1.SF-1 gene mutation and gonad development

Orphan nuclear receptor steroidogenic factor 1 (SF-1) is a member of the nuclear receptor superfamily. It regulates the expression of multiple genes involved in adrenal and gonadal development,steroidogenesis,and reproductive function. Twenty five different mutations have been described and their clinical features are various. Detection of SF-1 gene may be contribute to the etiological diagnosis for 46, XY disorder of sex development patients.
2.Video-assisted Thoracoscopic Surgery for Anterior Mediastinal Nodules:Report of 32 Cases

Objective To investigate the feasibility of video-assisted thoracoscopic surgery for anterior mediastinal modules. Methods Thoracoscopic surgery was performed with the patients in supine position and the thoracoscope set at 30?. Three trocars were used during the procedure. One of the trocars was inserted through the fifth or sixth intercostal space at the midaxillary line for direct observation. And the other two were placed at the second or third intercostal space at the midclavicular line,and the fifth intercostal space at the anterior axillary line. The nodules were removed directly during the operation. For inseparable nodules,the whole thymus and the fat tissues in the anterior mediastinum were removed all together. Results The procedure was completed under a thoracoscope in all of the 32 cases. Small assistant incision was made in 3 cases because of hemorrhage in the chest cavity. Postoperative pathological examination showed thymus tumor in 14 cases,hyperplasia of the thymus in 3,thymic cyst in 5,bronchial cyst in 3,pericardial cyst in 2,neurilemmoma in 2,lymph node in 2,and angioma in 1. The accordance rate of pre-and post-operative diagnosis was 68.8% (22/32). Postoperative complications were found in two patients,including one case of pericardial effusion,and one case of lung infection. Both the cases were cured afterwards. Conclusions Thoracoscopic surgery is a minimally invasive procedure for the treatment of anterior mediastinal nodules. It is optimal for young patients and cases with low lung function,as it causes less pain,quicker recovery,and satisfying cosmetic outcomes.
3.Diagnostic value of combination detection of CEA,CYFR21-1,NSE in bronchoalveolar lavage fluid for early lung cancer

Objective To explore the diagnostic value of combination detection of carcino‐embryonic antigen(CEA ) ,cytokeratin 19 fragment 21‐1(CYFR21‐1) ,neuron‐specific enolase(NSE) in bronchoalveolar lavage fluid (BALF) for early lung cancer and its correlation with clinical effects .Methods 69 cases of lung cancer were included into the lung cancer group and 50 cases of benign pulmonary disease were included into the lung benign disease group .All the patients were given bronchoalveolar lavage(BAL) .The chemiluminescence immunoassay was adopted to detect the levels of CEA ,CYFR21‐1 and NSE in BALF .The changes of tumor markers levels were compared between the two groups and among different clinical curative effects in the patients with lung cancer . Results The levels of CEA ,CYFR21‐1 and NSE in BALF of the lung cancer group were significantly higher than those of the be‐nign pulmonary disease group ,the difference was statistically significant (P < 0 .05) .The levels of tumor markers in the stage Ⅱ of lung cancer were higher than those in the stage Ⅰ of lung cancer ,the difference was statistically significant (P< 0 .05) ;the levels of CEA ,CYFR21‐1 and NSE in BALF were gradually increased with the decrease of clinical effect (P< 0 .05) ;the sensitivity and spe‐cificity of the combined detection of three tumor markers were 62 .3% and 82 .0% respectively ,which were significantly higher than those of any single tumor marker (P< 0 .05) .Conclusion The levels of CEA ,CYFR21‐1 and NSE in BALF of early lung cancer patients are obviously increased ,moreover which is closely correlated with the pathological stage of lung cancer ,the combined detec‐tion could contribute to improve the detection rate of early lung cancer and guide the evaluation of clinical effect .
4.Protective effect of vitamin E on rat liver mitochondrial respiratory dysfunction induced by lipid extract of burn eschar

Lipid extract of burn eschar (D_1) added to isolated rat liver mitochondrialcaused depression of respiratory control ratio (RCR), ADP/O ratio, rate of ATP produc-tion, respiration rate in state 3 but it stimulated the respiration rate in state 4. Pretreat-ment with vitamin E significantly prevented the inhibition of mitochondrial respiratoryfunction caused by D_1. The addition of vitamin E after that of D_1 had no protective ef-fect. Heat-treatment of D_1 markedly reduced its inhibitory effect on mitochondrial respira-tory function. Cumene hydroperoxide (CHP) markedly depressed mitochondrial respiration.Vitamin E caused a significant increase in RCR, which was inhibited by CHP at lowerconcentrations. Vitamin E reduced MDA formation considerably in normal mitochondriaand in those mitochondria incubated with D_1, heated D_1 or CHP. These findings indicatedthat D_1 -and CHP-induced depression of mitochondrial respiratory function was probablydue to lipid peroxidation damage which was proved by the protective effct of the pota-tive antioxidant vitamin E.
5.Difference in antimicrobial susceptibility of common clinical pathogens to different ratios of cefoperazone/sulbactam antimicrobial disks

Objective To understand the antimicrobial resistance of common clinical pathogens to antimicrobial disks containing different ratios of cefoperazone/sulbactam,so as to provide basis for rational application of cefoper-azone/sulbactam in clinic.Methods 1 141 pathogens isolated from clinical specimens in a hospital in the first half year of 2014 were collected,disk diffusion method was adopted to detect antimicrobial activity of two kinds of cef-operazone/sulbactam disks (70/35 μg and 75/75 μg).Results Of 1 141 pathogenic strains,675 (59.16%)were En-terobacteriaceae,447 (39.18%)were non-fermentative bacteria,and 19 (1 .66%)were other gram-negative bacilli. Resistance rates of pathogens to 70/35μg and 75/75 μg cefoperazone/sulbactam antimicrobial disks were as follows:extended-spectrumβ-lactamases(ESBLs)-producing Escherichia coli (n=221)were 7.69% and 2.26% respective-ly,ESBLs-producing Klebsiella pneumoniae (n=92)10.87% and 3.26% respectively,imipenem-resistant Acineto-bacter baumannii (IRAB,n=295)54.92% and 11 .19%respectively;there were significant differences in antimicrobial activity between two ratios of antimicrobial disks(P <0.05).While antimicrobial resistance rates of ESBLs-negative Enterobacteriaceae (Escherichia coli ,n = 135;Klebsiella pneumoniae ,n =98),imipenem-sensitive Acinetobacter baumannii (ISAB,n=51),Pseudomonas aeruginosa (n=48 ),and Stenotrophomonas maltophilia (n=22)were not significantly different (all P >0.05).Conclusion Antimicrobial activity of two different ratios of cefoperazone/sulbactam antimicrobial disks to ESBLs-producing Enterobacteriaceae and IRAB is different,attention should be paid to ratios of cefoperazone/sulbactam during the treatment ,so as to achieve the desired therapeutic effect.
6.Effects of Astragalus injection on proliferation of basal-like breast cancer cell line MDA-MB-468

OBJECTIVE: To investigate the effects of Astragalus injection (AI) on basal-like breast cancer cell line MDA-MB-468 and murine bone marrow stromal stem cells (mMSCs). METHODS: MDA-MB-468 cells and primary cultured mMSCs were treated by different concentrations of AI, and with untreated MDA-MB-468 cells as blank control. The morphology of cells was observed by phase-contrast inverted microscope and transmission electron microscopy. Cytotoxic effects of AI on MDA-MB-468 cells and mMSCs were evaluated by cell counting kit-8 (CCK-8) assay. Cell cycle and apoptosis of MDA-MB-468 cells induced by AI were measured by flow cytometry. Lactate dehydrogenase (LDH) activity in supernatants was measured by enzymatic colorimetric method. The expressions of epidermal growth factor receptor (EGFR) and p53 protein in MDA-MB-468 cells were evaluated by streptavidin-biotin-peroxidase complex method. RESULTS: A time-dependent cytotoxic effect of 1 g/ml AI was observed in MDA-MB-468 cells. 1 g/ml AI also had cytotoxic effect on mMSCs, but its effect was not better than cisplatin. 0.1 g/ml AI could promote the proliferation of mMSCs. Different concentrations of AI could all induce the apoptosis of MDA-MB-468 cells. There was no significant difference in LDH activity in the supernatants between blank control group and AI-treated and cisplatin-treated groups. AI could down-regulate the expressions of EGFR and p53 protein. CONCLUSION: The effects of AI on MDA-MB-468 cells and mMSCs are related to the concentration of AI, and its mechanism of inhibiting the proliferation of MDA-MB-468 cells may be due to down-regulation of the expressions of EGFR and p53 protein.
7.Effect of absorbable poly-D,L-lactide acid rod on development of rabbit epiphyseal plate

0.05).Both femur bone lengths in 3.2mm group were shorter than those in the 1.5 mm and 2.2 mm groups.No abnormalities of cartilage cells were observed,without obvious histological evidence of inflammation;only nonspecific foreign body reaction and macrophage infiltration were noticed.Conclusion: PDLLA rod itself does not cause any disturbance to the development of the epiphyseal plate.The growth disturbance of epiphyseal plate depends on the area of injured epiphyseal plate;a injured area over 7% of epiphyseal plate may cause the growth disturbance of epiphyseal plate.
8.Optimization of extraction method of total flavones in Lonicera japonica by orthogonal design

Objective To optimize and establish the extraction of total flavones in Lonicera japonica. Methods The ultraviolet spectrophotometry was used to determinate the total flavonoids. The effect of extraction time, the amount of solvent and extraction times,pH were tested by the orthogonal design.ResultsThe total flavonoids showed a linear relationship,the optimal extraction condition of total flavones in Lonicera japonica was with 16 times volume of sample weight water for three times, each time was 2 h.Conclusions The method is simple and applicable to the extraction of total flavones in Lonicera japonica.
9.Effect of leptin on nerve function in patient with Parkinson disease

Objective To investigate effect of leptin on nerve function in patient with Parkinson disease .Methods 68 cases patients with PD were selected and randomly divided into two groups, according to random number table, from October 2013 to December 2014 in the hospital.34 cases in control group were treated with conventional therapy, and 34 cases in experimental group were treated with leptin, with a consecutive treatment of twelve weeks.The score of activity of daily living scale ( ADL ) and mini-mental state examination ( MMSE ) were analyzed pre and post-treatment. ResuIts After treatment, the ADL score of experimental group was significantly lower than that of control group ( P<0.05 ) , MMSE score of experimental group was significantly higher than that of control group (P<0.05).The adverse reactions in both groups were minor.ConcIusion Leptin has good curative effect in treatment of Parkinson disease, and could significantly improve the neurological function of patients, with mild adverse reactions.
10.Relationship between osteopontin and hepatocellular carcinoma

In recent years,it has been reported that the osteopontin attracts great attention for its roles in tumor growth and metastasis.In many tumors,osteopontin plays an important role in promoting tumor growth,metastasis and enhancing adhesion and migration abilities.In particular,in hepatocellular carcinoma,osteopontin has very satisfactory sensitivity and specificity in early diagnosis and is relatively effective on prognosis prediction.Suppression osteopontin expression will inhibit liver cancer growth and metastasis,indicating that the osteopontin stays a new target for liver cancer treatment and possesses intangible clinical potential.
