1.Effect of lymphovascular invasion on prognosis in high-grade pT1 bladder cancer after first transurethral resection of bladder tumor

Objective To explore the effect of lymphovascular invasion (LVI) on prognosis in the high-grade stage pT1 bladder cancer after first transurethral resection of bladder tumor (TURBT). Methods Thirty-five high-grade stage pT1 bladder cancer patients who underwent first TURBT and showed LVI positive according to the postoperative pathological (observation group) and 70 high-grade stage pT1 bladder cancer patients who underwent first TURBT and showed LVI negative according to the postoperative pathological (control group) were enrolled into the study. The tumor recurrence rate, tumor progression rate, recurrence-free survival (RFS) and overall survival (OS) were compared between 2 groups. Results The follow-up time in observation group was 15 - 71 (42.1 ± 18.1) months, and in control group was 20-72 (50.9 ± 12.2) months. The tumor recurrence rate and tumor progression rate in observation group were significantly higher than those in control group:88.6%(31/35) vs. 34.3%(24/70) and 54.3%(19/35) vs. 22.9%(16/70), and there were statistical differences (P<0.01). The RFS and OS in observation group were significantly shorter than those in control group: (19.7 ± 4.5) months vs. (46.8 ± 9.2) months and (43.4 ± 12.6) months vs. (63.4 ± 7.1) months, and there were statistical differences (P<0.05). Conclusions There is a significant correlation between LVI and tumor recurrence and progression in the high-grade pT1 bladder cancer after first TURBT, and the LVI can predict disease progression and poor prognosis.
2.MRI in diagnosis of fetal meconium peritonitis

Objective To explore the value of MRI in diagnosis of fetal meconium peritonitis.Methods Seven meconium peritonitis fetuses proved by surgery and pathology were enrolled.The prenatal MRI findings and clinical data were analyzed retrospectively.Results Six fetuses showed a large amount of ascites,intestinal tube floating in the abdomen,small intestine gathered together.One fetus showed a giant abdominal cystic mass,with bowel compressed,displaced and uneven dilated.Four fetuses showed small colon and rectum,or without meconium signal.Two fetuses were accompanied by bilateral hydrocele.Amniotic fluid increased in 3 cases.After the neonates were born,1 case of them died from sudden heart rate decline during operation,1 case died from severe pulmonary edema after operation,and 5 cases survived after operation.Conclusion MRI has some features in the prenatal diagnosis of meconium peritonitis,which can provide an important basis for postpartum treatment and evaluation of prognosis.

4.In situ Determination of Anthracene Adsorbed onto Mangrove Root Surface Micro-zone Using Microscopic Fluorescence Spectrometric Analysis System

A microscopic fluorescence spectrometric analysis ( MFSA) system was set in the laboratory. A novel method for in situ determination of Anthracene ( Ant) adsorbed onto root surface micro-zone of two kinks of mangrove plant, named Kandelia obovata ( K. obovata) and Avicennia marina ( A. marina) by MFSA was established. Fluorescence spectra of Ant adsorbed on root surface micro-zone were obtained by synchronous scanning mode. The signal to noise (S/N) of Ant (5. 3 pg/μm2) adsorbed on K. obovata and A. marina root surface micro-zone increased up to 5. 5 and 6. 8 while wavelength offset (△λ) both were at 60 nm, respectively. The linear dynamic ranges of established method were 5. 3-63. 2 pg/μm2 for K. obovata and 10. 5-52. 6 pg/μm2 for A. marina, with the detection limits of 1. 1 pg/μm2 and 5. 5 pg/μm2 , respectively. The relative standard deviations were both less than 12 . 5% ( n=9 ) , and the recoveries were 98 . 1% -117. 0% and 81. 2%-110. 9%, respectively. The result showed that the MFSA system had ability to obtain quantitative information of fluorescence spectra and fluorescence image of Ant adsorbed onto plant roots surface micro-zone.
5.Influences of different cryoprotectants by ladder-style freezing from low temperature refrigerator to liquid nitrogen on cryopreservation of hemopoietic stem cells

BACKGROUND: Uncontrolled-rate freezing in -80 ℃ refrigerator is convenient, while controlled-rate freezing in -196 ℃ liquid nitrogen is reliable and long-term, the combination of the two can simplify the process and has been successfully used in clinics. OBJECTIVE: To explore the influences of different cryoprotectants by ladder-style freezing from -80 ℃ low temperature refrigerator to liquid nitrogen on the cryopreservation of hemopoietic stem cells. METHODS: The experiments were divided into four groups: 10% dimethyl sulfoxide (DMSO) group, 5% DMSO combined with 3% hydroxyethyl starch group, 5% DMSO combined with 0.25 mol/L trehalose group, 5% DMSO combined with 3% hydroxyethyl starch and 0.25 mol/L trehalose group. Peripheral hemopoietic stem cells were cryopreserved by ladder-style freezing from -80 ℃ low temperature refrigerator to liquid nitrogen. The ultrastructural changes were examined by transmission electron microscopy, the expressions of Annexin-V, PI and Caspase-3 were detected by flow cytometry. RESULTS AND CONCLUSION: There was no significant difference in survival rate, apoptotic rate and necrotic rates of the cryopreserved cells in the four groups (P > 0.05). The ultrastructural changes had no significant difference under the transmission electron microscopy. The viability was more than 90% in frozen-thawed mononuclear cell colonies, and the apoptosis was roughly 50% in the frozen-thawed CD45+ cell population, which contained many mature cells. Of hemopoietic stem cells, early stage cells have greater resistance to damage of cryopreservation than late stage cells. It is concluded that the addition of hydroxyethyl starch or trehalose into DMSO exhibits no synergistic protective effect on the cryopreservation of hemopoietic stem cells.

Objective To investigate the changes of IL‐1β,IL‐2 ,IL‐4 ,IL‐6 ,IL‐10 and INF‐γexpressions in bone marrow of chro‐nic myeloid leukemia(CML)patients .Methods The IL‐1β,IL‐2 ,IL‐4 ,IL‐6 ,IL‐10 and INF‐γexpression levels were detected by u‐sing flow cytometry in 30 cases of CML chronic phase(CML‐CP) ,21 cases of CML accelerated phase(CML‐AP) ,15 cases of CML blastic phase(CML‐BP) ,42 cases of CML remission after treatment and 7 cases of non‐remission .Then the detection results were compared with those in the control group .Results The expression levels of INF‐γ and IL‐2 in each CML groups were lower than those in the control group(P<0 .05) ,while the expression levels of IL‐1β,IL‐4 ,IL‐6 and IL‐10 were higher than those in the con‐trol group(P<0 .05) .With the disease condition progression ,the INF‐γand IL‐2 levels were gradually decreased ,i .e .,CML‐BP
7.Experience of laparoscopic treatment of ureteral calculi

Objective To summary the therapy outcome of retroperitoneal laparoscopic treatment on upper ureteral calculi.Methods The retroperitoneal laparoscopic treatment were performed to all cases from Jun.2011 to Jun.2012 in our hospital.All cases were treated with 3-hold method (two 10 mm and one 5 mm ports).The retroperitoneal space was made by a combination of blunt and balloon dissection,and the space was maintained with CO2 Ureteral longitudinal incision was made to remove the stones,and double J catheter was served as stent drainage.Absorbable suture was used to suture ureteral incision.Results A retroperitoneal approach was performed in 12 patients,and another patient was conducted the open surgery because the stone cower in the kidney calices.Operative periods ranged from 55 to 132 min (average was 85 min).There were no significant postoperative complications.Conclusion It is a minimally invasive and effective approach in the therapy of upper urerteral calculi with laparoscopic.
8.Impact on the Expression of CD123 on K562 Cells Induced by rhIFN-γ

Objective To investigate the inhibitory effect of rhIFN-γ on human chronic myeloid leukemia cell line K562,and the impaction on the expression of CD123.Methods MTT method was used to test cell relative viability with rhIFN-γ at different concentrations.The expression of CD123 on K562 ceils was detected by flow cytometry.Results The relative inhibition of K562 cells proliferation was hampered when the cells were treated with rhIFN-γ for 72 h at the concentration of 5 x 10P ~ 108 U/L,respectively.However,rhIFN-γ at 2 x 105 U/L was benefit to K562 cells proliferation.After treatment with rhIFN-γ at 0,2 x 105 U/L and 107 U/L,the percentages of CD123 expression on K562 cells were (4.10 ± 1.46) %,(7.2 ± 2.50) % as well as (21.6 ± 1.17) %,respectively.Compared with the control group,107 U/L rhIFN-γ significantly increased the expression of CD123 on K562 cells (P<0.05).Conclusion The effect of rhIFN-γ on the growth of K562 cells has two aspects (inhibition or proliferation),and it can increase the expression of CD 123.
9.Stimulating test of bear bile eye drops on rabbit eyes

Objective To explore the stimulating effect of bear bile eye drops on rabbit eyes frequently in order to provide experimental basis for drug use. Methods Bear bile eye drops dripped into the right eye of rabbit conjunctival sac as experimental group ( different frequency as 1, 5, 10 min) while normal saline was used in the left eye of rabbit as the control group ( one drop once and 10 times continually) . We observed the stimulation of rabbit eyes at 0, 6, 12, 24, 48 hours after the last dripping. Results At the observation of 5 and 10 min interval set, the right eyes of rabbit had no hyperemia, no edema, no secretion, no maculae corneae, and normal iris that was 1 score according to eye stimulation response scale belonging no irritation. After 1 min interval set observation of right eyes, they presented light conjunctiva hyperemia, light edema, increased secretion of conjunctiva sac with 5 score belonging to light stimulation;but it decreased to 3 scores at 6 h interval set belonging to no stimulation;the average score declined underneath 1 score at the observation of 12 h interval set. Conclusions Bear bile eye drops can arouse reversible light stimulation when used in high frequency, so the safety is high, but we should avoid the high frequency of use.
10.The diagnostic value of prenatal MRI on jejunal and ileal atresia in fetus

Objective:To evaluate the value of MRI in the diagnosis of jejunal and ileal atresia in fetus.Methods:This study included thirteen neonates with surgically and pathologically confirmed jejunal or ileal atresia without other gastrointestinal diseases treated in our institution between January 1, 2010 and December 31, 2018. MRI was performed on all fetuses subsequent to routine prenatal sonographic examinations indicated bowel dilation or ascites. MR images were analyzed by two radiologists,and MRI diagnosis was compared with surgical and pathological results.Results:On prenatal MRI, eight fetuses exhibited single or multiple dilated small bowel loops, one of them exhibited a cystic mass in the abdomen; five fetuses exhibited massive ascites, and collected bowels. On final surgical and pathological diagnosis, nine fetuses had ileal atresia, five of them had intestinal perforation; four fetuses had jejunal atresia, one of them had intestinal perforation and a meconium pseudocyst. Among the 7 cases without intestinal perforation, the atresia location was diagnosed correctly in 5 cases. The 6 fetuses with intestinal perforation were diagnosed as intestinal atresia and meconium peritonitis by prenatal MRI, but the location of atresia could not be determined.Conclusions:Jejunal and ileal atresia have their characteristic manifestations on prenatal MRI. The location of atresia can be inferred from the distribution of amniotic fluid and meconium in the intestine. Prenatal MRI has a complementary role in the assessment of fetal bowel abnormalities after standard obstetric ultrasound.
