Progesteron could significantly inhibit the contracted effect of strips of tracheal and lung smooth muscles isolated from guinea pigs, which induced by Ach His and 5- HT. It also inhibited the contractility of phase I caused by intracellular calciumefflux in the course of 5 -HT stimulating tracheal smooth muscles and the Schultz - Dale responsiveness of sensilized of tracheal smooth muscles guinea pigs.

The application of genetic engineering technology in modern agriculture shows its outstanding role in dealing with food shortage. Traditional medicinal plant cultivation and collection have also faced with challenges, such as lack of resources, deterioration of environment, germplasm of recession and a series of problems. Genetic engineering can be used to improve the disease resistance, insect resistance, herbicides resistant ability of medicinal plant, also can improve the medicinal plant yield and increase the content of active substances in medicinal plants. Thus, the potent biotechnology can play an important role in protection and large area planting of medicinal plants. In the development of medicinal plant genetic engineering, the safety of transgenic medicinal plants should also be paid attention to. A set of scientific safety evaluation and judgment standard which is suitable for transgenic medicinal plants should be established based on the recognition of the particularity of medicinal plants.

8.An Electrochemical Biosensor for Detection of Breast Cancer Cells Based on Exonuclease I-catalyzed DNA Degradation Reaction

In terms of the characteristic of exonuclease I that can selectively catalyze the degradation of single-stranded DNA, we report a new sensitive electrochemical biosensor for the detection of breast cancer cells by using the aptamers of MUC1 that is overexpressed on the surface of MCF-7 cells. Because the binding of aptamer and MUC1 that is over-expressed on the surface of breast cancer cell MCF-7 can prevent its hybridization with the complementary probe DNA, the unhybridized single-stranded probe DNA that is immobilized on the electrode surface will be selectively digested by Exonuclease I and result in the removal of the signal molecule methylene blue. Therefore, by tracing the changes in electrochemical responses, the biosensor can be used to sensitively detect MCF-7 cells in a linear range from 103 to 106 cell/mL with a detection limit of 330 cell/mL, and easily distinguish the control islet β cells with high specificity.

Objective: To investigate the expression and identification of CH50 polypeptide in bladder cancer cell line BIU-87 by gene transfection. Methods:The eukaryotic expressing vector pCH510 of polypeptide CH50 was introduced into BIU-87 cells by gene transfection Lipofectimine TM2000.The expressed product was identified by immunohistochemistry and Western blot method. The expression of the transfected gene was identified by RT-PCR. Results:The BIU-87 cells could produce CH50 polypeptide by transfection. Conclusion:When the vector of pCH510 was transfected into BIU-87 cells in vitro, the cell adhension characteristic would be changed.
10.Effects of ganoderma spores on apelin expression in rats with myocardial ischemia injury induced by isoproterenol

AIM:To investigate the changes of apelin in blood and myocardium, and the protective mechanisms of ganoderma spores on myocardial ischemia injury induced by isoproterenol in rats. METHODS: The model of myocardial ischemia injury was induced by subcutaneous injection of high dose isoproterenol. Enzyme linked immunosorbent assay (ELISA) was used to measure the apelin contents in blood and myocardium. Semi-quantitative RT-PCR was used to measure the apelin mRNA level in myocardium. Electron microscope was used to observe the pathomorphological changes of myocardium. Ganoderma spores was administered i.g. for 7 weeks.
