Objective To investigate the effect of melatonin (MT) on the proliferation and apoptosis in human pancreatic cancer cell SW1990 in vitro. Methods SW1990 cell line were treated with MT at different concentrations (0.1,0.5,1.0,2.5 and 5.0 mmol/L) and at different time points (24 h,48 h and 72 h).Cell proliferation was evaluated by MTT and apoptosis was determined by AnnexinV/Pl,cell cycle was determined by flow cytometry, and Western Blot was used to detect the expression of Bcl-2 and Bax.Results MT could inhibit the proliferation of SW1990 cell in a time and dose dependent manner.48 h after 0.1~5.0 mmol/L MT treatment, the proliferation inhibitory rate was 7.4%~85.8%,the proportion of SW1990 cell in phase G0/G1 was 72.6%~85.33%.48 h after 1~5.0 mmol/L MT treatment, the apoptosis rate was 21.5% ~41.7%,and the expression of Bcl-2 was down-regulated and the ratio of Bcl-2/Bax decreased. Conclusions MT could inhibit the proliferation of SW1990 pancreatic cancer cells by up-regulating the expression of Bax and down-regulating the expression of Bcl-2,as well as enhancing apoptosis and blocking SW1990 cell in phase G0/G1.

Objective To explore the common bleeding location, disease features, and the management strategies of intractable aged epistaxis. Methods 134 serious epistaxis patients were examined step by step according to nasal anatomic structure and treated by a stepwise way under endoscopy. Results The bleeding location of the intractable aged epistaxis were found as follows:2 cases (1. 49%) in anterosu-perior area, 44 cases (32. 84%) in anteroinferior area, 18 cases (13. 43%) in posterosuperior area, 22 cases (16. 42%) in posteroinferi-or area, 3 cases (2. 24%) in nasopharynx, and the bleeding site of the other 45 cases (33. 58%) were not found. All the patients were healed. The final treatment way were:46 cases (34. 33%) with pharmacotherapy or cauterization (grade Ⅰ~Ⅲ), 88 cases (65. 67%) with anterior nasal packing (grade Ⅳ), 9 cases (6. 72%) with posterior packing(gradeⅤ),1 case (0. 75%) with selective angiographic embolization (grade Ⅵ). There were 82 cases (61. 19%) succeed with the initial hemostasis methods while 52 cases (38. 81%) succeed with the upgrade therapy. Both hemostasis methods and upgrade therapy had statistical significance in different nasal position groups ( hemo-stasis methods:χ2 =16. 35,P=0. 00; upgrade therapy: χ2 =16. 35,P=0. 00). Conclusion Compartmental examination and classified treatment steps by using endoscope may locate and stop nose bleeding promptly while decrease patients' pain and medical cost.

Objective Recently, the role of TREK-1 in anti-depression and neuroprotection was demonstrated definitely. In this paper,a systematic review about the functional structure of TREK-1 as well as the underlying mechanisms which involved in depression resistance and nerve protection was made for purpose of providing new ideas for the treatment of clinical diseases. Methods The electronic search of Medline/PubMed,CNKI,CBM and Wanfang Data were performed to collect the introduction of TREK-1 and the studies of depression and nerve protection in which TREK-1 participated from inception of each database to October 2015 ,besides the relevant references of the included studies were also retrieved. Results 30 studies were included in this paper.14 studies were about the interpretation of TREK-1's structure, distribution and the regulatory factors, and the number of researches regarding the involvement of TREK-1 in anti-depression and neuroprotection was 13 and 7 respectively.Based on these studies,it was easily to conclude that inhibition or blocking of TREK-1 channels can produce obvious antidepressant effect, however, activation of TREK-1 channels will induce the dilation of local cerebral vascular and silence hyperactive neurons. Conclusion Some achievements about the antidepressant and neuroprotective effect of TREK-1 channels in vivo have been made gratifyingly, thus future research should focus on exploring the best TREK-lactivation ratio in different brain regions to gain the best balance between anti-depression and neuroprotection.

OBJECTIVE:To explore the therapeutic effect of early use of alprostadil and the effects on PTX-3,renal function and stability of carotid plaque in patients with acute cerebral infarction. METHODS:93 patients with acute cerebral infarction in our hospital from Aug. 2011 to Aug. 2015 were selected, and divided into observation group(47 cases,2 case withdrew from the test and 45 cases completed the test)and control group(46 cases,1 case withdrew from the test and 45 cases completed the test) by odd and even number of registration order. Control group was given Citicoline sodium injection 0.5 g + Extract of ginkgo biloba leaves injection 20 ml added into Sodium chloride injection(NS)250 ml,ivgtt,qd;observation group was additionally given Al-prostadil injection 20 μg added to NS 250 ml,ivgtt,qd. They were treated for 2 weeks. Clinical efficacy, and the differences of PTX-3,blood urea nitrogen(BUN),serum creatinine(SCr),24 h urinary protein(Upro),stability of carotid plaque before and after treatment, and the incidence of adverse reactions in 2 groups were observed. RESULTS:The total effective rate in observation group(91.1%)was higher than control group(73.3%),the difference was statistically significant(P<0.05). There was no statisti-cal significance in PTX-3,BUN,SCr,24 h Upro between 2 groups before treatment (P>0.05). After treatment,PTX-3,BUN, SCr,24 h Upro in 2 groups decreased, and observation group was lower than control group;stability of carotid plaque in observa-tion group(100.0%)was higher than control group(64.0%),the differences were no statistically significant(P<0.05). The ad-verse reactions in 2 groups relieved themselves with no special treatment, and with no statistical significant(P>0.05). CONCLU-SIONS:The early use of alprostadil is effective for acate cerebral in farction, and can obviously reduce PTX-3 and improve renal functions, increase stability of carotid plaque with good safety.

Objective To analyze the molecular characterization of PrM/C and E genome of Japanese encephalitis virus(JEV) strain,CQ11-66,a newly strain isolated from patients with epidemic encephalitis B Chongqing Municipal. Methods The samples were collected from Children's Hospital of Chongqing Medical University, and inoculating BHK-21 cells were used to detect and isolate the Japanese encephalitis virus(JEV) strain, computer analysis of the phylogenetic, nucleic acid data and deduced amino acid sequence was accomplished using the Clustal X(1.8) and MEGA5 programs. Results Only one JEV strain was isolated from patient's cerebrospinal fluid specimen, named CQ11-66.Comparison of the PrM/C genome sequence of strain CQ11-66 with other 31 JEV isolates showed a 74.8%-97.4% nucleotide sequence homology among them, which resulted in 85.6%-98.7% amino acid sequence homology; Meanwhile, comparison of the E genome sequence of strain CQ11-66 with other 35 JEV isolates showed a 81.6%-99.6% nucleotide sequence homology among them, which resulted in 94.8%-99.6% amino acid sequence homology. There were high homology between CQ11-66 and JEV isolates from Fujian province on nucleotide sequence and amino acid sequence.Phylogenetic analysis of PrM/C and E genome showed that the CQ11-66 belonged to genotype Ⅲ.Conclusion Only one JEV strain was isolated from patient's cerebrospinal fluid specimen. There were some differences between CQ11-66 strain and other JEV isolates, and CQ11-66 strain belonged to genotype Ⅲ.
