Objective To investigate the effect of melatonin (MT) on the proliferation and apoptosis in human pancreatic cancer cell SW1990 in vitro.Methods SW1990 cell line were treated with MT at different concentrations (0.1,0.5,1.0,2.5 and 5.0 mmol/L) and at different time points (24 h,48 h and 72 h).Cell proliferation was evaluated by MTT and apoptosis was determined by AnnexinV/Pl,cell cycle was determined by flow cytometry,and Western Blot was used to detect the expression of Bcl-2 and Bax.Results MT could inhibit the proliferation of SW1990 cell in a time and dose dependent manner.48 h after 0.1～5.0 mmol/L MT treatment,the proliferation inhibitory rate was 7.4%～85.8%,the proportion of SW1990 cell in phase G0/G1 was 72.6%～85.33%.48 h after 1～5.0 mmol/L MT treatment,the apoptosis rate was 21.5% ～41.7%,and the expression of Bcl-2 was down-regulated and the ratio of Bcl-2/Bax decreased.Conclusions MT could inhibit the proliferation of SW1990 pancreatic cancer cells by up-regulating the expression of Bax and down-regulating the expression of Bcl-2,as well as enhancing apoptosis and blocking SW1990 cell in phase G0/G1.

Objective:To investigate the relationship of the mRNA expression of BRCA1 andβ-tubulinⅢwith docetaxel-resistance in esophageal cancer. Methods:The genes BRCA1 andβ-tubulinⅢwere determined at the mRNA level using RT-qPCR in 36 esophageal carcinoma specimens. Results:The mRNA expression of BRCA1 andβ-tubulinⅢwas determined in the 36 tumor samples using RT-qPCR. The median BRCA1 mRNA expression level in relation to that ofβ-actin was 6.27. The medianβ-tubulinⅢmRNA expression level in relation to that ofβ-actin was 4.44. The patients were divided into two groups using these cutoff values. The BRCA1 mRNA expression level was not correlated with the sensitivity of esophageal cancer patients to docetaxel (P=0.733). The response rate of the tumors with highβ-tubulinⅢexpression was (38.9%), which is significantly lower than in patients with lowβ-tubulinⅢexpression (83.3%) (P=0.015). Conclusion:Theβ-tubulinⅢexpression levels in the tumor tissues were probably an important biomarker for the efficacy of docetaxel chemotherapy in esophageal cancer patients. Our study may provide new insights into taxane chemotherapy for advanced esophageal cancer patients.

Objective To analyze the molecular characterization of PrM/C and E genome of Japanese encephalitis virus(JEV) strain,CQ11-66,a newly strain isolated from patients with epidemic encephalitis B Chongqing Municipal.Methods The samples were collected from Children's Hospital of Chongqing Medical University,and inoculating BHK-21 cells were used to detect and isolate the Japanese encephalitis virus(JEV) strain,computer analysis of the phylogenetic,nucleic acid data and deduced amino acid sequence was accomplished using the Clustal X(1.8) and MEGA5 programs.Results Only one JEV strain was isolated from patient's cerebrospinal fluid specimen,named CQ11-66.Comparison of the PrM/C genome sequence of strain CQ11-66 with other 31 JEV isolates showed a 74.8％-97.4％ nucleotide sequence homology among them,which resulted in 85.6％-98.7％ amino acid sequence homology; Meanwhile,comparison of the E genome sequence of strain CQ11-66 with other 35 JEV isolates showed a 81.6％-99.6％ nucleotide sequence homology among them,which resulted in 94.8％-99.6％ amino acid sequence homology.There were high homology between CQ11-66 and JEV isolates from Fujian province on nucleotide sequence and amino acid sequence.Phylogenetic analysis of PrM/C and E genome showed that the CQ11-66 belonged to genotype Ⅲ.Conclusion Only one JEV strain was isolated from patient's cerebrospinal fluid specimen.There were some differences between CQ11-66 strain and other JEV isolates,and CQ11-66 strain belonged to genotype Ⅲ.

Objective: To study the preoperative blood glucose management model and its implementation effect in the multidisciplinary team of ophthalmology under the background of diagnosis related groups (DRGs). Methods: A total of 170 patients underwent vitrectomy, vitreous injection, and cataract extraction surgery were enrolled in the CS03Z and CS09A groups from January 1, 2018 to December 31, 2018. The cases from January 1, 2018 to June 31, 2018 were in the control group, accounting for 76 cases, and routine blood glucose management was performed. The cases from July 1, 2018 to December 31, 2018 were observed in the observation group, and 94 cases were implemented. The multidisciplinary blood glucose management model was implemented. The average hospitalization days, average hospitalization costs, preoperative blood glucose management quality, and hospitalization satisfaction were compared between the two groups. Results: In the control group, the average hospitalization cost of the three operations (PPV/IV/Phaco group) was (13 949.71±2 099.36) yuan, (4 933.22±2 269.33) yuan, (6 521.16±1 006.40) yuan, the average hospitalization cost of the three operations of the observasion group was (12 937.37±1 447.33) yuan, (2 649.53±1 105.92) yuan, (5 315.76±1 037.02) yuan, the difference between the two groups was statistically significant (t=2.266, 3.818, 4.074, all P<0.05 or 0.01) . The average hospitalization day of the 3 operation group was (9.98±3.91) d, (9.63±5.46) d, (7.65±3.88) d, and the observation group was (6.37±3.54) d, (3.97±2.29) d, (4.17±2.23) d,the difference between the two groups was statistically significant (t=3.980, 3.979, 3.632, all P<0.01). The preoperative blood glucose accident rate in the control group was 14.47% (11/76), The observation group was 2.13% (2/94), the difference between the two groups was statistically significant (χ²=11.642, P<0.01). The surgical delay rate of the control group was 2.63% (2/76) ; the surgical delay rate of the observation group was 0(0/94), the difference between the two groups was statistically significant (χ²=7.045, P<0.01) .The nurse blood glucose management of the control group was 66.67% (24/36) , and that in the second half was 97.22% (35/36) ,the difference between the two groups was statistically significant (χ²=9.005, P<0.01). The satisfaction rate of the patients in the control group was 82.89% (63/76), and that of the observation group was 94.68% (89/94). the difference between the two groups was statistically significant (χ²=6.166, P<0.05). Conclusion Under the background of DRGs, the preoperative multidisciplinary blood glucose management team with clinical pathway as the core can effectively reduce the average hospitalization cost and average hospitalization day, improve the quality of blood glucose management, and improve the hospitalization satisfaction of the patients.

N-acetyl-L-cysteine (NAC) capped quantum dots (QDs) were synthesized by a hydrothermal method and coated with 2-amino-2-deoxy-D-glucose (DG), polyethylene glycol (PEG), and 9-D-arginine (9R). The optical properties, morphology and structure of 9R/DG-coated CdTe QDs were characterized by ultraviolet-visible spectrometry, fluorescence spectrum, Fourier transform infrared (FTIR), proton nuclear magnetic resonance (1H NMR), liquid chromatography-mass spectrometer (LC-MS), sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and transmission electron micrographs (TEM). Furthermore, the biocompatibility, tumor targeted ability and transmembrane action of 9R/DG-coated CdTe QDs were studied. Results indicated that 9R/DG-coated CdTe QDs was constructed successfully by ligand exchange. The 9R/DG-coated CdTe QDs with the size of 8-10 nm had good dispersity and the absorbance and fluorescence peaks of CdTe QDs after modification were red shifted from 480 nm to 510 nm and 627 nm to 659 nm, respectively. In addition, the CdTe QDs modified by PEG, DG and 9R displayed good biocompatibility, high targeted ability to the cancer cells with glucose transporter type 1 (GLUT1) receptor high expression and obvious transmembrane ability.

Objective To compare different beginning time of acid simulation to reduce the acute damage of salivary glands after high-dose of iodine-131 treatment for the post-surgery patient with differentiated thyroid cancer (DTC) and screen out the optimal beginning time of acid simulation .Methods Total 309 cases of post-surgery DTC patients accept high-does ioding-131 treatment (average dose of ioding-131 :4 .28 ± 0 .43 GBq) ,and all patients was divided into three group randomly ,the beginning time of acid stimulation(vitamin C :10 mg per time ,three time a day ,lemonade:50 mL per 2 h) for each group is 2 ,12 and 24 h .Then observe the incidence and time of acute damage of salivary glands for each group .At same time ,we analyses the relation between acute dam-age of salivary glands with sex and age .Results The incidence of acute damage of salivary glands is 13 .21% (2 h) ,24 .51% (12 h) , 26 .73% (24 h)respectively ,the incidence of 2 h is lower than those of 12 h and 24 h obviously(P<0 .05) ,and there is no difference between 12 h and 24 h(P>0 .05) .The occurrence time of acute damage of salivary glands for 84 .85% patients is between 10-24 h .And there is no difference of incidence of acute damage of salivary glands between different sex and age group .Conclusion 2 h maybe the optimal beginning time of acid simulation to reduce the acute damage salivary glands for the post-surgery DTC patients after high-dose iodine-131 treatment in this study .Sex and age are no influence to the occurrence of acute damage of salivary glands .

Objective To establish a novel direct radio-labeling method for 99Tcm-IgG and evaluate the biologic distribution of 99Tcm-IgG.Methods IgG protein was modified with 2-mercaptoethanol.Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE),UV-vis spectrophotometer,matrix-assisted laser desorption ionization-time of flight (MALDI-TOF) were used to identify the concentration,integrity of the modified protein.Then radiolabeled IgG-SH with 99TcmO4-was analyzed with radio-HPLC.The product was prepared as frozen kits.The distribution and metabolic process of 99Tcm-IgG were observed in New Zealand white rabbits.Results The relative molecular mass of IgG-SH and IgG measured by SDA-PAGE were similar.The relative molecular mass measured by MALDI-TOF was 1.47× 105.The radiolabeling yield was over 95％,the specific activity was 1.7× 105 GBq/mmol.All the radioactive conjugates of 99Tcm-IgG showed excellent stability in vitro.And more than 95％ conjugates retained their original structures for 6 h in 5％ BAS.Gamma imaging in New Zealand white rabbits showed blood retention in first 4 h after injection,and prominent uptake of radiotracers in the liver,kidneys,and urinary bladder at 24 h after injection,which indicated that 99Tcm-IgG was excreted mainly through the renal route.99Tcm-IgG kept its original biological activity after the modification.Conclusions Direct radio-labeling of 99Tcm-IgG was successfully established.The methods may be useful for radio-modification of monoclonal antibody.

Objective To investigate the feasibility of ultrasound-guided percutaneous drainage for the treatment of peripancreatic abscess. Methods The clinical data of 36 patients with peripancreatic abscess who were admitted to the General Hospital of the Chengdu Military Command were retrospectively analyzed. All the puncture sites were designed according to the region, range and shape of the abscess, and then the angle and the direction of the needle penetration were determined according to the spatial relationship between the puncture site and the abscess. Finally, the drainage tubes were placed under the guidance of the ultrasound. Results The technique was successfully performed on all the patients, and 33 patients were cured with the cure rate of 92%.The mean healing time was 37 days. Three patients were converted to laparotomy because of the unsatisfied therapeutic effects. Enterocutaneous fistula was detected in 3 patients after the surgery and they were cured after receiving nonoperative management. All patients were followed up for 3-48 months, and neither residual abscess nor recurrence was detected. Two patients were complicated with type one diabetes, one with dyspepsia, two with gallstone, and they were cured by symptomatic treatment. The body weights of 27 patients were increased compared to those before the operation. Conclusion Ultrasound-guided percutaneous drainage is effective for the treatment of peripancreatic abscess.

The response of glueagon to the change of glucose and its relation to blood pressure in 71 eidely patients with type 2 diabetes was investigated. The results showed that the postprandial increment of glucagon in the group of patients with postprandial 2h plasma glucose increment<2.5 mmol/L was significantly higher than that in the group with plasma glucose inerement≥ 2.5 mmoi/L (P<0. 05). The postprandial increment of glucagon in patients with normal blood pressure was significantly higher than that in patients with hypertension (P<0.05). The results suggest that the decreased response of glucagon to the change in plasma glucose in elderly patients with type 2 diabetes is related to increased blood glucose and high blood pressure.
