1.Generation of Highly Expandable Intestinal Spheroids Composed of Stem Cells

Many of early findings regarding intestinal stem cells (ISCs) and their niche in the human intestine have relied on colorectal cancer cell lines and labor-intensive and time-consuming mouse models. However, these models cannot accurately recapitulate the physiologically relevant aspects of human ISCs. In this study, we demonstrate a reliable and robust culture method for 3D expanding intestinal spheroids (InSexp ) mainly comprising ISCs and progenitors, which can be derived from 3D human intestinal organoids (HIOs). We did functional chararcterization of InSexp derived from 3D HIOs, differentiated from human pluripotent stem cells, and optimization culture methods. Our results indicate that InSexp can be rapidly expanded and easily passaged, and show enhanced growth rates via WNT pathway activation. InSexp are capable of exponential cell expansion and cryopreservation. Furthermore, in vitro-matured HIO-derived InSexp proliferate faster than immature HIO-derived InSexp with preservation of the parental HIO characteristics. These findings may facilitate the development of scalable culture systems for the long-term maintenance of human ISCs and provide an alternative platform for studying ISC biology.
2.Association of Bone Densities with Anthropometric Indices and Lifestyles in Elderly People.

This study was performed to investigate the relationships among bone mineral densities (BMD), anthropometric data and lifestyle factors in the elderly. Subjects included 138 elderly (male: 38, female: 100) aged over 65 years, who were home-dwelling in a low-income area of Puchon City. The BMDs of the lumbar spines (LS), femoral necks (FN), Ward's triangles (WT) and trochanters (TC) were measured by dual energy X-ray absorptiometry. The females showed significantly lower BMDs in four sites (p < 0.0001). The elderly aged over 75 revealed significantly more decreased femoral BMDs than the elderly aged 65 to 74. Female with BMIs of 20 to 25, showed significantly higher BMDs in LS, FN and trochanter than those with BMIs of less than 20. However, males displayed significantly higher BMDs in only LS, with increasing BMIs. THe BMDs of LS correlated with weights (r = 0.543, p < 0.001), heights (r = 0.477, p < 0.001), upper arm circumferences (r = 0.368, p < 0.01), waist circumferences (r = 0.367, p < 0.001), subscapular skinfold thicknesses (r = 0.363, p < 0.001) and hip circumferences (r = 0.231, p < 0.01). Non-smokers and non-drinkers showed significantly higher BMDs in trochanters only in the case of the males. Female milk-drinkers showed significantly elevated LS BMDs. Eighteen percent of the males were assessed as having osteoporosis, as compared to fifty percent of the females. Ninety-three percent of the females and 81.6% of the males responded that they often or always had "difficulty in standing for a long time".

Species of the genus Aspergillus have a variety of effects on humans and have been considered industrial cell factories due to their prominent ability for manufacturing several products such as heterologous proteins, secondary metabolites, and organic acids. Scientists are trying to improve fungal strains and re-design metabolic processes through advanced genetic manipulation techniques and gene delivery systems to enhance their industrial efficiency and utility. In this review, we describe the current status of the genetic manipulation techniques and transformation methods for species of the genus Aspergillus. The host strains, selective markers, and experimental materials required for the genetic manipulation and fungal transformation are described in detail. Furthermore, the advantages and disadvantages of these techniques are described.
5.Unveiling the Functions of the VosA-VelB Target Gene vidD inAspergillus nidulans

The velvet regulators VosA and VelB are primarily involved in spore maturation and dormancy. Previous studies found that the VosA-VelB hetero-complex coordinates certain target genes that are related to fungal differentiation and conidial maturation in Aspergillus nidulans. Here, we characterized the VosA/VelB-inhibited developmental gene vidD in A. nidulans.Phenotypic analyses demonstrated that the vidD deleted mutant exhibited defect fungal growth, a reduced number of conidia, and delayed formation of sexual fruiting bodies. The deletion of vidD decreased the amount of conidial trehalose, increased the sensitivity against heat stress, and reduced the conidial viability. Moreover, the absence of vidDresulted in increased production of sterigmatocystin. Together, these results show that vidD is required for proper fungal growth, development, and sterigmatocystin production in A. nidulans.

Purpose:
This study examined the diet quality and its relationship with risk factors for diabetes complications and the related markers in Korean adults with type 2 diabetes (T2D).
Methods:
This study analyzed 1,774 adults aged 31–79 years with T2D who participated in the 8th Korea National Health and Nutrition Examination Survey. The diet quality was examined by the Korean Healthy Eating Index (KHEI), and the dependent variables were the risk factors for diabetes complications (overweight/obesity, hyperglycemia, dyslipidemia, and hypertension) and related markers (HbA1c, fasting plasma glucose [FPG], triglycerides, low-density lipoprotein-cholesterol, high-density lipoprotein-cholesterol [HDL-C], systolic blood pressure, and diastolic blood pressure). The data were analyzed using SPSS v. 29.0.
Results:
The total KHEI scores were Q1 45.80, Q2 58.98, Q3 67.91, and Q4 77.90 in men and Q1 48.96, Q2 60.79, Q3 69.62, and Q4 80.34 in women. The odds ratio [OR]s of the adverse level HDL-C were significant for men in KHEI Q1 (OR, 1.844; 95% confidence interval [CI], 1.115–3.052), Q2 (OR, 1.777; 95% CI, 1.085–2.911), and Q3 (OR, 1.982; 95% CI, 1.220–3.220) compared to those in Q4. The KHEI ‘have breakfast’ score was inversely and significantly associated with the adverse level FPG in men (OR, 0.942; 95% CI, 0.891–0.996) and women (OR, 0.935; 95% CI, 0.883–0.990). The ORs of dyslipidemia were significant for men in KHEI Q2 (OR,2.026; 95% CI, 1.179–3.483) and Q3 (OR, 1.817; 95% CI, 1.062–3.110) compared to those in Q4.
Conclusion
A good diet quality can help with the proper control of HDL-C in men with T2D, and having breakfast can help control the FPG level in people with T2D.
