The average recoveries were obtained as 98.4%, 98.2%, 98.5% and 98.2%(n=5). CONCLUSION: The method is simple, sensitive, accurate and suitable for the determination of four constituents in Ganmaokang Spray.
3.Molecular Typing of Neisseria Gonorrhoeae by Pulsed field Gel Electrophoresis and Random Amplified Polymorphic DNA Analysis
Chinese Journal of Dermatology 1995;0(03):-
Objective To evaluate the ability of restriction endonuclease analysis by pulsed field gel electrophoresis (PFGE REA) and random amplified polymorphic DNA (RAPD) analysis for molecular typing of Neisseria gonorrhoeae isolates. Methods Genomic DNA from 36 Neisseria gonorrhoeae isolates were digested with SpeI and analysed by contour clamped homogeneous electric fields electrophoresis. The 36 isolates were also characterised by RAPD analysis with arbitrary primer OPA 03. Results 12~19 DNA fragments from 5 to 600kb were obtained among the 36 isolates after PFGE REA. The 36 isolates were discriminated into 20 PFGE patterns. Amplification of Neisseria gonorrhoeae DNA with primer OPA 03 produced 15 different DNA fragments (280~1900bp), and 3~9 fragments per strain could be seen. The 36 isolates showed 18 different RAPD patterns. Strains sharing common auxotypes and antibiotic spectrum could be differentiated by PFGE REA and RAPD analysis. General agreement was found between these two techniques. Isolates from different geographic areas and even some isolates in the same area showed considerable amount of DNA polymorphism. In addition, some isolates shared common or very similar patterns were unique to a given geographic area. Conclusion It is concluded that both PFGE REA and RAPD analysis are useful, sensitive molecular techniques for differentiation of clinical isolates of Neisseria gonorrhoeae. They should be helpful in the investigation of strain origin, clonal relation among strains and spread of antibiotic resistance. Compared with PFGE REA, RAPD analysis is faster, relatively simple and more economical.
4.Short-segment versus long-segment pedicle screw fixation for repair of thoracolumbar fracture:one-year follow-up of vertebral compression rate
Chinese Journal of Tissue Engineering Research 2015;(13):2040-2044
BACKGROUND:The effects of posterior decompression fixation in the repair of thoracolumbar fracture are affirmative. The fixation was divided into short-segment and long-segment pedicle screw fixation according to different treatment methods. The therapeutic effects of above two methods are controversial in the clinic at present. OBJECTIVE:To compare and observe the anterior vertebral compression ratio and the changes in Cobb angle after posterior short-segment and long-segment pedicle screw placement fixation in the repair of thoracolumbar fracture. METHODS:120 patients with thoracolumbar fracture were enrol ed in this study, and assigned to two groups according to fixation methods. Study group received posterior short-segment pedicle screw fixation. Control group received posterior long-segment pedicle screw fixation. Anterior vertebral compression rate and Cobb angle were observed before fixation, immediately and 12 months after fixation in both groups. RESULTS AND CONCLUSION:There was no significant difference in anterior vertebral compression rate and Cobb angle between the two groups before fixation (P>0.05). Anterior vertebral compression rate and Cobb angle were significantly smal er in the study group than in the control group immediately and 12 months after fixation (P<0.05). Besides, surgical time and blood loss volume were less in the study group than in the control group (P<0.05). These results indicated that posterior short-segment pedicle screw fixation for treatment of thoracolumbar fracture can effectively improve kyphosis, recover vertebral compression degree, and its surgical time is short and blood loss volume during surgery is little, so it is better than posterior long-segment pedicle screw fixation.

Chinese Journal of Experimental Ophthalmology 2016;34(3):227-233
Background miRNAs are a group of non-coding small RNA molecules,and they play an important role in regulating the apoptosis of LECs.The biological effects and mechanisams of miRNAs on LECs in agerelated cataract still need to be further elucidated.Objective This study was to investigate the anti-oxidative effects of miR-204 on age-related cataract in vitro.Methods The specimens of anterior lens capsules from agerelated cataract patients and normal donors were collected in Shandong Eye Institution and 20 subjects for each.The expression level of miR-204 was detected and compared between cataractous eyes and normol eyes by real-time fluorescence quantitative PCR (RT-qPCR).HLE-B3 cells,a human LEC line,were cultured,and the oxydative stress models of LECs were established by adding 200 μmol/L H2 O2 in the medium.The models were divided into model control group,miR-204 agomir group,agomir negative control group,miR-204 antgomir group and antagomir negative control group according to the difference of tranfected agents,and normal cells served as the control group.Twenty-four hours after transfection,the expression levels of miR-204 mRNA in the cells of various groups were detected by RT-qPCR to varify the transfection rate.Apoptosis rate of the cells was assayed by flow cytometry.The relative expression levels of TP53INP1 mRNA and p53 mRNA as well as their proteins were detected by RT-qPCR and Western blot,respectively.Results The mean expression level of miR-204 was significantly lower in the anterior lens capsules of cataractous eyes than that in the normal eyes (t=14.21,P<0.05).The mean apoptosis rate of cells was 1.31±0.12,4.90±0.28,2.60±0.15,4.39±0.20,5.74±0.13 and 4.34±0.63 in the normal control group,model control group,miR-204 agomir group,agomir negative control group,miR-204 antgomir group and antagomir negative control group,respectively.The apoptosis rate was significantly increased in the model control group compared with the normal control group (t =-20.69,P<0.01) and the apoptosis rate was also increased in the miR-204 antagomir group compared with antagomir negative control group (t =3.79,P<0.05);while the apoptosis rate in the miR-204 agomir group was significantly declined in comparison with agomir negative control group (t=-12.20,P<0.01).The relative expression levels of TP53INP1 and p53 mRNA and proteins in the cells were significantly higher in the model control group than those in normal control group (mRNA:t =6.44,11.71,both at P<0.0I;protein:t =10.72,19.40,both at P<0.01),and so were between the miR-204 antagomir group and antagomir negative control group (mRNA:t =4.07,3.74,both at P< 0.05;protein:t =7.18,10.58,both at P<0.05).However,the expression levels of TP53INP1 and p53 mRNA and protein were significantly reduced in the miR-204 agomir group in comparison with the agomir negative control group (mRNA:t =-19.28,-10.58,both at P<0.05;protein:t=-6.50,-6.36,both at P<0.05).Conclusions miR-204 induces oxidative damage of age-related cataract via targeting TP53INP1,which suggests that the activation of TP53INP1-p53 signaling may be involved in the development of age-related cataract.

Acta Anatomica Sinica 1953;0(01):-
Objective\ The present study was carried out to examine the relationship between the distance of axotomy and axonal regeneration systematically,and the effect of a pre\|degenerated peripheral nerve(PN) graft on axonal regeneration of retinal ganglion cells(RGCs) axotomized at different distances. Methods\ The optic nerve(ON) was transected at 0\^5,1,1\^5,2,3 or 7mm from the optic disc and a normal (the normal group) or pre\|degenerated(the pre\|degeneration group) PN graft was transplanted onto the ocular ON stump in adult hamsters. Results\ In both groups,the number of regenerating RGCs 28 days after grafting decreased significantly when the distance of axotomy increased from 0\^5 to 7mm with a sharp decline between 0\^5 and 3mm.When the corresponding distance points between the two groups were compared,enhanced regeneration was observed at 2 and 3mm in the pre\|degeneration group. Conclusion\ These results show that the distance of axotomy on the ON of adult hamsters is critical in determining the number of regenerating RGCs.The beneficial effect of a pre\|degenerated PN graft on axonal regeneration of axotomized RGCs is conditional on the distance of axotomy.\;
