experimental group were obviously increased as compared to those in the controls [(302±197) vs (64±16),P＜0.05].A positive correlation was found between the level of lung homogenates LOXL2 and the level of lung homogenates TGF-β1 (r=0.520,P＜0.05).Conclusion LOXL2 participates in the pathological process of bleomycin induced pulmonary fibrosis in mice.It predicts pulmonary fibrosis in the preclinical stage and is associated with increased risk for disease progression.The elevated levels of LOXL2 and TGF-β 1 may have some relationship.They both are involved in the disease progression of pulmonary fibrosis.

Objective: To discuss the methods and experience of 64-slice spiral CT which used in infant cases with congenital heart disease, to select the appropriate scan parameters for reducing artifact and the radiation dose. Methods: Thirty-five cases with congenital heart disease were arranged CT examination by using Philips Brilliance 64-slice spiral CT scanner with Blous Tracking techniques. Results: All the images of the heart chambers, pulmonary artery, pulmonary vein, the ascending aorta and the descending aorta contrast agent filling showed good and had clear display. The density of superior vena cava is moderate. The various vascular CT value is approximate, no hardening artifacts. The atrioventricular of Cardiac and vascular of contrast agent filled fully and had clear boundary, no significant artifacts and no effect on the impact reconstruction. Conclusion: Appropriate adjustment of scanning parameters can reduce hardening artifact and the impact of artifacts. Following the principle of as low as reasonably achievable (ALARA) can obtain diagnostic images and reduce radiation damage to the infant.

Objective:To investigate the role of clinical pharmacists in the therapy for a patient with acute promyelocytic leukemi-a. Methods:Clinical pharmacists participated in the treatment process of a patient with acute promyelocytic leukemia complicated with retinoic acid syndrome after tretinoin therapy. The rational treatment plan was put forward according to the literatures,the characteristics of drugs and the general situation of the patient. Results: Promising therapeutic effect was obtained,and the patient was discharged from the hospital. Conclusion:Clinical pharmacists play an active role in improving rational drug use and providing better individual-ized treatment for patients.

CCK correlates with the generation and progression of pancreatic cancer. The research aims to construct eukaryotic expression plasmid pIRES2-EGFP/CCK (CCK pDNA) and transiently express it in COS-7 cells. Total RNA was extracted from porcine intestinal mucosa. RT-PCR was used to amplify the aimed segments CCKcDNA which was then digested with EcoR1 and BamH1 and inserted into a eukaryotic expression plasmid pIRES2-EGFP to construct CCK pDNA. The constructed plasmid was transfected into COS-7 cells by lepofectamin 2000-mediated transfer method. The expression of CCK in transfected COS-7 cells was detected 24, 48 and 72 h post-transfection with fluorescence microscopy and the expression level of CCK mRNA in transfected COS-7 cells was assayed by using RT-PCR. The results showed CCK pDNA was successfully constructed and expressed transiently in COS-7 cells. Green fluorescent protein could be detected in the COS-7 cells transfected with porcine CCK pDNA 24 h post-transfection. At 48th h post-transfection, the number of positive cells was increased significantly and much brighter green fluorescence could be detected. And 72 h post-transfection, the green fluorescence of positive cells became even stronger, while no green fluorescence was detected in the control group. The expression of CCK mRNA in the cells was detectable by using RT-PCR. In COS-7 cells transfected with CCK pDNA a high level of porcine CCK mRNA was detected while no expression of porcine CCKmRNA was found in the cells transfected with null plasmid. It was concluded CCK pDNA was expressed successfully in COS-7 cells, which lays a foundation for further research on the relationship between CCK and tumor.

OBJECTIVE: To summarize the latest research progress in formulations and technologies of transdermal drug delivery system (TDDS) for natural drugs.

Objective To monitor intra-abdominal pressure (IAP) in critically ill patients.Methods A prospective cohort study was conducted.IAP was measured through the bladder technique.Patients were screened for intra-abdominal hypertension (IAH,IAP ≥ 12 mmHg,1 mmHg=0.133 kPa) upon ICU admission.The patients with IAH/abdominal compartment syndrome (ACS) were given appropriate treatment and management for IAH and/or ACS.Mean arterial pressure (MAP),IAP,abdominal perfusion pressure (APP),filtration gradient (FG) and serum creatinine (Cr) were determined in patients with or without IAH,as well as in survivors and non-survivors.Results The entire protocol of IAP measurement was completed in 88 patients.Number of IAH and ACS patients was 25 (28.4％) and 2 (2.3％),respectively.The number of survivors was 80 (90.9％),with 8 (9.1％) non-survivors.Compared with non-IAH patients,IAP and SCr were increased in IAH patients [IAP (mmHg):14.16 ± 2.43 vs.8.13 ± 2.28,t=10.984,P=0.000; SCr (μmol/L):126.72 ± 83.02 vs.73.41 ± 37.59,t=3.087,P=0.005],with a lower FG (mmHg:59.32 ± 17.08 vs.70.24 ± 15.03,t=-2.956,P=0.004).There were no significant differences in MAP and APP between IAH group and non-IAH group [MAP (mmHg):79.18 ± 12.33 vs.88.71 ± 17.34,t=-1.368,P=0.190; APP (mmHg):73.40 ± 16.11 vs.78.37 ± 14.32,t=-1.415,P=0.161].Compared with survivors,non-survivors showed significantly lower APP and FG [APP (mmHg):60.88 ± 14.58 vs.78.56 ± 14.06,t=3.382,P=0.001 ; FG (mmHg):50.38 ± 16.18 vs.68.81 ± 15.44,t=3.208,P=0.002],and higher SCr (μmol/L:129.12 ±83.62 vs.84.36 ± 55.15,t=-2.082,P=0.040).There was no significant difference in IAP and MAP between survivors and non-survivors [MAP (mmHg):71.00 ± 25.46 vs.84.38 ± 13.53,t =1.224,P=0.238 ; IAP (mmHg):10.62 ±5.34 vs.9.76 ± 3.40,t=-0.647,P=0.519].Conclusions Earlier IAP measurements in critically ill patients are essential for the detection of IAH/ACS and renal injury.With appropriate management of IAH/ACS,significant decrease in morbidity and mortality of patients has been achieved.

Objective To observe the role and effect of allitridi to cerebral vasospasm following expe rimental subarachnoid hemorrhage.Methods The models of Japanese rabbits with symptomatic cerebral vasospasm were established by blood injection of twice cisternal magna.Via one side subclavian artery,allitridi was continuously infused by a mini pump.Results After continuous intra arterial infusion allitridi 2 d,the diameter of basilar artery in the treatment group increased more obviously than that before treatment ( P 0.05).There was marked ischemic pathological change in the structure of basilar artery and brain in the control group,however there was no obvious change in the treatment group.Conclusion Allitridi can improve acute cerebral vasospasm;and the continuous intra arterial infusion allitridi has preventive effects in delayed cerebral vasospasm.

Objective To evaluate the effect of procedure for prolapse and hemorrhoid(PPH)for the treatment of prolapsed internal hemorrhoids. Methods 23 patients with the third or fourth degree circular prolapsed internal hemorrhoids were managed by means of PPH. Results The mean operative time was 22 minutes,and hospital stay was 3.5 days after operation.Pain in 48 hours after operation was evaluated and 18 cases had no pain(78 3%),5 cases less pain(21 7%).The anastomoses healed in (4～5) weeks and there was no recurrence in (1～6) months after the patients were discharged from hospital. Conslusions Compared with the traditional operation,PPH has the advantages of simpleness,less pain,less complications and quiker recovery,but the cost is higher.
