1.The Protective Effects of Polypeptides from Chlamys farreri against UVB Radiation

Objective To study the protective effects of th e polypeptides isolated from Chlamys farreri(PCF)against ultraviolet B radiatins.Methods An oxidative damage model of HeLa epi thelial cell line irradiated by ultraviolet B(UVB 7.15?10 -5 J /cm 2 )was established.The HeLa cells were randomly divided into six groups:control group,UVB radiated groups(radiated control,0.5%PCF,1%PCF,2%PCF,1%Vit C).The enzymic activities of HeLa cells were tested by MTT methods,the apoptosis rate an d death rate of HeLa epithelial cells and the content of free calcium in those cells were tested by using flow cytometry(FCM).The contents of MDA and the activities of GSH-Px ,SOD,CAT in the supernatants of cells were determin ed by biochemical methods.Results The results indicated that polypept ides fromChlamys farreri could enhance the activities of GSH-Px,SOD,CA T and decrease the contents of MDAin the su pernatants of HeLa cells,increase t he concentration of cellular free calciumin the cells,and decrease the rates of apoptosis and cell death of the cells .Conclusions The PCF has the protective effects on the HeLa cells irradiated by ultraviolet B.The mechanism of th is effects of PCF may be mediated by e-liminating the free radicals,by increasing the activities of antioxidative enzymes to inhibit the oxidativ e damages caused by UVB .
2.Immunomodulation of polypeptides from Chlamys farreri(PCF) in mice

Objective To investigate the immunomodulation of PCF on the immunosuppressed mice. Methods The immunosuppressed model was established by injection of dexamethasone (DEX) intraperitoneally. Effects of PCF on the immunity were studied by the observation of morphology of immune organ, the analysis of T lymphocytes subsets and the measurement of lymphoproliferative ability. Results PCF could markedly improve the low immunoreaction of immunosuppressed mice caused by DEX. It was showed that the white pulp of spleen was enlarged, the quantity of T lymphocytes of the peripheral blood was increased and the response of lymphoproliferation to ConA was enhanced in the test group compared to those in the model group. Conclusion PCF had the abilities to up-regulate the immunity in vivo,which may be due to its influences on the peripheral mature immunocytes.
3.Aortic arch surgery under mild hypothermic for Standford type A aortic dissection via single upper hemisternotomy approach

Objective:To evaluate the feasibility and safety of total aortic arch surgery under mild hypothermicvia single upper hemisternotomy approach.Methods:From January 2019 to July 2019, 35 patients(31 male and 4 female) with Stanford A type aortic dissection were diagnosed, who were(43.7±5.7)years old. Aortic arch surgeries were carried out under mild hypothermic via single upper hemisternotomy approach and the perioperative mortality, time of cardiopulmonary bypass(CPB), aortic cross clamp(ACC), circulation arrest(CA) and morbidity of neurological dysfunction were respectively were recorded.Results:All patients were finished aortic arch surgery under mild hypothermic single upper hemisternotomy approach, with 8.6% of mortality(3 patients died perioperation). The time of CPB, ACC and CA were respectively(202±53)min, (128±28)min and(8±3)min. There were 6 cases of transient neurological dysfunction(17.1%) and 1 case of permanent neurological dysfunction(2.9%).Conclusion:Aortic arch surgery under mild hypothermic for Standford A dissectionvia single upper hemisternotomy approach is safe and feasible.

5.Comparison of the Genetic Alterations between Primary Colorectal Cancers and Their Corresponding Patient-Derived Xenograft Tissues.

Patient-derived xenograft (PDX) models are useful tools for tumor biology research and testing the efficacy of candidate anticancer drugs targeting the druggable mutations identified in tumor tissue. However, it is still unknown how much of the genetic alterations identified in primary tumors are consistently detected in tumor tissues in the PDX model. In this study, we analyzed the genetic alterations of three primary colorectal cancers (CRCs) and matched xenograft tissues in PDX models using a next-generation sequencing cancer panel. Of the 17 somatic mutations identified from the three CRCs, 14 (82.4%) were consistently identified in both primary and xenograft tumors. The other three mutations identified in the primary tumor were not detected in the xenograft tumor tissue. There was no newly identified mutation in the xenograft tumor tissues. In addition to the somatic mutations, the copy number alteration profiles were also largely consistent between the primary tumor and xenograft tissue. All of these data suggest that the PDX tumor model preserves the majority of the key mutations detected in the primary tumor site. This study provides evidence that the PDX model is useful for testing targeted therapies in the clinical field and research on precision medicine.

6.Study on the diagnostic value of superb microvascular imaging ,advanced dynamic flow and color Doppler flow imaging in breast microcarcinoma and their correlation with pathological microvessel density

Objective To investigate the diagnostic value of superb microvascular imaging ( SM I ) , advanced dynamic flow ( ADF ) and color Doppler flow imaging ( CDFI) in breast microcarcinoma ,and to compare the correlation coefficients between these three indicators and postoperative pathological tumor microvascular density ( M VD ) . Methods A total of 85 patients ( 87 lesions ) with BI‐RADS 4 and the maximum diameter≤1 cm were selected ,of which ,46 lesions were benign ( benign group) and 41 lesions were malignant( malignant group) . All the patients and the corresponding lesions were examined by SM I , ADF and CDFI . Blood flow grading was performed on the images using Adler grade of blood flow ,and the difference of blood flow among the 3 methods was compared . T he expression level of M VD in pathological tumor tissues was detected and analyzed for its correlation with Adler classification by three detection techniques . Results T he areas under the ROC curve( AUR) of CDFI ,ADF and SM I were 0 .694 ,0 .705 and 0 .776 respectively based on the gold standard with pathological diagnosis . T he sensitivity ,specificity , positive predictive value ,negative predictive value ,and accuracy of CDFI were 78 .0% ,54 .3% ,60 .4% , 73 .5% ,and 65 .5% , respectively ; those of ADF were 75 .6% , 60 .9% , 63 .3% , 73 .7% and 67 .8% , respectively ; and those of SM I were 78 .0% ,69 .6% ,69 .6% ,78 .0% and 73 .6% ,respectively . T he Adler grades of CDFI , ADF and SM I were positively correlated with M VD ( P < 0 .05 ) , w hich the highest correlation coefficient between SM I and M VD ( r =0 .430 , P <0 .001 ) . Conclusions SM I is superior to ADF and CDFI in detecting the abundance of breast microcarcinoma , and has the highest correlation coefficient among those 3 detection techniques with tumor pathological M VD ,which indicates that SM I may be used for differential diagnosis of breast microcarcinoma and indirectly evaluate the prognosis of patients .

The sensitivity, specificity, positive predictive value, negative predictive value, and accuracy of CDFI were 78.0%, 54.3%, 60.4%, 73.5%, and 65.5%, respectively; those of ADF were 75.6%, 60.9%, 63.3%, 73.7% and 67.8%, respectively; and those of SMI were 78.0%, 69.6%, 69.6%, 78.0% and 73.6%, respectively. The Adler grades of CDFI, ADF and SMI were positively correlated with MVD(
