1.DETERMINATION OF CONJUGATED BILE ACIDS IN BILE BY HIGH PERFORMANCE LIQUID CHROMATOGRAPHY

A reversed-phase high performance liquid chromatographic method for the determination of conjugated bile acids in human bile had been reported. The procedure of sample preparation is simple since a guard column is used in this system. Recovery of the added bile salts was in the range of 87.01~110.75%. The limit of detection ranged from 0.09 ?g to 0.21 ?g. The method is used for the analysis of conjugated bile acids of bile from the patients with gallstone and control groups.
2.Facial nerve and the surrounding vessels: an anatomic study

Objective:To study the relationship between facial nerve and surrounding vessels. Methods:Dissect skull of cadaver 40 sides of 20 cases: open the calvaria and clean the cerebrum, then open the tentorium of cerebellum, object and measure distance between the facial nerve and the nearest vessel, follow the course of it. Results:60%(24/40) of the arteries have the comprission with the facial nerve; among the nearest vessels, 87.5%(35/40) of them are anterior inferior cerebellar artery, and 12.5% (5/40) are posterior inferior cerebellar artery; the distances between facial nerve and the nearest vessels are from zero to 5.78mm, with an average of 1.14 mm; 94.3%(33/35) of the anterior inferior cerebellar artery emitted from the lower 2/3 of the basic artery. Conclusion:The most familiar vessel next to the facill nerve is the anterior inferior cerebellar artery, and the second is the posterior inferior cerebellar artery; most of the anterior inferior cerebellar artery emitted from the basic artery, exactly, the lower 2/3 of the artery.
3.Nurses′participation in the management of anticoagulation among patients with atrial fibrillation:a literature review

Atrial fibrillation (AF) is the most common sustained cardiac arrhythmia worldwide with a trend of increasing morbidity and prevalence. It usually links to a high disability and mortality rate. Stroke is the most important and severe complication of AF, as a core strategy to prevent it, anticoagulation therapy is recommended to reduce the risk of stroke events. However, management risks including poor treatment adherence, individual difference and bleeding risk can′t be ignored. In the western countries, they have successively established a nurse-led integrated chronic care program for patients with atrial fibrillation. We can use it to guide our future work. Anticoagulation therapy is a long-term managing process. To integrate the existing medical resources, nurses must give their full play to access the deep needs and guide them to learn the core skills of management, thus improving the adherence to anticoagulation therapy.
4.Methods of suppressing physiological myocardial uptake in 18F-FDG imaging

18F-FDG PET has been widely used in myocardial oncology and inflammatory imaging.Suppression of physiological uptake in heart is required for accurate diagnosis of some thoracic diseases,cardiac inflammatory diseases and coronary atherosclerosis.There is no unified criteria to reduce myocardial 18F-FDG uptake.This review summarizes the research progress of inhibiting physiological myocardial 18F-FDG uptake by controlling diet,mediating blood glucose and applying drugs.
5.Fecal calprotectin in differential diagnosis of ulcerative colitis

Objective To investigate the clinical value of fecal calprotectin as a marker for diagnosis of ulcerative colitis(UC).Methods ELISA assay was used to measure the concentrations of calprotectin in feces of 30 UC patients and 20 healthy controls.The disease activity of UC was determined by Truelove Witts histological criteria.Results The level of calprotectin in feces was significantly higher in feces with active UC(578?g/g) than with inactive UC(8.4?g/g,P0.05),but significant difference existed between each active grade and inactive phase of UC.The results revealed siginificant correlation between the endoscopic grades of disease activity and fecal calprotectin concentration(r=0.85,P
6.Preliminary application of rep-PCR in epidemiological research of extended spetrum ?-lactamase(ESBL)+ Escherichia coli

Objective To establish better reaction factors for rep-PCR and to investigate if the extended spectrum ?-lactamase(ESBL)+ Escherichia coli isolated from different patients in the respiratory ward have the same origin by using rep-PCR.Methods The ESBL confirmation was taken by double disc confirmatory test.The susceptibility testing was performed with K-B test.By applying the widespread enterobacter repetitive intergenic consensus as a primer,the stains were typed by rep-PCR following electrophoresis in agarose gel.Results The analysis of the PCR productions indicated that it would create useful DNA ban-prints and all these 22 ESBL(+)Escherichia coli strains were of three origins.Conclusion The method of rep-PCR is practical for epidemiological research in nosocomial infection.
7.Trigeminal nerve and superior cerebellar artery:An anatomic study

Objective To study the relation between the trigeminal nerve and the superior cerebellar artery. Methods The authors dissected 20 cadaver skulls (40 sides):we firstly opened the calvaria and cleaned the cerebrum, then opened the tentorium of cerebellum and observed the relation between the trigeminal nerve and the superior cerebellar artery, as well as measured the distance between them, finally inspected the trend of the superior cerebellar artery. Results In the study the nearest blood vessel to the trigeminal nerve was the superior cerebellar artery, with a distance of 0~8 1 mm (mean, 2 19 mm). All the superior cerebellar arteries had rostral and caudal branches. Approximate 47 5% of the superior cerebellar arteries (19/40) crossed with trigeminal nerves, and in 87 5% of the superior cerebellar arteries (35/40) the closest site to the trigeminal nerve was the junction of the main trunk and the rostral or caudal branches. Conclusions The superior cerebella artery usually gives off rostral and caudal trunks. The junction of the main trunk and the rostral or caudal branches is nearest to the trigeminal nerve.
8.Effects of FGF-2 on the intercellular communication in the cardiac myocytes

Objective To investigate the effects of FGF 2 on the intercellular communication in the cardiac myocytes. Methods The primary neonatal ventricular myocytes of rats were cultured with FGF 2 (0,1,10,100 and 1000ng/ml) for 1 or 4 days. Then the mean fluorescence recovery rate (MFRR, %/min) of these myocytes labeled with carboxyfluocein diacetate after photo bleaching was examined with laser scanning cytopmetry (ACAS Ultima). Results The MFRR levels of the myocytes cultured with FGF 2 (10 and 100ng/ml) groups for 1 day were significantly decreased compared with the control group, while no statistic changes were found among the other groups (FGF 2, 1 and 1000ng/ml) and the control. After FGF 2 exertion on the myocytes for 4 days, the increased cell number, enlarged cellular body and decreased MFRR level were found in the myocytes cultured with FGF 2 (10 and 100ng/ml) groups. And the decreased cell number, disarranged and shrunk cells and significantly increased MFRR level were gotten in the group (FGF 2 1000ng/ml) compared with the control group. Conclusion These results suggested the FGF 2 may have both short and long term effects on the cell cell communication in the cardiac myocytes mediated by gap junction, and be necessary to the cardiac myocyte coordination during the cell growth, differentiation and perhaps apoptosis.
9.Construction of recombinant lentiviral vector expressing HCV core-Ant

Objective To construct a recombinant lentiviral vector for HCV core-Ant and then study its effect on the transdifferentiation of hepatic stem cells. Methods The HCV core-Ant was obtained by PCR of two primers template with each other and T-vector cloning method,and then subcloned to pLenti6/V5-D-TOPO. The restriction endonuclease and T4 DNA ligase were used to construct the vector. pLenti6/V5-D-HCV core-Ant and the ViraPowerTM PackagingMix (containing three packaging plasmids pLP1,pLP2 and pLP/VSVG) were cotransfected into 293ET cells to produce replication in competent lentivirus after transfection. The viral supernatant on 293T cells was collected. The expression of the lentiviral vector containing HCV core-Ant in Hela cells was measured by immunohistochemistry. Then we constructed the lentiviral vector containing green flurosecent protein by the similar method,and the titers were determined. Results HCV core-Ant was identified and analyzed by restriction enzyme digestion and sequencing,respectively. The expression of the recombinant lentiviral vector plasmid containing HCV core-Ant in Hela cells was confirmed by immunohistochemistry. The 3T3 cells transfected the lentiviral vector containing green flurosecent protein were found to show strong expression of GFP,which confirmed that the four-plasmid system of the lentiviral vector was successfully constructed. Conclusion The recombinant lentiviral vector expressing HCV core-Ant was successfully constructed by molecular cloning and recombination techniques in vitro,which will be beneficial to guiding further study on gene therapy of cancer.
