1.Significance of detecting the mutations of K-ras gene in faecal for diagnosis colorectal carcinoma

Objective To study the significance of detecting the mutations of K-ras gene in faecal for diagnosis colorectal carcinoma.Methods 36 cases suffering from colorectal carcinoma,24 cases from colorectal benign diseases and 24 cases from healthy subjects,all cases had been diagnosed by enteroscope. K-ras gene mutation at the codon 12 was detected by PCR-RFLP,DNA was taken from faecal in modified method.Results K-ras mutation rate in colorectal carcinoma was 77.8 %,colorectal benign diseases was 25.0 %,healthy cases was 8.3 %.Conclusion Detecting the mutations of K-ras gene in faecalmay improve the rate of diagnosing colorectal carcinoma.
2.Effect of Xiang-Qi-Tang on coagulation factors in mice with endotoxemia

To study the anticoagulant effect of Xiang-Qi-Tang (XQT),the mice model of endotoxemia was established to detect the expression of coagulation factors and their regulatory proteins in serum and aorta.The results showed that XQT could decrease the expression of TF and increase the expression of tPA in the aorta of mice with endotoxemia,and also decrease the expression of sEPCR in the serum.We further found that XQT caused the decrease of sEPCR through the regulation of PKC δ and ADAM17 to contribute the anticoagulation in mice.This study may provide a new strategy for treating endotoxin-induced disease and provide evidences for further researching the pharmacological action of XQT.
3.Effect of VEGF-C interfering on the lymphangiogenesis of a mouse model bearing human colon cancer

Objective To construct recombinant adenovirns containing small interfering RNA (siRNA) targeting human VEGF-C mRNA,then to study the inhibitory effects of adenovirus-mediated VEGF-C siRNA on growth and lymphangiogenesis of human colon cancer in BABL/c nude mice model.Methods In vitro:the specific siRNA sequence targeting human VEGF-C mRNA was selected.The homologous double-strand DNA was designed and synthesized.After such DNA was inserted into pDC316-EGFP-U6 by BamHI and HindⅢ,pDC316-VEGF-C siRNA-EGFP-U6 was obtained,then it was co-transfected into 293 cells with the hone plasmid pBHGF35.After generated by homologous recombination,Ad5F35-VEGF-C siRNA-EGFP-U6 was obtained,and it was packaged and amplified in 293 cells.The human colon cancer model was established in nude mice by hypo-injection LoVo cells.They were divided randomly into four groups (n=6):adenovirus,virus without target gene,single siRNA and PBS control group.Injecting intervention intra-tumorly in each groups,calculating the tumors volume and drawing the growth curve,calculating micro-vascular density (MVD) and micro-lymphatic density (LVD) by staining respectively of the lymphatic and vascular with anti-LYVE-1 monoclonal antibody and anti-CD<,34> monoclonal antibody were completed.Results Adenovirus group tumor sizes were smaller than other groups.The LVD were 8.47±2.1 and 17.35±4.7 (P<0.05) in adenovirns group and the PBS control group.The MVD were 22.65±6.04 and 23.19±7.63 (P>0.05) in adenovims group and the PBS control group respectively.Conclusion The adenovirus-mediated VEGF-C siRNA can significantly inhibit the growth and lymphangiogenesis of human colon cancer in nude mice model,hut have no effect on microangium vessels growth.
4.MR Diffusion-Weighted Imaging and ADC Value Measurement in the Diagnosis of Breast Axillary Lymph Node Cancer

Objective To value diffusion-weighted imaging and ADC value measurement of benign and malignant axillary lymph node identification value. Methods From 2006-06 to 2008--02, by collecting in the hospital for breast dynamic con-trast-enhanced scans and diffusion-weighted imaging, 217 lesions in 56 cases with a clear outcome of the surgery and pathology and/or needle biopsy results were retrospectively analyzed.Results The DWI could accurately show the lymph node of the armpit abnormality, 163 cases of 47 breast cancer cases with lymph node metastasis showed high signal in DWI image that did not decrease with increasing b value, similar to the 54 cases of 9 benign enlarged responsive prolifer-ative lymph nodes. But the former showed higher signal than the later. The ADC values were(1.017±0.114)×10-3mm2/s and (1.905±0.136)×10-3mm2/s respectively, and showed significant difference between two groups (P<0.05). Conclusion The dynamic contrast-enhanced subtraction MR technology benign and malignant breast lesions has high diagnosis and differ-ential diagnosis importance. DWI and ADC value measurement is a safe, non-invasive, accurate and rapid diagnosis of metastasis axillary lymph nodes and benign proliferative response of the lymph nodes means.
5.Diagnosis and treatment of the extrahepatic bile ducts carcinoma

Objective To summarize the methods of diagnosis and operation of carcinoma of the extrahepatic bile ducts. Methods The clinical data of 100 cases of carcinoma of the extrahepatic bile ducts which were treated in our hospital from 1972 to 1999 were retrospectively analysed. In this series, there were 68 cases of the cancer located in the upper portion of exlrahepatic duct(proximal cancer), 12 in midder portion(midder cancer), 18 in lower portion(distal cancer), and 2 in whole bile duct. Results The initial symptom was upper abdominal discomfort or vague pain, abdominal distension, weakness, weight loss and progressive jaundice. BUS, CT and MRI were scatheless. If the intrahepatic bile duct dilatation or extrahepatic cholestatic jaundice were revealed, PTC(13 cases in this series) or ERCP(42 cases in this series) were to further determine the location of tumor. According to the position and type of the tumor, the different operations were selected. Twenty-five cases(36.8%) of the proximal cancer were resected, including 15 cases of type Ⅰ treated with localresection or “skeletonization” resection, 9 cases of type Ⅱ treated with resection of the tumor and caudate lobe, 1 case of type Ⅲb treated with resection of the tumor, caudate lobe and left hepatic trisegmentectomy. Nine cases(75%) of midder cancer were resected. After resected the proximal and midder cancer, bile duct reconstruction by Roux-en-Y hepaticojejunostomy was performed on all the cases. Fourteen cases(77.8%) of distal cancer were treated by pancreatoduodenectomy. The total resection rate in this series was 48%. Of the cancer resected cases, 35 were followed up, the five-year survival rate was 58%. 32 of the 52 cases without cancer resection were followed up, and all of them died one to one and half year after operation. Conclusions BUS, CT and MRI are the first selective methods for early diagnosis of the carcinomas of extrahepatic bile duct. If needed, PTC or ERCP should be done because of these methods have more accurate diagnostic value. Surgical resection of the tumor is the only likelihood for effective treatment.
6.Diagnostic Value on Mammary Gland Focal Benign and Malignant with MRI Digital Subtraction

Objective To study the diagnostic value on mammary gland focal benign and malignant with MRI digital sub- traction. Methods By analyzing 109 histopathology cases of breast neoplasm which can be use three dimensional dynamic contrast and MRI digital subtraction. Based on early-phase enhancement rate peak time, time-signal intensity curve and enhanced lesions way, the diagnostic value on it was evaluated. Results The specificity signs of breast carcinoma are en- hanced ways on MRI enhancement scanning. The filling type is contrast agent filled from edge to centre. The sensitivity is 82.9%; specificity is 79.0%; accurate rate is 80.7%. Conclusion The aggrandizement form on filling type of breast MRI digital subtraction scanning focus has obvious advantages in breast carcinoma diagnosis.
7.Clinical Applications of MR Whole Spine Imaging with Total Imaging Matrix

Objective To study the values of MR whole spine imaging with TIM(Total Imaging Matrix) technique in diagnosing spine and spinal cord diseases.Methods Eighty-five patients who were suspected having spine diseases underwent whole spine MR.High resolution MR is performed when a local lesion was found on a Avanto 1.5T MR system(Siemens Company) by using different TIM in coils combination with automatic moving table and no-gap composing techniques.Results All 75 cases can be obtained clear images of total spine and spinal cord.In the all of 75 cases whole spine MR,the results show that normal in 15 cases,vertebral diseases in 41 cases,and spinal cord diseases in 19 cases.Conclusion The MR whole spine imaging with total imaging matrix has shorter scanning time and better image quality,position and nature of high accuracy,resolved the controversy of imaging covering and resolution.It can be improved the accuracy in diagnosing spine and spinal cord diseases.
8.Effects of tissue engineering bladder reconstruction in canines on upper urinary tract

Objective To analyze and evaluate the effects of the tissue engineering bladder reconstruction on the upper urinary tract structure and function.Methods The 8 male beagles were randomly divided into the two groups:sham-operation group (group A,n=4)and the collagen scaffold repair group (group B,n=4).The bladder defect animal model was established in the group B by using the collagen scaffold materials to repair the bladder.The renal function related biochemical indicators were detec-ted and the renal Doppler ultrasonic examination was performed in each group before repair and in 23 weeks after repair.The speci-mens from the two groups were performed the gross morphology observation and the histology examination on postoperative 24 weeks.Results The renal Doppler ultrasound examination showed the normal kidney morphology and normal blood flow signal.In the general observation,no calculi and neoplasm were found in the kidney and ureter of the experimental dogs.The renal function related biochemical indicators had no statistically significant differences between the two groups(P>0.05).The histological exami-nation indicated that the organization structure was integrity,the nephrons in each group had no obvious pathological changes.Con-clusion Using the collagen scaffold materials to reconstruct the canine bladder has no adverse influence on the upper urinary tract structure and function,this tissue engineering approach has good feasibility.
9.MRI Diagnosis of Bone Giant Cell Tumor

Objective To analse MRI appearances and diagnostic value of giant cell tumor of bone.Methods MRI features of giant cell tumor of bone proved by pathology in 14 cases were reviewed.Results The lesions located at distal femur in 7 cases,at approximal tibia in 3 cases,at distal humerus,approximal femur,thoracic vertebrae and sacrum in 1 case respectively.The characteristic appearances of MRI were cystic and solid masses,there was fluid-fluid level within tumors in company with bleeding.Tumors were slight low or low signal intensity on T1WI,slight high signal intensity on T2WI.Conclusion MRI is of significant value in diagnosis of bone giant cell tumor.
10.Effect of spindle checkpoint on Akt2-mediated paclitaxel-resistance in A2780 ovarian cancer cells.

Recent evidence has suggested that Akt2 plays an important role in the protection of cells from paclitaxel (PTX)-induced apoptosis and control of the cell cycle. In addition, some scholars suggested that the PTX sensitivity depends on a functional spindle assembly checkpoint. In the present study, we investigated the role of the Akt2/Bub1 cross-talking in apoptosis and cell cycle after exposure of the A2780 ovarian cancer cells to paclitaxel (PTX). Recombinant expression plasmid WT-Akt2 was transfected into A2780 cells by lipofectamine2000, and then the expression level of Akt2 gene was detected by using RT-PCR and Western blotting. Cell apoptosis and cell cycle were detected by flow cytometry and Hoechst 33342 staining after treatment with PTX. Moreover, we compared the expression level of Bub1 in different groups by Western blotting. Our study showed that up-regulation of Akt2 contributed to A2780 ovarian cancer cells overriding PTX-induced G(2)/M arrest, and inhibited Bub1 expression. Our findings might shed light on the molecular mechanism of PTX-induced resistance in ovarian cancer and help develop novel anti-neoplastic strategies.
